Thyroid disorders are the most common endocrine disorders worldwide. PDE8B is strongly expressed in the thyroid and to a lower extent in the pituitary and hypothalamus. PDE8B gene affects the synthesis of T3 and T4 and regulates the release of TSH by the pituitary gland. Therefore, PDE8B single nucleotide polymorphisms (SNPs) rs 6885099 and rs2046045 play a crucial part in the incidence of thyroid disorders, but the existing data is insufficient and contradictory. Materials & Methods. Meta-analysis was carried out to assess the relationship between PDE8B SNP’s rs 6885099 and rs 2046045 and thyroid diseases. A meta-analysis of the associations between common variation in the PDE8B gene and thyroid hormone parameters, including assessment of longitudinal stability of associations over time and effect of thyroid hormone replacement Abstract Objective: Common variants in PDE8B are associated with TSH but apparently without any effect on thyroid hormone levels that is difficult to explain. Furthermore, the stability of the association has not been examined in longitudinal studies or in patients on levothyroxine (L-T 4 ). Design: Totally, four cohorts were used (nZ2557): the Busselton Health Study (thyroid function measured on two occasions), DEPTH, EFSOCH (selective cohorts), and WATTS (individuals on L-T 4 ). Methods: Meta-analysis to clarify associations between the rs4704397 single nucleotide polymorphism in PDE8B on TSH, tri-iodothyronine (T 3 ), and T 4 levels. Results: Meta-analysis confirmed that genetic variation in PDE8B was associated with TSH (PZ1.64!10 K10 0.20 S.D./allele, 95% confidence interval (CI) 0.142, 0.267) and identified a possible new association with free T 4 (PZ0.023, K0.07 S.D./allele, 95% CI K0.137, K0.01), no association was seen with free T 3 (PZ0.218). The association between PDE8B and TSH was similar in 1981 (0.14 S.D./allele, 95% CI 0.04, 0.238) and 1994 (0.20 S.D./allele, 95% CI 0.102, 0.300) and even more consistent between PDE8B and free T 4 in 1981 (K0.068 S.D./allele, 95% CI K0.167, 0.031) and 1994 (K0.07 S.D./allele, 95% CI K0.170, 0.030). No associations were seen between PDE8B and thyroid hormone parameters in individuals on L-T 4 . Conclusion: Common genetic variation in PDE8B is associated with reciprocal changes in TSH and free T 4 levels that are consistent over time and lost in individuals on L-T 4 . These findings identify a possible genetic marker reflecting variation in thyroid hormone output that will be of value in epidemiological studies and provides additional evidence that PDE8B is involved in TSH signaling in the thyroid.

Thyroid-stimulating hormone (TSH) controls thyroid growth and hormone secretion through binding to its G protein-coupled receptor (TSHR) and production of cyclic AMP (cAMP). Serum TSH is a sensitive indicator of thyroid function, and overt abnormalities in thyroid function lead to common endocrine disorders affecting~10% of individuals over a life span. By genotyping 362,129 SNPs in 4,300 Sardinians, we identified a strong association (p ¼ 1.3 3 10 À11 ) between alleles of rs4704397 and circulating TSH levels; each additional copy of the minor A allele was associated with an increase of 0.13 mIU/ml in TSH. The single-nucleotide polymorphism (SNP) is located in intron 1 of PDE8B, encoding a high-affinity cAMP-specific phosphodiesterase. The association was replicated in 4,158 individuals, including additional Sardinians and two genetically distant cohorts from Tuscany and the Old Order Amish (overall p value ¼ 1.9 3 10 À20 ). In addition to association of TSH levels with SNPs in PDE8B, our genome scan provided evidence for association with PDE10A and several biologically interesting candidates in a focused analysis of 24 genes. In particular, we found evidence for association of TSH levels with SNPs in the THRB (rs1505287, p ¼ 7.3 3 10 À5 ), GNAQ (rs10512065, p ¼ 2.0 3 10 À4 ), TG (rs2252696, p ¼ 2.2 3 10 À3 ), POU1F1 (rs1976324, p ¼ 3.9 3 10 À3 ), PDE4D (rs27178, p ¼ 8.3 3 10 À3 ), and TSHR (rs4903957, p ¼ 8.6 3 10 À3 ) loci. Overall, the results suggest a primary effect of PDE8B variants on cAMP levels in the thyroid. This would affect production of T4 and T3 and feedback to alter TSH release by the pituitary. PDE8B may thus provide a candidate target for the treatment of thyroid dysfunction.

In it, he based his conclusions on the theory of tendencies, arguing that Biden (or rather, those who stand behind him) would behave like an insensitive and distrustful paranoiac at a critical moment. He claimed this would bring great misfortune to the world, including Russia. That moment has now arrived. The American president has allowed Ukraine (formerly part of Russia) to use long-range missiles against Russian territory. This puts the world on the brink of disaster, as these missiles and their carriers were not produced by Ukraine but by the United States, Great Britain, and their allies, who have thus joined the conflict. "Who can lead the world except the United States of America?" the president declared on August 19. "<...> America is winning, and this is for the benefit of the world." This was not merely a statement for the American public. Biden spoke at the US Democratic National Convention on the eve of the presidential election during a time when America is desperately trying to redefine its leading role in the world to confirm its status as a superpower. This is happening as the world becomes irreversibly multipolar and begins to lose faith in the "American dream," the American currency, military power, and the political influence of the American elite, as well as its government institutions, education, and culture-everything that the so-called "Western world" was based on, if not the entirety of "Western and Christian civilization." Who could have written such nonsense for Biden? After all, we know from live interviews that presidents have not been responsible for creating or voicing their own ideas for a long time-since the Reagan era. As Ronald Reagan, the 40th President of the United States (1981-1989), admitted, a person enters the office each morning with a detailed plan of action and prepared speeches. "I don't know who he is, but he's the one who runs America!" Of course, this raises questions about who is managing him. Who exactly is that? Probably someone who knows more and is smarter than anyone else. But who determines their mental and heuristic abilities? Family, ethnicity, or other clans? Secret services? Please, don't make me laugh. These people are on the other side of good and evil. They seem more interested in playing hide-and-seek with the devil himself than in serving the highest ideals of humanity. And they don't care about the fate of the world or our children.

A sharp increase in the number of so-called hillforts in many parts of Late Bronze Age Europe has usually been considered a key indicator of rising levels of intergroup violence during the final centuries of the second millennium BC. With over a hundred Late Bronze Age hillforts, Ireland provides an excellent case-study region to test this hypothesis. What little hard evidence we have for the function of these sites only lends very limited direct support to viewing increased levels of intergroup violence as a dominant factor in Irish Bronze Age society and rather suggests that other phenomena, such as regular communal gatherings and feasting activities, may have played an equally or even more significant role in the social life of Irish Late Bronze Age communities, ultimately driving the appearance of-both enclosed and unenclosedhilltop sites during the final centuries of the second millennium BC.
