Formulation of paracetamol-containing pastilles with in situ coating technology

Katona, Szalontai, Budai-Szcs, Csnyi, Szab-Rvsz, Jjrt-Laczkovich

Abstract

The focus of this research was to apply the in situ coating technology for producing paracetamol- (PCT-)
containing pastilles for pediatric use from a eutectic of two sugar alcohols (sorbitol, xylitol) in one step. This
type of melt-technology is more cost-efficient and simpler than other conventional tableting technologies,
whereby the formation of the pastilles and their coating occur upon the same fabrication step.

Thesis Statement:

A paracetamol containing tablet is available as a chewable one in pediatric therapy with the aim of easy
administration and fast release. Although it is most often used for a localized painkiller effect in the mouth,
it can also be used for systemic therapy provided that the drug is in contact with the oral cavity for an
extended period or is swallowed. By means of buccal absorption, several disadvantages of oral
administration can be avoided, including the hepatic first-pass metabolism or enzymatic degradation within
the gastrointestinal tract, which prevent the oral administration of certain classes of drug particularly of
protein and peptides.

Abstract

The objective of the study was to reveal the influence of necessarily added barium sulfate (BaSO4) X-ray
contrast material on floating drug delivery tablets. Based on literature survey, a chosen floating tablet
composition was determined containing HPMC and carbopol 943P as matrix polymers. One-factor factorial
design with five levels was created for evaluation of BaSO4 (X1) effects on experimental parameters of
tablets including: floating lag time, total floating time, swelling-, erosion-, dissolution-, release kinetics
parameters and X-ray detected volume changes of tablets

Thesis Statement:

In order to evaluate and compare floating preparations, imaging techniques such as gamma scintigraphy, x
ray radiography, or more advance x ray computed tomography may be used to track in vivo floating a
gastroretention. X ray imaging utilizes barium sulfate as an opacifying agent and in some cases, it can alter
the absorption of other drugs.

Mesoporous silica nanoparticles in target drug delivery system: A review.

Bharti, Nagaich, Pal, Gulati.

Abstract

Due to lack of specification and solubility of drug molecules, patients have to take high doses of the drug
to achieve the desired therapeutic effects for the treatment of diseases. To solve these problems, there
are various drug carriers present in the pharmaceuticals, which can used to deliver therapeutic agents to
the target site in the body. Mesoporous silica materials become known as a promising candidate that can
overcome above problems and produce effects in a controllable and sustainable manner

Thesis Statement:

In particular, silica nanoparticles are widely used as a delivery reagent because silica possesses favorable
chemical properties, thermal stability and biocompatibility.
