Microscopy

ARRIVAL PROCEDURES

 Put on Face Mask


 Find Work Station
 Place personal belongings in cabinet near
Work Station
 Roll Up Sleeves and Tie Back Hair, if necessary
 Wash Hands
 Put on Lab Coat
o Roll up sleeves, if long

 Put on Goggles
o Select pair from white cabinet near autoclave room

 Put on Gloves
o Be careful not to touch your face, hair, face mask, or goggles with
gloves on
o Change gloves whenever they become contaminated

 Disinfect Work Station


Microscopy

Simple Stain

Prepare a Smear:
1) Collect sample with clean wooden stick
and apply to slide, rough side up
2) Dispose of swab/stick in small biohazard
container on table
3) Air-dry slide COMPLETELY (may use
slide warmer for this step)
4) Heat-fix, using clothespin

Simple Stain:
1) Cover smear with methylene blue for 1
minute
2) Rinse stain off gently with wash bottle
3) Remove clothespin, and pat slide dry
with Bibulous Paper
4) Observe with microscope

HANGING-DROP
1) Place slide and coverslip separately on top of “Kimwipe” tissue
2) Put a dot of Vaseline on the 4 corners of the cover slip
3) Collect 2 loops of Proteus mirabilis and place on a cover slip:
a) Flame loop
b) Open tube, flame lip
c) Collect first sample
d) Flame lip, close tube
e) Apply sample to cover slip, repeat steps a)-e) at least one
more time
4) Carefully attach a depression slide to the cover slip
5) Turn, so cover slip is on top
6) Observe on HIGH POWER, before proceeding to Oil Immersion
7) When you are finished, place slide and cover slip both in a
designated Beaker of Disinfectant on Hanging-Drop Cart,
DO NOT Discard Depression Slides into Sharps Container!!!

IF YOU CANNOT SEE BACTERIA MOVING, DO NOT MAKE A FRESH SLIDE UNTIL YOU TRY
THE TRICKS BELOW:
1) Use low light: Keep your lamp BRIGHT, but close the IRIS DIAPHRAGM
2) Locate the edge of your drop on SCAN. The drop will appear grainy. The graininess
is the bacteria
3) As you increase magnification, keep the edge of the drop in view and increase light
as needed
4) If you see a grainy background that DOES NOT MOVE when you move your slide,
RAISE OR LOWER THE CONDENSOR to eliminate the background
5) If you see images that move when you rotate your OCULAR LENSES, clean the
ocular lenses with lens paper, and open the IRIS DIAPHRAGM to let in a bit more light

IF YOU STILL CANNOT SEE BACTERIA MOVING, DO NOT MAKE A FRESH SLIDE UNTIL YOU
CHECK WITH YOUR INSTRUCTOR

CLEAN UP
 Submit Lab Data Sheets – front lab bench

 Hanging-Drop Depression Slides – Disinfectant Beaker on


Hanging-Drop Cart
 Simple Stain Slides – Sharps Bin (red box on tabletop)
 Box of clean, unused slides – Label with your name and
place in drawer at work station
 Stains – Stain Waste Jug in Fume Hood
 Test Tubes – Remove labels and place in rack next to
autoclave
 Paper Towels – Trash

 Disinfect work area with disinfectant in spray bottle, wipe


with paper towels.
 Clean Microscope ocular and objective lenses with Formula
409 and lens paper.
 Clean Microscope stage, mechanical arms, and adjustment
knobs with Formula 409 and paper towels
 Check that the Microscope 4x/Scanning Objective is in the
downward position

 Lab Coat, Goggles – Plastic bag labeled with your name

 Gloves – “Gloves Only” bin beside sink


 Wash hands
