Proposal Title: Possible antiviral drug for Covid-19 patients

RATIONALE

Let us be really rational about this pandemic that every country is now facing . The Covid-19
is not going to vanish in the near future. This virus has been shown to be extremely
infectious with a fast incubation time in humans that cause hundreds of new cases everyday
here in the Philippines. That's why the government is keen to discover a way to suppress the
replication of the virus, and this research aims to develop possible antiviral medications.

REVIEW OF LITERATURE

The novel coronavirus disease or COVID-19 caused by SARS-CoV-2 infection has spread
quickly in many countries worldwide. The transition from early signs to acute respiratory
distress syndrome is very likely to be due to uncontrolled production of cytokines and it can
be dangerous due to an overreaction of the human immune system (A. George,2020). An
antiviral drug called chloroquine (CQ) is being developed for the medication of malaria and
HIV-1/aids, a drug well known for its effectiveness in the treatment of autoimmune disease,
and insights into its antiviral activity have also been obtained from in vitro studies (Dr. A.
Savarino et al,. 2006). Also, there’s a Hydroxychloroquine (HCQ) which has the same
chemical formula with chloroquine is known to have a strong immunomodulatory potential
which really stops flare-up inflammation and organ damage (Dan Zhuo et al., 2020).

HCQ and CQ,in terms of immune regulation is said to inhibit the binding of receptors and
membrane fusion, two main steps should be taken for coronavirus cell entry. CQ has been
shown to have an antiviral effect throughout pre-and post-infection conditions by
suppressing angiotensin-converting enzyme 2 (ACE2) glycosylation (SARS-CoV cellular
receptor) and blocking virus fusion fast (Dan Zhuo et al., 2020).

OBJECTIVES

The objective of the study is emphasize the possible effectiveness of hydrochloroquine


(HCQ) and chloroquine(CQ) for the treatment of fast growing number of Covid -19 patients.

METHODOLOGY

Since, hydrochloroquine is already a drug which is known as Plaqueniel, and used as to treat
malaria and to treat rheumatoid arthritis,it might also used as antiviral drug for Covid-19
patients. In addition, HCQ can improve intracellular pH and suppress lysosomal activity in
antigen-presenting cells (APCs), including plasmacytoid dendritic cells (pDCs) and B cells,
therefore preventing the processing of antigens and major histocompatibility complex
(MHC) class II-mediated autoantigen presentation to T cells (Dan zhouet al,.2020)

HCQ is expected to reduce the extreme development of COVID-19 by suppressing the


cytokine storm by reducing CD154 production in T cells. Since CQ and HCQ share similar
chemical structures and pathways to serve as a weak base and immunomodulator, it is
simple to summon up up the possibility that HCQ might be a potent agent for SARS-CoV-2
infection.

Since it is available drug in the market, test it to Covid-19 patients and more laboratoy test in
vitro is necessary. There is an immediate need to distinguish the effectiveness of this drug.
