The aim of this in vitro study was to evaluate the effects of nicotine, cotinine, and caffeine on the viability of some oral bacterial species. It also evaluated the ability of these bacteria to metabolize those substances. Single-species biofilms of Streptococcus gordonii, Porphyromonas gingivalis,o rFusobacterium nucleatum and dual-species biofilms of S. gordonii -F. nucleatum and F. nucleatum -P. gingivalis were grown on hydroxyapatite discs. Seven species were studied as planktonic cells, including Streptococcus oralis, Streptococcus mitis, Propionibacterium acnes, Actinomyces naeslundii, and the species mentioned above. The viability of planktonic cells and biofilms was analyzed by susceptibility tests and time-kill assays, respectively, against different concentrations of nicotine, cotinine, and caffeine. Highperformance liquid chromatography was performed to quantify nicotine, cotinine, and caffeine concentrations in the culture media after the assays. Susceptibility tests and viability assays showed that nicotine, cotinine, and caffeine cannot reduce or stimulate bacterial growth. High-performance liquid chromatography results showed that nicotine, cotinine, and caffeine concentrations were not altered after bacteria exposure. These findings indicate that nicotine, cotinine, and caffeine, in the concentrations used, cannot affect significantly the growth of these oral bacterial strains. Moreover, these species do not seem to metabolize these substances.

O objetivo do presente estudo foi determinar os parâmetros farmacocinéticos da ropivacaína encapsulada em lipossomas após anestesia local em 14 voluntários sadios. Neste estudo randomizado, cruzado e duplo-cego, os voluntários receberam anestesia infiltrativa na maxila de ropivacaína a 0,5% encapsulada em lipossomas e ropivacaína 0,5% com epinefrina a 1:200.000 em duas sessões distintas. Amostras de sangue foram coletadas antes e após (de 15 a 1440 min) a administração das formulações de ropivacaína. A quantificação da concentração plasmática de ropivacaína foi feita por meio de HPLC com detecção por UV. Os parâmetros farmacocinéticos AUC 0-24 (área sob a curva de concentração × tempo do tempo 0 até 24 horas) , AUC 0-∞ (área sob a curva de concentração x tempo do tempo 0 até o infinito), C max (concentração máxima da droga), CL (clearance renal), T max (tempo em que ocorre a concentração máxima); t 1/2 (meia vida de eliminação) e V d (volume de distribuição) foram analisados pelo teste de Wilcoxon. Nenhuma diferença (p > 0,05) foi observada entre as duas formulações em cada parâmetro farmacocinético avaliado e as concentrações plasmáticas de ropivacaína, considerando cada período de tempo. Ambas as formulações apresentaram perfil farmacocinético semelhante, indicando que a formulação lipossomal poderia ser uma opção mais segura para o uso deste anestésico local, devido à ausência de vasoconstritor.

Study Objective: To compare the topical and the pulpal anesthesia efficacy of liposomal and plain benzocaine formulations. Design: Double-blinded, randomized crossover study. Setting: University ambulatory dental center. Patients: 30 ASA physical status I volunteers.

RESUMO A anestesia local é essencial para a realização da grande maioria de procedimentos em odontologia. Casos de mortalidade devido à anestesia local são raros. No entanto, casos de morbidade são mais comuns, mas nem sempre são relatados. Portanto, o cirurgiãodentista deve possuir conhecimento da farmacologia e da toxicidade das soluções anestésicas locais, com o intuito de evitar possíveis complicações sistêmicas decorrentes da sua administração.

RESUMO Este trabalho busca identificar possíveis interações medicamentosas que são relevantes para a terapêutica medicamentosa adotada na prática odontológica. As interações decorrentes de fatores individuais (patologias, idade, obesidade, etc) e da associação de dois ou mais medicamentos (medicamento-medicamento) serão revisadas e discutidas em uma série de três artigos.

Aims: To evaluate the antimicrobial activity of 10% and 37% carbamide peroxide during dental bleaching in three different modes. Materials and Methods: This five-week double-blind randomized controlled trial included 32 volunteers assigned to four groups (n= 8). Each group received bleaching agents or placebo as an in-office and at-home treatment.

The aim of this study was to evaluate the effect of sodium diclofenac on the bioavailability of amoxicillin. In this randomised, crossover study with a 1-week washout period, 20 volunteers received a 2 g oral dose of amoxicillin (Amoxil ®) (Group 1) or a 2 g oral dose of amoxicillin with 100 mg of sodium diclofenac (Voltaren ®) (Group 2). Blood samples were collected at 0, 0.25, 0.5, 1, 1.5, 2, 2.5, 4, 6, 8, 12 and 24 h following drug administration. High-performance liquid chromatography with ultraviolet detection was used to quantify plasma amoxicillin concentrations. Bioassay (Micrococcus luteus ATCC 9341) was performed to verify the antimicrobial efficacy of amoxicillin in vitro. The pharmacokinetic parameters area under the plasma concentration-time curve (AUC), maximum plasma concentration observed during the 24-h study period (C max) and renal clearance (CL) were analysed by analysis of variance, and time at which C max occurred (T max) and volume of distribution (VD) were analysed by Wilcoxon test (P < 0.05). For Group 1, the mean (± standard deviation) AUC 0-24 , C max and T max values were 3391.8 ± 1186.7 g min/mL, 17.3 ± 6.5 g/mL and 121.5 ± 20.6 min, respectively; and for Group 2, the values were 2918.4 ± 1024.8 g min/mL, 15.5 ± 5.8 g/mL and 136.5 ± 30.0 min, respectively. Lower values of AUC and C max were observed for Group 2 (P < 0.05). CL of amoxicillin increased (P < 0.05) by 18.5% in Group 2, suggesting that sodium diclofenac may interfere with amoxicillin renal excretion. In conclusion, sodium diclofenac can significantly reduce the bioavailability of amoxicillin.
