Testosterone

Testosterone: A steroid hormone which
determines the primary and secondry male
sexual characteristics.
 Site of synthesis: Testes,ovaries, and adrenal glands
Avalible forms: two forms endogenous
and exogenous
 Endogenous forms: Testosterone,DHT
 Exogenous forms:oxandronol stanozol
Pathway of synthesis: Multistep
pathway
 1. Release of GnRh from Hypothalmus
 2.Release of Lh from ant. Pituatary gland by action of GnRh
 3.Action of Lh on leydig cells of seminiferous tubule
Action of Lh on Leydigs cell:
 Lh binds to receptor on leydig cell
 Causes cascade of intracellular signalling
 Ultimately leydig cells releases Testosterone.
Testosterone blood transport:

 Mainly in three forms


 1.By binding to SHBG
 2.By binding to plasma albumin
 3.As free form
MOA:

 As testosterone is a steroid hormone it passes freely through cell membrane


 Binds to cytosolic receptor to form homone receptor complex
 Hormone receptor complex enters the nucleus binds to specific site on dna
which ultimately causes protein synthesis
Action of testosterone in body:
 Acts in two forms:
 1.without undergoing any biochemical change in organs like liver,adipose
tissues and and muscles.
 As DHT in tissues like skin and prostate gland by the action of 5-alpha
reductase on testosterone
Effect on liver:
 Increases liver protein synthesis
Effect on Bone:

 Increases Bone density


 Deficiency can cause osteoporosis
Effect on muscle mass:

 Has anabolic effect on muscle mass i.e increase muscle mass by increasing
muscle proteins.
 Indivisuals with low testosterone levels will suffer from muscle weakness.
Effect on mode and libido:

 Normal levels stabalizes mode,increases libido,and may have positive


cognitive ability
 Indivisual with low serum level will have opposite effect
Effect on sex organs:

 Normal levels causes penile growth,high sperm count and prostate growth
 Low serum concentration can cause erectile dysfunction, low sperm count
Effect on skin and heamatopoises:

 Causes hair growth in chest and genitalia.


 Causes skin thickening
 Increases heamatopoises will low levels can cause anemia
Therapeutic uses:

 Used to treat hypogonadism


 Useful therphy in osteoporosis.
 Used to treat wasting syndrome
 Used to treat severe burns
 Pituitary dwarfism
Toxicities:

 In male high serum conc. Can caue androgenic alopecia and


gynaecomastia.
 In females high level of testosterone causes hairsutism and dysmenorrhea
Anti testosterone:

 GnRH inhibitors: e.g leuprolide used mainly in prostatic carcinomas


 Receptor inhibitors: flumatide use in treating prostatic carcinomas
 5-alpha reducatase inhibitors: e.g finasteride usefull in treating BPH and hair
loss
