The choline head group containing phosphatidylcholine (PC) and sphingomyelin (SPM) are major eukaryotic lipid components playing an important role in forming membrane microdomains and serve as precursor of signaling molecules. Both lipids can be monitored by positive ion mode electrospray tandem mass spectrometry using a parent ion scan of m/z 184. Although PC species appear at even m/z and SPM species at odd m/z, there may be a significant overlap of their isotopes. In order to separate PC and SPM species, an isotope correction algorithm was established, which utilizes calculated isotope percentages to correct the measured peak intensities for their isotopic overlap. We could demonstrate that this approach was applicable to correct the isotope overlap resulting from spiked PC and SPM species. Quantification was achieved by addition of different PC and SPM species prior to lipid extraction. The developed assay showed a precision, detection limit and robustness sufficient for routine analysis. Furthermore, an analysis time of only 1.3 min combined with automated data analysis using self-programmed Excel Macros allows high-throughput analysis. In summary, this assay may be a valuable tool for detailed lipid analysis of PC and SPM species in a variety of sample materials. D

Background: An increasing number of publications point to the importance of voriconazole plasma monitoring. Our aim was to develop a fast and simple LC-MS/MS method for the quantification of plasma voriconazole. Methods: 20 μL of plasma was extracted by adding a precipitation reagent containing the internal standard (d 3 -voriconazole). Analysis was performed on a Quattro Micro tandem mass spectrometer equipped with an Alliance HPLC 2795 separations module. MRM transitions were m/z 350.0→281.4 for voriconazole and m/z 353.0→284.4 for d 3 -voriconazole. Quantification was done by both linear calibration curves and multiplication of the response ratio by a predefined factor. Results: A method using protein precipitation as only pretreatment with an analysis time of 2 min was developed. Within-and between-run precision, expressed as coefficient of variation, at 6 concentrations ranged from 2.8% to 11.7%. Accuracy, expressed as a percentage of the theoretically added concentration, ranged from 89.0% to 109.0%. Linearity was demonstrated from 0.06 mg/L (0.17 μM) to 20.0 mg/L (57.3 μM). Conclusions: The described method offers a simple and rapid analysis of voriconazole in human plasma for clinical routine. Quantification with a predefined factor permits omitting calibration to each run and thereby reduces workload, costs and turn-around time.

Tepoxalin is a veterinary drug registered for use in the dog as a dual inhibitor (cyclooxygenase -5 lipoxygenase). In the horse, it predominantly triggers a strong cyclooxygenase inhibition; this bias seems to be due to the action of its metabolite(s). Among these, only the RWJ-20142 is well known, while to the best of our knowledge no information is available on the other metabolites produced in vivo. Hence, the identification of its main metabolic pathway is pivotal to better understand its clinical activity.

Coli surface antigen 6 (CS6) is one of the most prevalent colonization factors among enterotoxigenic Escherichia coli (ETEC) isolated in developing countries. Although it is known that CS6 is encoded by a plasmid, there are no reports on the sequence analysis of the CS6-encoding plasmid or genes exhibiting similar behavior to CS6. Here, we report the isolation of the CS6-encoding plasmid, pCss165Kan, from 4266 DcssB::kanamycin (Km) and its complete nucleotide sequence. This plasmid consisted of 165,311 bp and 222 predicted coding sequences. Remarkably, there were many insertion sequence (IS) elements, which comprised 24.4% of the entire sequence. Virulence-associated genes such as heat-stable enterotoxin, homologues of ATP-binding cassette transporter in enteroaggregative E. coli (EAEC), and ETEC autotransporter A were also present, although the ETEC autotransporter A gene was disrupted by the integration of IS629. We found that 2 transcriptional regulators belonging to the AraC family were not involved in CS6 expression. Interestingly, pCss165 had conjugative transfer genes, as well as 3 toxin-antitoxin systems that potentially exclude other plasmid-free host bacteria. These genes might be involved in the prevalence of CS6 among ETEC isolates.

This case describes the experiences of a healthcare organization in the North of England that introduced a multi-lingual electronic patient information system, the first of its kind in this part of the country. This information technology (IT) based project, commissioned as a pilot for the establishment of an information system to cater to the needs of ethnic minority languages, failed to make progress in-spite of its great potential, and the user satisfaction that it offered. The case presents a number of issues related to how the design and deployment of the project resulted in its lack of satisfactory progress. The findings from this case suggest why the project failed: the failure being a direct result of the approach taken to the project.

This paper concerned with the comparative study of design and implementation of a 32bit FPGA Implementation of RISC Processor Using VHDL Reduced Instruction Set Computer (RISC) processor on a Field Programmable Gate Arrays (FPGAs). The processor has been designed with VHDL, synthesized using Xilinx ISE 9.1i Webpack, simulated using ModelSim simulator, and then implemented on Xilinx Spartan 2E FPGA that has 143 available Input/output pins and 50MHz clock oscillator and the Field Programmable Gate Array (FPGA) based 64-bit RISC processor with built-inself test (BIST) feature implemented using VHDL and was, in turn, verified on Xilinx ISE simulator. The VHDL code supports FPGA, System-On-Chip (SOC), and Spartan 3E kit. This paper also presents the study of architecture, data path and instruction set (IS) of the RISC processor.

When the mechanisms of fading channels were first modeled in the 1950s and 1960s, the ideas were primarily applied to over-the-horizon communications covering a wide range of frequency bands. The 3-30 MHz high-frequency (HF) band is used for ionospheric communications, and the 300 MHz-3 GHz ultra-high-frequency (UHF) and 3-30 GHz super-high-frequency (SHF) bands are used for tropospheric scatter. Although the fading effects in a mobile radio system are somewhat different than those in ionospheric and tropospheric channels, the early models are still quite useful to help characterize fading effects in mobile digital communication systems. This tutorial addresses Rayleigh fading primarily in the UHF band. That affects mobile systems such as cellular and personal communication systems (PCS). The purpose of this work was to explore the relation between protein-protein interactions (PPIs) and solution viscosity at high protein concentration using three monoclonal antibodies (mAbs), two of the IgG 4 subclass and one of the IgG 1 subclass. A range of methods was used to quantify the PPI either at low concentration (interaction parameter (k D ) obtained from dynamic light scattering, DLS) or at high concentration (solution storage modulus (G 0 ) from ultrasonic shear rheology). We also developed a novel method for the determination of PPI using the apparent radius of the protein at either low or high protein concentration determined using DLS. The PPI measurements were correlated with solution viscosity (measured by DLS using polystyrene nanospheres and ultrasonic shear rheology) as a function of pH (4-9) and ionic strength (10, 50 and 150 mM). Our measurements showed that the highest solution viscosity was observed under conditions with the most negative k D , the highest apparent radius and the lowest net charge. An increase in ionic strength resulted in a change in the nature of the PPI at low pH from repulsive to attractive. In the neutral to alkaline pH region the mAbs behaved differently with respect to increase in ionic strength. Two mAbs (A and B) showed little or no effect of increasing ionic strength, whereas mAb-C showed a remarkable decrease in attractive PPI and viscosity.

The number, variety and complexity of projects involving data mining or knowledge discovery in databases activities have increased just lately at such a pace that aspects related to their development process need to be standardized for results to be integrated, reused and interchanged in the future. Data mining projects are quickly becoming engineering projects, and current standard processes, like CRISP-DM, need to be revisited to incorporate this engineering viewpoint. This is the central motivation of this paper that makes the point that experience gained about the software development process over almost 40 years could be reused and integrated to improve data mining processes. Consequently, this paper proposes to reuse ideas and concepts underlying the IEEE Std 1074 and ISO 12207 software engineering model processes to redefine and add to the CRISP-DM process and make it a data mining engineering standard.

Primarily used for administration use, more precisely for the execution of financial data, the computational processing became involved for the manipulation of the health records (EHR) for the population only from the 1970’s. The main reason for this was because the organizations did not already have in place the necessary technology nor the answers concerning the use or storage of such information. The process of automated work has therefore intrigued the medical professionals who were considering the benefits that such new machines could bring in the area of health observation (Sethi and Theodos, 2009). Nevertheless Walsham (2012) argues that the past few decades saw the tremendous influence and development of such technology namely through the increase in personal computer use, graphical workstations, the development of the new methods for human–computer interactions, the innovations and changes brought to the storage of mass data, the Internet or to the wireless communications. The intriguing role of such technology for the health society is the main attention for this proposal.
Based mainly on previous theoretical studies the theses seeks to show that a better collaboration between the involved disciplines could not just simplify but also bring considerable improvements to such an important domain that paradoxically seems to be the one advancing the less.
After a theory inquiry that helps classify the information, a more particular analyses is given to the existing technology and the health informatics research. The means of sharing the electronic data and the recent developments in genetics, informatics and medical science are an important step for the understanding the research problem. In order to do this the analyses follows both qualitative and quantitative approaches and it is primarily using explanatory and descriptive methods with outcomes from observational studies or past research cases. Moreover, a closer attention is given to the analyses, recognition and classification of the existing software applications by comparatively looking at some of the most used browser and desktop applications. A final chapter regards the discussion of the results, the impact of such study and it makes references for future research.

Measurement of the information system (IS) project success has become the interesting topic for researchers and practitioners since the Standish Group published their findings in 1994. Project success theory is the main concept in this topic, but this theory is still an ambiguous concept and lack in agreement among researchers and practitioners. They are also still tending to focus on single or partial dimension. Therefore, they did not get a clear picture of the system measurements. This study developed an alternative model of the project success measurement based on input-process-output (IPO) model. The development was conducted using comparison, adoption, adaptation, and combination the previous theories and models: Davis's IPO model, the project success theories, Delone and McLean' model, and the project classificatory framework. As indicated in most studies that most of models are developed using the previous theories and models rather than on empirical proofs. The result is a IS project success model consisting of 9 variables and 36 relationships among the variables. Although, the model is only a conceptual model, but it was developed completely and coherently considering three main aspects of project success measurement, namely: processional and causal models, project success theories, and the influence concept of project environment.

In this paper an introduction to the principles and methods used in logistics and supply chain management is presented. It begins by a discussion on fundamentals and explains the relevant terms. Next policy and practice associated with logistics and freight services are regarded with a focus on the EU policy for the sector which greatly influences the development of logistics chains and services. Mathematical formulation of typical transport and logistics-related problems is also presented followed by a discussion on the concept of sustainability.

A comprehensive review with 185 references for the analysis of commonly prescribed members of an important class of drugs, non-sedating antihistamines (NSAs), is presented. The review covers most of the methods described for the analysis of cetirizine (CTZ), ebastine (EBS), fexofenadine (FXD), ketotifen (KET) and loratadine (LOR) in pure forms, in different pharmaceutical dosage forms and in biological fluids. The review covers the period from 1991 till now.

One of the main challenges in personal communication service (PCS) is to locate many mobiles that may move frequently from place to place. Such a system operation is called location tracking. Many network signals flow, and database queries are required to achieve such a task. In addition to the two-level hierarchical strategy in IS-41 and GSM, several strategies have been proposed to improve the efficiency of location tracking. Pointer forwarding was used to reduce the expensive home location register (HLR) accesses. Previously, the distributed HLR scheme was proposed to prevent HLR from becoming a bottleneck in the signaling network. However, the length of a forwarding pointer chain may be lengthened in a distributed HLR environment. In this paper, we propose a more efficient strategy to overcome this potential problem. This strategy attempts to migrate the locating chains in a distributed HLR system when a mobile issues a registration operation. As a consequence, the length of any forwarding pointer chain does not exceed one in our strategy. Simulation results indicate that our strategy significantly decreases the locating cost. In fact, this strategy provides an upper bound of location tracking time owing to the fact that the length of any locating path does not exceed one. Furthermore, obsolete entries in local databases visiter location registers can be reclaimed in this strategy.

This pioneering study investigates the seventy Bruneian corporate customers' adoption intentions towards using the mobile services. The study uses and validates the VAM (value-based adoption model) for investigating mobile service in different geographical setting contrary to the original study for Mobile Internet. The study combines four external variables such as perceived usefulness, perceived enjoyment, technicality and perceived fee with perceived value in predicting the adoption intentions of using mobile services. The study results indicate that only perceived usefulness is significant predictor of perceived values which are significant indicator of adoption intentions of using mobile services. Based on the results, some recommendations are made for the managerial implications.

A reliable gas chromatographic mass spectrometric method has been validated for the determination of trace levels (<10 lgL À1 ) of patulin in apple products and quince jam. The method was based on extraction of patulin with ethyl acetate-hexane, alkalinisation and silylation with N,O-bis-trimethylsilyltrifluoroacetamide with 1% of trimethylchloro-silane. The accurate determination of patulin was achieved by employing commercial 13 C 5-7 patulin labelled as an internal standard, which allowed compensating target analyte losses and enhancement or suppression matrix effects. Limits of detection and quantification of method using real samples were 0.4 and 1.6 lgkg À1 , respectively. Recoveries of patulin from samples spiked at 8-50 lgkg À1 levels ranged between 71% and 89%. The repeatability of measurements (expressed as relative standard deviation) was lower than 16%. The method was successfully applied to the determination of patulin in apple fruit and apple products including juice, cider and baby food, and also in quince fruit and quince jam. A new PCR system for the detection of Penicillium expansum in samples containing highly degraded DNA was developed which permitted the detection of the mould in 2/3 of the samples containing patulin, including juices and jams.

The strAS or rpsL+ gene, encoding a ribosomal protein, S 12, expresses its streptomycin-sensitivity (SmS) phenotype dominantly over strAR or rpsL gene. Therefore, strAR cells that harbor plasmids with strAS alleles are phenotypically SmS. It was found that the SmS phenotype is unstable, and such cells eventually switch to the Sm-resistance (SmR) phenotype, especially when the strAS gene was cloned on high-copy-number (HCN) plasmids. It seemed that the strA gene cloned on HCN plasmids was toxic to Escherichiu coli host cells and, during prolonged cultivation, plasmids with an inactivated strAS gene, mostly carrying insertion sequence elements, such as ISI, IS5 and ~6, were selected. The instability of the strA gene was particularly enhanced when the Va151 residue in the middle of S12 protein was replaced by Leu, suggesting enhanced toxicity of the altered S12. Since the strAS gene was stably maintained throughout approx. 100 cell doublings when its expression was abolished, most probably it is the gene product rather than the nucleotide sequence itself that is responsible for the instability of strA gene on HCN plasmids. To improve the stability of the SmS phenotype, the previously reported ampicillin-resistance-conferring and Sms-enforcing plasmid vector, pHSG670, was reconstructed. The resulting vector, pHSG683, confers chloramphenicol resistance, enforces SmS on strAR and supE_ host bacteria, and has multiple cloning sites within the coding region of synthetic rpsL gene. When pHSG683 DNA was prepared from strAR and sup+ cells grown in tryptophan-rich medium with Cm and Sm, less than 10m6 plasmids failed to enforce SmS on strAR and supEm cells in tryptophan-less medium with Cm. This vector is extremely powerful and useful for efficient detection of the rare incidence of DNA recombination or genetic alterations in vitro and in vivo.

DATAID-is a database design methodology developed as part of the DATAID project financed by the Italian National Research Council within the Progetto Finalizzato Informatica research project. Several working groups, both from academic and industrial environments, participate in the DATAID project. At present, the authors coordinate the project. The importance of developing methodologies and tools for database design has been indicated in recent years by the constant growth of computer-based systems which involve a database to provide their users with a controlled and centralized access to data. Accordingly, the DATAID project was undertaken in September 1979 with the objective of developing a methodology to support the specification, design and implementation ofdatabases, including the construction of tools that would support the designer. In this study the main features of DATAID-I are discussed with respect to other approaches existing in literature. A later article will completely describe a case study in DATAID-I [I].

To assess the perceptions of healthy women of their genital anatomy and sexual sensitivity, and to provide suggestions for genitoplasty based on this information, as the success of genitoplasty has historically relied upon the surgeon's perception of the patient's anatomy and function, rather than the patient's perception of outcome in terms of appearance and erotic sensitivity. SUBJECTS AND METHODS Fifty healthy, sexually active, adult women (aged 20-56 years) with no history of genital surgery completed the female version of the Self-Assessment of Genital Anatomy and Sexual Function. This selfreport questionnaire comprises written text and images enabling women to rate the appearance, size and position of clitoris and vagina, as well as the intensity of orgasm and effort required for achieving orgasm in specified areas around the clitoris and within the vagina. Anatomical locations were compared for these ratings by repeatedmeasures analysis of variance. RESULTS Anatomically, 46% of women described their clitoris as 'moderate-sized and raised', 42% as 'small and raised', and 78% reported that their vaginal opening was adequate for sexual penetration. The women reported the strongest orgasm and least effort to obtain an orgasm with stimulation of the area on and above the clitoris. For vaginal sensitivity, scores for orgasm intensity increased, and for orgasm effort decreased, with increasing vaginal depth, and they indicated less sexual sensitivity for the vagina than for the external genitalia. CONCLUSION The skin above the clitoris, and the clitoris itself, appeared to be the most sexually sensitive. During genitoplasty, attention to preserving skin-flap integrity in this area seems appropriate.

Despite the implicit recognition of trust in IS outsourcing, comparatively few research could be found in this area. While addressing the nature of trust in IS outsourcing, we found surprisingly many aspects on the concept of trust which have not been captured in the mainstream research on IS outsourcing. This paper presents an interdisciplinary view on trust and attempts to call for a greater understanding and focus on the role and impact of trust in an outsourcing context. It is argued that trust does play a significant role in many aspects of IS outsourcing and potentially could be one of the key elements that contributes to the success of an outsourcing activity. The paper suggests a multidimensional approach to the exploration of trust in outsourcing relationships with a greater focus on multi-faceted aspects of trust as well as the mediating factors that influence the success and/or failure of IS outsourcing activities.

In 1998, Feeny and Willcocks published a core IS capabilities framework suggesting four tasks and nine capabilities for any future IT function. This paper revisits the framework, examining the challenges and learning points from its implementation in three organizations from 2000 to 2005. The cases, studied longitudinally, involved a medium-sized organization beginning to outsource, and a multinational and a national bank with international interests, both of which were several years into large-scale outsourcing arrangements. Longitudinal research revealed a range of omissions and resulting problems and underlined the importance of: retaining enough architecture planning and technical doing capability; ensuring future, businessoriented leadership tasks are fully resourced together with succession planning for these; ensuring informed buying and other capabilities to manage external supply and leverage business value from supplier performance are built; and ensuring that operational lynch-pin tasks such as relationship building (to business units) and contract facilitation are in place. In comparing findings for a medium-sized organization with those from two large-scale outsourcing contracts, the study notes the different challenges arising in organizations of different size and at different stages in their sourcing strategies.

The paper presents the research activity developed by the authors in the field of computer-aided inventive problem solving: an original model and a dialogue-based software application have been developed by integrating the logic of ARIZ (Algorithm for the Inventive Problem Solving) with some OTSM-TRIZ (General Theory of Powerful Thinking) models in order to guide a user also with no TRIZ education to the analysis of inventive problems. The paper demonstrates that through a dialogue-based interaction it is possible to guide the user towards a proper formulation of the problem statement, which is an essential step of any conceptual design activity. The proposed software system, although still at a prototype stage, has been tested with students at Politecnico di Milano and at the University of Florence. The paper details the structure of the algorithm and the results of the first validation activity; then, it discusses about the possibility to integrate the proposed approach into a new generation of CAD systems.

This paper describes a simple, fast, sensitive and reliable method for the simultaneous determination of albendazole sulfoxide (ASOX) and albendazole sulfone (ASON), the two most important metabolites of the drug albendazole (ABZ), in plasma samples using liquid chromatography and tandem mass spectrometry. After liquid-liquid extraction with dichloromethane, the two albendazole metabolites and the internal standard phenacetin were resolved in a CN column using the mobile phase methanol-water (4:6, v / v) acidified with 1% acetic acid. Detection by electrospray mass spectrometry was carried out in the positive ion mode. The method was linear up to 2500 and 250 ng / ml for ASOX and ASON, respectively, with mean recoveries of more than 85%. The precision and accuracy data, based on within-and between-day variations over 5 days, were lower than 15%. The quantitation limits of 0.5 and 5.0 ng / ml for ASON and ASOX are low enough for the method to be suitable for pharmacokinetic studies. Pharmacokinetic data obtained with the proposed method following oral administration of ABZ to a patient with neurocysticercosis are also reported. 

Activated charcoal (AC) is a sorbent that has been shown to remove urinary toxins like urea and indoxyl sulfate. Here, the influence of AC on kidney function of rats with experimental chronic renal failure (CRF) is investigated. CRF was induced in rats by feeding adenine (0.75%) for four weeks. As an intervention, AC was added to the feed at concentrations of 10%, 15% or 20%. Adenine treatment impaired kidney function: it lowered creatinine clearance and increased plasma concentrations of creatinine, urea, neutrophil gelatinase-associated lipocalin and vanin-1. Furthermore, it raised plasma concentrations of the uremic toxins indoxyl sulfate, phosphate and uric acid. Renal morphology was severely damaged and histopathological markers of inflammation and fibrosis were especially increased. In renal homogenates, antioxidant indices, including superoxide dismutase and catalase activity, total antioxidant capacity and reduced glutathione were adversely affected. Most of these changes were significantly ameliorated by dietary administration of AC at a concentration of 20%, while effects induced by lower doses of dietary AC on adenine nephrotoxicity were not statistically significant. The results suggest that charcoal is a useful sorbent agent in dietary adenine-induced CRF in rats and that its usability as a nephroprotective agent in human kidney disease should be studied.

In this study, secondary school teachers' acceptance of a digital learning environment (DLE) was investigated. Questionnaires were taken on three times (T1/T2/T3) during the same school year, with the Unified Theory of Acceptance and Use of Technology (UTAUT) as theoretical framework. Next to questionnaires, user-logs were collected during the entire school year. A total of 72 teachers completed a questionnaire on at least one occasion: 64 teachers responded at T1, 41 at T2, and 55 at T3. We first investigated which factors influence teachers' acceptance of a DLE. The main predictors of DLE acceptance were performance expectancy and social influence by superiors to use the DLE. Effort expectancy and facilitating conditions were of minor importance. We then investigated how well the amount of final observed use could be predicted, and found that at T1 about one third, at T2 about one fourth and at T3 about half of the variance in observed use was predicted by attitude, behavioral intention and self-reported frequency of use. Our study showed that to maximize use of a DLE, its usefulness should be demonstrated, while school boards or principals should strongly encourage teachers to (start to) use the DLE.
