• Take out epidermal peel from upper
and lower surfaces of Petunia leaf
and put them in separate watch glasses
•Stain the peels with safranin by adding few drops of the stain in the watch gluHS

•Take the peels and mount them in glycerine on sen~9'!1te slides cq'

. Cover the peels with coverslips and •
observe under the microscope
•Count the numbers of stomata and the numbers of Petunia
epidermal cells in the peels of both upper and lower
epidermis of the leaf appearing in the microscopic field

• Similarly prepare the slides of the peels of lily and count the numbers of
stomata and numbers of epidermal cells in the microscopic field
