II. Questions
1. Based on the graph constructed in Part A, give a statement relating enzyme activity to substrate
concentration.
As the substrate concentration increases, the enzyme activity also increases. An optimum rate is reached at the enzyme’s optimum substrate

concentration.

2. Explain why at 70oC enzyme activity decreased.


The rate of an enzyme-catalyzed reaction increases as the temperature increases, as it does with many chemical reactions. The rate, however,

decreases at high temperatures because the enzyme becomes denatured and can no longer function.

3. On what type of solution – acidic, basic, or neutral – will polyphenoloxidase work best. Explain your
answer.
Since polyphenoloxidase is dependent on the pH, it works best on a neutral solution. The ideal pH of a solution should be close to the pH of 7

because more acidic and basic solution will slow the reaction and will cause denaturation of the polyphenoloxidase enzyme, which greatly affect its

function.
