Basic Understanding of

1.0 Features of NIIN Designs

Continuing with its history of innovations, NIIN provides leading Fluidize Bed Granulator system is
an advanced, multifunctional system designed to perform three distinct functions in one machine:
powder spray granulation, pellet coating, and pellet growing. This machine is suitable for a
versatile production environment, reduces labor requirements and eliminates additional
equipment. Conventional machinery requires multiple units to complete the same tasks. When
binder solutions are added to the fluidized powder bed, fluidization characteristics can be
determined by air flow, pressure, temperature, and spray parameters, all of which can be adjusted
by the user for optimal results.

The design of this machine reduces the risk of contamination during transport of in-process
materials and saves manpower and production time. The machine creates uniform pellets and
excellent flow properties, resulting in better control in the downstream tablet processing.
Machine operation is simple and ergonomic; operators can be easily trained. Furthermore, easy
to clean, and easy to maintain are also its key advantages. After all, its excellent functional
advantages are aimed to reduce the user’s total cost of ownership.

Model NI-EXE-DB-30T is suitable for use in:

Pharmaceuticals: Enzymes, antibiotics, proteins, fermentation products, drugs, herbal

medicines, vitamins

Explosion Barrier Valve

The explosion barrier valve prevents automatically and without external energy supply the
propagation of a dust explosion beyond the
installation point and acts as a mechanical flame trap.
explosion barrier valves are suitable for application in
dust transport systems, for dust explosion categories 1
and 2. The use of the valves as barrier against
explosions of dust category 3 and hybrid gas mixtures
is subject to clarification with the manufacturer.
Function
The movable internal valve poppet is suspended in
central position by two springs. The maximum

permissible velocity of air is 20 m/s

relative to the nominal width of the
connection pipe. In case of an explosion
the valve closes automatically by the
kinetic energy of the pressure wave,
which moves ahead of the flame front.
In the process the internal valve poppet
is hurled to the rubberized valve seat. A
mechanical shut-off device
simultaneously retains the valve in an
airtight position "SHUT" and acts as a
flame trap. Limit switches which signal
the "SHUT"-position at random monitoring points, can be supplied upon request.

1.1 Main Body

The main body is comprised of a retarding chamber and a product container receptacle
underneath the chamber. The retarding chamber is constructed from 316L stainless steel
and is the core of the Fluidize Bed system. A depiction of the main body is shown

Page 29

The retarding chamber is comprised of an upper filter chamber and a lower drying chamber.
The filter chamber includes filter bags sealed around the
inner circumference of the chamber and a bag shaking on
top. A sight glass provides visibility into the filter chamber.
The drying chamber is the chamber volume beneath the
filter bags and is open at the bottom to connect to the
product container. The drying chamber includes three
large access ports: the front access port is installed with a
sight glass for visibility into the chamber; two side access
ports are installed with blind plates. When equipped
with the granulation function, a blind plate can be
removed and installed with solution spray guns; when
equipped with the automatic product in-feed function, the
blind plate can be removed and installed with an in-feed
port; when equipped with the wash-in-place (WIP) function,
the blind plate can be removed and installed with the
WIP spray balls or nozzles.

The retarding chamber has been designed for operating

pressures up to 2 kg/cm2.

The product container receptacle is fitted with a circular

gasket around its circumference. To raise the product container, compressed air pressurizes
the hidden cavity underneath the circular gasket, raising the gasket and the container. On the
bottom rim of the retarding chamber, a silicon soft gasket seals the gap between the product
container and the chamber. Since the rise mechanism and the seal due not require the use of
lubricants or chemical sealants, there is no risk related contamination.

In operation, filtered air from the air handling unit)enters the main body through its air inlet
behind the product container receptacle. Air flows from the product container receptacle,
through the product container, drying chamber, bag filter assembly, filter chamber, and exits
through the its air outlet on top of the filter chamber.

1.2 Bag Filters, Filter Lift, and Shaking System

The bag filters are made of antistatic (conductive) polyester fabric with stain weaves. The bag
filter assembly is comprised of a top ring bracket for holding the bag and sealing ring with an
inflatable sealing gasket at the bottom. When installed in place, the fabrics
are grounded to allow the discharge of static generated during the drying process and
the sealing gasket inflates to create a tight seal on the retarding chamber’s inner wall. A

The shaking system and the filter lift are controlled by the same linear actuator. A schematic
of the shaking system is shown. When the linear actuator is extended the lift cable is
lowered. A diret cylinder for all motion movement.
At the same time, the response of the air outlet damper is also linked to the shaking
sequence. When the Pause Time has elapsed, the air outlet damper is closed, shaking timer
is started, shaking actuator engages in an up-and-down motion, and the accumulated
powder on the bag filter is shaken off. When the Shaking Time has elapsed, the shaking
actuator stops and retracts to its default position, the air outlet damper opens, and the
pause timer is started. The purpose of the shaking sequence is to shake off accumulated
powder on the bag filter, which blocks exhaust air flow; however, shaking disrupts
fluidization in the drying chamber, therefore the system is designed with the intermittent
shaking sequence.

1.3 Product Container

The product container is constructed of 304 or 316 stainless steel. The bottom of the
container is made of a perforated sheet, on top of which is a fine mesh. A sight glass
on the front side of the container is available for visibility, and a sampling port on the side
of the container allows in-process sampling. Below the sight glass on the bottom rim of
the container is a J-hook; on the two sides of the container’s top rim are two buckles.
Both the J-hook and the buckles are used to secure the container to the main body once
the container is raised in place and keep the container securely in place in explosion
scenarios. Finally, the product container is attached to a castered stainless steel frame
for mobility.

When raised in place, the product container is grounded to allow the discharge of static
generated during the drying process. In operation, air enters the product container
through its bottom mesh screen. The product is fluidized by this stream of incoming
air inside the enclosed space between the drying chamber and the product container.

1.4 Damper System

The damper system consists of a mechanical damper on the top-right corner of the fluid bed
dryer’s upper enclosure, actuated by a rotary actuator, per Figure 7-13. The damper system
acts as a gate to the incoming and outgoing air flow. The damper’s open-and-close is
controlled automatically by the PLC. On “open”, the percentage opening is set manually by
the operator.

1.5 Air Handling Unit (AHU)

The heater unit (heater unit) consists of (in order of sequence) a 60% prefilter, dehumidifier
(optional), heater, 95% HEPA filter, and a 99.97% HEPA final filter, per Figure 7-14, and all
inside a stainless steel housing. The output hot air passes through the automatic damper
into the main body. The HEPA efficiency rating may be different based on user specifications.
When the AHU is supplied without the dehumidifier, it is also referred to as the Heater Unit.

H2. Dehumidifiy System

The dehumidifier is constructed of copper tubing and aluminum fins. Plant-supplied chilled
water supply passes through the copper tubing. Water vapour from the inlet air condenses as
it passes through the copper tubing and aluminum fins. Condensed water is
drained. A downstream humidity sensor feeds back humidity measurements and modulates a
3-way control valve that controls the chilled water supply and return flow. As a reference, chilled
water continuous supplied at 7°C can attain an absolute humidity of approximately 10g/m3.

H3. Air Mixing System

The Air Mixing System is a self control in AHU Damper for cooling air (above section) and heating
air (lower section) mixing the air for stable temperature. The damper system provides quick
heating and quick cooling with by-pass its function if not in function. With Pneumatic auto control
valve, PID controls the motion of the system by PLC calculation.

H4 Heater and Temperature Control

Based on user specifications, the heater may be heated by electricity or by plant

steam.

For plant steam-heated units, prior to entering the heater, plant steam
supply passes through a pressure-reducing valve to maintain the inlet pressure at

around 1.5 to 2 kg/cm2. Pressure gauges before and after the pressure-reducing valve
provide pressure readings.
Steam on-off control is modulated by a temperature control loop, which maintains the
temperature of the outlet air at setpoint ±1.5°C. The temperature control loop includes
the inlet air temperature sensor (located at the fluid bed dryer’s air inlet duct) and an
overheat temperature sensor (located at the heater unit’s air outlet). Steam on-off control
is based on the feedback from the inlet air temperature sensor. The overheat
temperature sensor measures the air temperature against the setting value,
above which the heater will be forced shut and an alarm message will appear on the HMI
screen. This overheat alarm is intended to protect the heater from damages.

1.6 Exhaust Cartridge Dust Collector

The dust collector is used to collect granular solids from the Fluidize Bed ’s exhaust air.
Dust collection is essential in minimizing pollution, cross-contamination, and safety risk of the
operators.

The cartridge dust collector system consists of 16 cartridge-type filters in 8 accessible cartridge
wells. “Dirty” air from the Fluidize Bed enters the dust collector from the top, passes through
the filter media, and “clean” air exits the dust collector through its bottom-side outlet. Dust
is collected on the surface of the filters; excess dust falls off the surface of the filters and is
collected in the portable waste bin through the bottom hopper. See Figure 7-16 for a
schematic of the system.

Cleaning is carried out with pulses of compressed air to

flush off dust accumulated on the filter cartridges, one
set at a time. Compressed air is released from the
“clean” side of the filters against normal air flow
direction). Dust on the “dirty” side of the filters are
blown off and collected in the waste bin. In
automatic mode, pulse intervals can be set by the user;
in manual mode, the user can toggle the compressed air
valve manually.

1.7 Exhaust Filter Chamber and Exhaust Blower

Exhaust air from the dust collector passes through an exhaust filter chamber for
fine-filtration and is finally expelled to the atmosphere through the exhaust blower.
Fine filtration provides an additional layer of pollution, cross-contamination, and safety
control. The user may specify different grades of filters depending on the nature of the
product. Typical design includes an initial, 95% HEPA filter and a final, 99.97% HEPA filter.
A schematic of the blower is shown in Figure 7-18.

2.0 PRINCIPLE OF OPERATION

With modular design, NIIN’s Fluidize Bed systems can be configured with different
dehumidification, heating, air handling, solution delivery, coating, spraying, dust collection,
and programmable logic control modules. The end user will also have the option to
change the coating pan on demand, either for use with different batch sizes, different
products, or to minimize the risk of cross-contamination. The result is a flexible, efficient
system suitable for a wide variety of coating applications, such as film coating, enteric
coating, sugar coating, controlled release coating, and granule coating for the
pharmaceutical industry, chocolate, chewing gum coating for the food industry, and catalytic
agents coating, seed coating, fertilizer coating, and feed coating for the agrochemical
industry. Built-in wash-in-place (WIP) system provides automated cleaning of interior
and exterior surfaces of the coating pan and the air outlet duct with purified water The
automated WIP system is designed to save cleaning time and manpower, and to reduce the
likelihood of human errors.

All components in the system that have direct contact with the product are constructed of
non-reactive, non-shedding, 304 or 316 stainless steel with Teflon or Silicon gaskets.
Welding of stainless steel components are carried out by qualified technicians, and proper
inspection, polishing, acid-treatment, and cleaning are performed at NIIN’s facility in
Taoyuan, Taiwan, prior to shipping. Pressure piping, including steam and water piping,
have been designed and pressure-tested in accordance with ASME B31.3 (ii) for safety.

5.1 Proportional-Integral-Derivative (PID) Control

This system utilizes Propotional-Integral-Derivative (PID) control algorithm for selective

analog feedback control loops. A PID controller can provide excellent response time and
accuracy, minimum overshoot, and user-adjustable control parameters suitable for a wide
range of input and output conditions.

The main component of a PID controller consists of a setpoint, a process output, a “P” value,
an “I” value, and a “D” value, as depicted in Figure 5.1. Aside from the output, which is a
measured value, all other values are user-defined. The controller output depicted in Figure
5.1 is a value calculated by the controller and fed into the process to arrive at the process
output. For example, for a typical temperature PID control, the setpoint would refer to a
user-defined temperature setpoint, the process output would refer to the actual
temperature measurement, and the “P”, “I”, and “D” values are user-defined values that
may be arrived by experience or trial.

diagram

“P” Value: Proportional action is the action where the controller output varies in proportion
to the deviation between the setpoint and the process output. If the proportional band
(“P” value) is narrowed, it generally helps improve response time even with small changes to
the process or the setpoint. However, if the proportional band is narrowed too much, it
could become over-sensitive to small disturbances like signal noise.

Page 47

E-mail: [email protected] http:// www.niin.com.tw/En/ Date. : 2014 01 03

“I” Value: Integral action is the action where the controller output varies in
proportion to the time-integral of the deviation between the setpoint and the process
output. When the integral time (“I” value) is shortened, it generally helps
narrowing the response offset sooner. However, shortening the integral time
can also result in response oscillations.

“D” Value: Derivative action is the action where the controller output varies in
proportion to the time-derivative of the deviation between the setpoint and the process
output. When the derivative time (“D” value) is shortened, it generally helps restore
response overshoots more in a smoother but slower fashion. When the derivative time
is extended, the response overshoots are reduced faster but it could result in reverse
overshoot and oscillations.

For best results with manual adjustments, it is suggested to gradually vary the “P”, “I”,
and “D” values and observing the responses until an optimal response is obtained.
Alternatively, use the PID auto-tuning feature described in the following section.

5.2. PID Auto-tuning Feature

The equipment may feature auto-tuning feature for the PID controllers. The auto-tuning
feature may be used when temperature or humidity control becomes too slow or too fast, the
actual output deviates increasingly from the set point, or actual output fluctuates excessively
(which can also pose potential damage to the equipment or risk to the operator). The
auto-tuning algorithm may react differently to different pre-conditions, examples of the
responses when the initial condition is below the set point, at the set point, or above the
setpoint are shown in Figure 5-2-1, Figure 5-2-2, and Figure 5-2-3, respectively.

outlet of AHU-01, issues alarms on high
temperatures to protect the heater from overheating.

4. Air enters the main body through the air inlet

behind the product container receptacle, flows
through Air Distributor and main chamber, bag filter
assembly, and exits through the its air outlet on top
of the filter chamber. As air passes through the the
product container, the product within is lifted or
“fluidized” by the air in drying chamber and gradually
dried. Filter Bag blocks the product but allows air
to pass through; product remains in the drying
chamber. Some materials will build up under filter bag, preventing a smooth air
flow. Therefore, Shaking Cylinder generates a periodic shaking motion through
repeated extension and retraction motions in order to shake off materials
accumulated under filter bag.

5. Air then travels through Inlet Damper, which is HMI adjusted. Container screens for
powder in the air stream; if found, an alarm is issued.

6. Air enters the Filter Bag to remove gross particles, and then through Dust Collector to
remove fine particles, before being exhausted through Blower.

7. Suction proceeds for a timed period set by Primary Time.

5.4. Dust Collection Sequence

This section describes the dust collection sequence. Dust collection sequence is carried
out whenever the dust collector is switched on in automatic mode, including during the
drying sequence and the granulation sequence.

1. As Dust Collector are turned on and air enters, some materials will build up on the
bag filters. Therefore, after a 10-second pause time (default; user-changeable) the
reverse air flush valve opens for 100 milliseconds (default; user-changeable) to blow
off the accumulated materials on the top row of bag filters.

2. Following another pause period, reverse air flush is carried out for the next row of
bag filters for 100 milliseconds.

3. After the bottom row has been flushed, flushing resumes from the top row.
The cycle continues until Dust Collecor is shut off.

5.0 Maintenance Service

A Preventive Maintenance (PM) program is a required component of a compliant Quality System
for the safe and proper operation of the equipment. When properly carried out, a good PM
program can also maximize the equipment’s operational time by minimizing down time.

8.1 First-Time Startup Maintenance

After starting up the equipment for the first time, and after running the equipment for one
week, the following maintenance work are recommended:

- Verify the all nuts, bolts, fittings, flanges, valves, and tri-clamp fittings remain tight.

- Clean all utility supply strainers and check the state of all filters.

- Clean the all filters and look for signs of excessive dust or debris

After operating the equipment for one month, the following maintenance work is recommended:

- Replace the gear reducer oil. If the equipment has been used heavily, the oil should not wait
until one month into its usage, but be replaced after 300 hours.

After operating the equipment for three months, the following maintenance work is
recommended:

- Replace the gear reducer oil. If the equipment has been used heavily, the oil should not wait
until one month into its usage, but be replaced after 1000 hours.

8.3.1. General and Safety Notices

Hydrostatic pressure testing is a statutory requirement for pressure vessels in many

jurisdictions. Many also require hydrostatic pressure testing at fixed periods. Consult the local
regulation for pressure testing requirements.

8.3.2. Recommended Pressure Testing Equipment

Hydrostatic pressure booster pump. Up to 12 bar is recommended.
Associated hoses, connectors, tri-clamps, gaskets and other connectors that are suitable for the
temperature, pressure, and chemical used in the process.

Pressure gauge to monitor the pressure.

8.3.3. Pressure Testing Procedures

The procedures described herein are for a general scenario only.

1. Reference the pressure vessel or piping working pressure that are indicated on the boiler
plate affixed to the vessel or on manufacturer’s documentation.

2. Ensure the equipment and all its supply and return utilities have been shut down or closed
off.Allow the equipment to cool to ambient temperature, and all piping and vessels to equalize to
atmospheric pressure. Drain the system.

3. Using hoses and tri-clamps, connect the inlet and outlet of the equipment section to be
tested to the pressure booster pump to create a continuous circulation loop. Install a
pressure gauge in the loop to monitor the loop pressure and an air removal valve.

4. Fill the vessel with the same grade of water that is used in the process (e.g. DI water for a
DI water loop). When water draining from the air removal valve turns air-free, shut off the
valve.

5. Pressurize the vessel with the pressure booster pump to ½ (one half) the normal working
pressure. Inspect for leaks and abnormality.

6. If the vessel is free of leaks and abnormality, continue to increase the pressure to the
normal working pressure. Inspect for leaks and abnormality.

7. If the vessel is free of leaks and abnormality, continue to increase

the pressure to 1½ (one-and-a-half) the normal working pressure. Inspect for leaks and
abnormality.

8. Hold the pressure and monitor for a period of one hour. The pressure should not drop
during this period.
