Nevertheless, Blepharomastix is a serious pest of okra plant as it feeds on the leaves, soft stems, fruit of okra and eventually damage okra production. Therefore, the aim of this study is to characterize the total cellulase and endo-β-1, 4-glucanase (digestive enzymes) present in the larvae of Blepharomastix as it is a popular trend to apply enzymes in biofuels production and application of enzyme inhibitors to protect crops from damaging throughout the world. The result analysis showed that the whole gut from the larvae exhibited the activities of total cellulase and endo-β-1, 4-glucanase enzymes that hydrolyzed crystalline cellulose and carboxymethyle cellulose (CMC) to glucose and the activities in insect were 0.076 µmol min-1 ml-1 and 0.398 µmol min-1 ml-1 respectively. However, the optimum temperature for the activity of total cellulase and endo-β-1, 4glucanase in Blepharomastix were 45˚C and 50˚C respectively. The pH optima of total cellulase and endoβ-1, 4-glucanase in Blepharomastix was 9.0. The cellulase activity was inhibited by ethylenediaminetetraaceticacid (EDTA), sodium dodecyl sulphate (SDS) and urea, whereas enhanced by NaCl, KCl and MgCl2.
Insect guts, particularly of phytophagous insects, are considered as intriguing bioprospecting so... more Insect guts, particularly of phytophagous insects, are considered as intriguing bioprospecting sources of cellulase and xylanase due to their use in biofuel industry. In this study, the activities of cellulase and xylanase were identified in the gut fluid of grasshopper, Oxya velox, and characterized. Qualitative assays of gut fluid carried out for endo-β-1,4-D-glucanase (EC 3.2.1.4) and endo-β-1,4-D-xylanase (EC 3.2.1.8) activities, using substrate-agar plate method, revealed clear transparent zones against the red-stained background. When measured by dinitrosalicylic acid method, the gut fluid had 0.759 ± 0.005 U and 0.303 ± 0.002 U of endoglucanase and endoxylanase activities, respectively, per mg of protein. In the gel zymogram, four distinct cellulolytic protein bands and one xylanolytic protein band were detected against substrates carboxymethyl cellulose and xylan, respectively. The optimal temperature and pH of both endo-β-1,4-D-glucanase and endo-β-1,4-D-xylanase were 55EC and 5, respectively. Preincubation at 70EC for 20 min resulted in almost complete loss of endo-β-1,4-D-glucanase activity as shown by zymography. The presence of both cellulase and xylanase activities suggested that O. velox could be considered as a model for studying the process of lignocellulose digestion in insect gut; the insect could also serve as a good source of enzymes for biofuel production.

CDC-48 is a AAA (ATPases associated with diverse cellular activities) chaperone and participates ... more CDC-48 is a AAA (ATPases associated with diverse cellular activities) chaperone and participates in a wide range of cellular activities. Its functional diversity is determined by differential binding of a variety of cofactors. In this study, we analyzed the physiological role of a CDC-48 cofactor UBXN-6 in Caenorhabditis elegans. The amount of UBXN-6 was markedly increased upon starvation, but not with the treatment of tunicamycin and rapamycin. The induction upon starvation is a unique characteristic for UBXN-6 among C-terminal cofactors of CDC-48. During starvation, lysosomal activity is triggered for rapid clearance of cellular materials. We observed the lysosomal activity by monitoring GLO-1::GFP, a marker for lysosome-related organelles. We found that more puncta of GLO-1::GFP were observed in the ubxn-6 deletion mutant after 12 h starvation compared with the wild-type strain. Taken together, we propose that UBXN-6 is involved in clearance of cellular materials upon starvation in C. elegans.

The fruits were Averrhoa bilimbi (Bilimbi), Artocarpus lacucha (Monkey jack), Cucumis melo (Mask melon), Phoenix sylvestris (Wild date palm) and Flacourita jangomas (Indian plum). P. sylvestris had the highest total phenolic content (37.40 ± 1.72 mg GAE/10 g of extract), whereas C. melo had the lowest (6.02 ± 0.89 mg GAE/10 g of extract). All the fruits showed DPPH free radical scavenging activity with the IC50 values for P. sylvestris, A. lacucha, F. jangomas, C. melo and A. bilimbi were 1.90 µg/ml, 0.798 mg/ml, 1.144 mg/ml, 1.695 mg/ml and 3.683 mg/ml respectively. Highest level of reducing activity was found in P. sylvestris (O.D. 0.933 ± 0.02) and reducing power activity was lowest in A. bilimbi (O.D. 0.249 ± 0.01) at a concentration of 0.4 mg/ml. The principle objective of this study was to investigate the presence of antimicrobial activities... more The principle objective of this study was to investigate the presence of antimicrobial activities in the larvae of insect Musca domestica (house fly). For this purpose, the body wall of the insect larvae were injured by a sterile hypodermic needle and then the larval extract were prepared in acidic (pH 5.0), neutral (pH 7.0) and alkaline (pH 8.0) buffers or water. These extracts were used for the screening of antimicrobial activities against 11 bacteria and 10 fungi by an agar disc diffusion assay and by poisoned food technique respectively. All the three extracts showed inhibition against 2 bacterial species viz., Salmonela typhi, Bacillus subtilis and 5 fungi viz., Macrophomia phaseolina , Fusarium equiseti , Colletrotrichum corchori, Alternaria alternate and Rhizopus species. The acidic extract also showed inhibition against bacteria Escherichia coli. Of the three extracts, acidic extract showed highest inhibition against all the microbes. Cytotoxic activity (LC 50 ) of the acidic, neutral and alkaline extract was found to be 141.2 µg/ml, 152.2 µg/ml and 149.5 µg/ml respectively. ___________________________________________________________________________

The eight day-old larva showed maximum activity with 1.73 U mg(-1) of protein, which was confirmed by gel zymography. In zymogram, using Carboxymethyl Cellulose (CMC) as substrate, four distinct cellulolytic protein bands were detected in leaf borer gut fluids through out of its development. The optimum temperature and pH were 60 degrees C and 5.0, respectively. This endo-beta-1,4-glucanase showed maximum stability at 20-45 degrees C with approximately 20% remaining activity. Zymography also showed complete loss of endo-beta-1,4 glucanase activity at 55 degrees C. This is the first report that the cellulolytic enzyme is produced in the gut of P. quatuordecimpunctata through the whole developmental stages, from the 1st instar to the adult, except for pupae.
The goal of phototherapy is to lower the concentration of circulating bilirubin or keep it from i... more The goal of phototherapy is to lower the concentration of circulating bilirubin or keep it from increasing in the treatment of neonatal unconjugated hyperbilirubinaemia. As phototherapy decrease billirubin level of infants, it is important to fix the dose of phototherapy. Objectives: The aim of the study was to compare the effectiveness of double phototherapy versus conventional single phototherapy in the treatment of neonatal unconjugated hyperbilirubinaemia. Materials and Methods: For this purpose 50 term and preterm newborns were enrolled. They were divided into 2 groups, 38 newborns were taken in group 1 and 12 newborn were taken in group 2. The babies who got conventional single phototherapy were taken in group 1. Each group was again divided into 3 subgroups according to their birth weight Normal Birth Weight (NBW), = Low Birth Weight (LBW) and Very Low Birth Weight (VLBW). Results: The serum bilirubin level of term babies were found 17.32 ± 4.08 mg/dl and in preterm babies were found 13.17 ± 1.49 mg/dl. Neonatal hyperbilirubinaemia in enrolled babies were due to physiologic jaundice (38%), neonatal sepsis (50%) and perinatal asphyxia (12%). The serum bilirubin level in physiological jaundice was found 18.47 ± 5.38 mg/dl, in neonatal sepsis was found 14.90 ± 1.85 mg/dl and in perenatal asphyxia was found 14.10 ± 1.47 mg / dl. Decline rate of serum bilirubin per day was found 4.58 ± 2.43 mg / dl in NBW babies with the use of conventional single phototherapy compared to 6.27 ± 2.26 mg / dl with the use of conventional double phototherapy. In LBW babies decline rate of serum bilirubin per day was found 2.07 ± 0.84 mg / dl with the use of conventional single phototherapy but 4.70 ± 2.08 mg / dl with the use of conventional double phototherapy. In VLBW babies, decline rate of serum bilirubin per day was found 2.24 ± 1.10 mg / dl with the use of conventional single phototherapy. Any VLBW babies which were given conventional double phototherapy were not found. Conclusion: From this study it can be concluded that double phototherapy is more effective than conventional single phototherapy in the treatment of neonatal hyperbilirubinaemia in both groups of neonates; however the rate of fall of bilirubin was higher in NBW subgroups.

The cellulolytic bacteria from the gut of three different phytophagous insects were studied to is... more The cellulolytic bacteria from the gut of three different phytophagous insects were studied to isolate novel cellulolytic organism for biofuel industry. Among the threse, gut of P. quatuordecimpunctata larvae contained both highest no of total bacterial count (6.8x10 7 CFU/gut) and cellulolytic bacteria (5.42x10 3 CFU/gut). Fifteen different isolates were obtained from the gut of O. velox, A. miliaris and P. quatuordecimpunctata. All the isolates produced clear zone in CMC medium staining with Congo red. The isolates included Gram positive Enterococcus, Microbacterium and Gram negative Aeromonas, Erwinia, Serretia, Flavobacterium, Acenitobacter, Klebsiella, Yersinia, Xenorhabdus, Psedomonas and Photorhabdus. Out of the fifteen isolated and identified bacterial species, twelve bacterial species were novel being reported for first time as having cellulase activity.

The isolates included Gram positive Enterococcus, Microbacterium and Gram negative Aeromonas, Erwinia, Serretia, Flavobacterium, Acenitobacter, Klebsiella, Yersinia, Xenorhabdus, Psedomonas and Photorhabdus. Out of the fifteen isolated and identified bacterial species, twelve bacterial species were novel being reported for first time as having cellulase activity.
Phenolic content, antioxidant and anti-amylase activities were studied in the ethanolic extract o... more Phenolic content, antioxidant and anti-amylase activities were studied in the ethanolic extract of five available Bangladeshi fruits. The fruits were Averrhoa bilimbi (Bilimbi), Artocarpus lacucha (Monkey jack), Cucumis melo (Mask melon), Phoenix sylvestris (Wild date palm) and Flacourita jangomas (Indian plum). P. sylvestris had the highest total phenolic content (37.40 ± 1.72 mg GAE/10 g of extract), whereas C. melo had the lowest (6.02 ± 0.89 mg GAE/10 g of extract). All the fruits showed DPPH free radical scavenging activity with the IC 50 values for P. sylvestris, A. lacucha, F. jangomas, C. melo and A. bilimbi were 1.90 µg/ml, 0.798 mg/ml, 1.144 mg/ml, 1.695 mg/ml and 3.683 mg/ml respectively. Highest level of reducing activity was found in P. sylvestris (O.D. 0.933 ± 0.02) and reducing power activity was lowest in A. bilimbi (O.D. 0.249 ± 0.01) at a concentration of 0.4 mg/ml. But all the fruits showed a dose-dependent increase in reducing power. The fruit extracts showed very weak inhibition of α-amylase activity. But highest activity found in case of P. sylvestris was 11.88 ± 3.69% and the lowest activity found for F. jangomas was 3.33 ± 0.64%. Considering the data, it can be concluded that among the five fruits P. sylvestris is the most health beneficiary.
