Temperature and water availability appear to be important factors for temperate fruit trees in mild climates. The aim of this research was to study the involvement of temperature regime on biological processes, such as xylem differentiation and dormancy evolution, in apricot flower buds (Prunus armeniaca L.). Over 3 consecutive years, biological and anatomical investigations were carried out in Tuscany (Italy) and Murcia (Spain) on two cultivars characterized by a different chilling requirement (CR): Currot, a traditional Spanish cultivar with a very low CR, and Stark Early Orange (SEO), a North American cultivar with a very high CR. Currot had a regular bearing, and was characterized by a synchronism between endodormancy release and xylem differentiation. On the other hand, SEO showed an irregular flower bud growth leading to flowering and fruit-set failure, even with a high level of chilling accumulation. No relationship was found in SEO between xylem development and flower bud growth reactivation. The de-synchronism between these processes could determine the poor adaptability to different environmental areas of SEO cultivar, and its very high CR is not the main hypothesised cause of the altered flower bud development.

Effects of vitamin D repletion in young people with low vitamin D status have not been investigat... more Effects of vitamin D repletion in young people with low vitamin D status have not been investigated so far. We evaluated the effect of a single massive dose of cholecalciferol on calcium metabolism at 3, 15, and 30 d, compared to baseline. We conducted a prospective intervention study in an ambulatory care setting. Forty-eight young subjects with vitamin D deficiency participated in the study. A single oral dose of 600,000 IU of cholecalciferol was administered to each subject. We evaluated serum changes of 25-hydroxyvitamin D [25(OH)D], 1,25-dihydroxyvitamin D, calcium, and PTH induced by a single load of cholecalciferol. The 25(OH)D level was 15.8 ± 6.5 ng/ml at baseline and became 77.2 ± 30.5 ng/ml at 3 d (P < 0.001) and 62.4 ± 26.1 ng/ml at 30 d (P < 0.001). PTH levels concomitantly decreased from 53.0 ± 20.1 to 38.6 ± 17.2 pg/ml at 3 d and to 43.4 ± 14.0 pg/ml at 30 d (P < 0.001 for both). The trends were maintained in a subgroup followed up to 90 d (P < 0.001). Mean serum Ca and P significantly increased compared to baseline, whereas serum Mg decreased at 3 d. 1,25-Dihydroxyvitamin D significantly increased from 46.8 ± 18.9 to 97.8 ± 38.3 pg/ml at 3 d (P < 0.001) and to 59.5 ± 27.3 pg/ml at 60 d (P < 0.05). A single oral dose of 600,000 IU of cholecalciferol rapidly enhances 25(OH)D and reduces PTH in young people with vitamin D deficiency.
Subclinical hypercortisolism (SH) has been associated with increased prevalence of hypertension, ... more Subclinical hypercortisolism (SH) has been associated with increased prevalence of hypertension, type 2 diabetes mellitus, dyslipidaemia, central obesity, osteoporosis and vertebral fractures. We aimed to investigate the accuracy of different SH diagnostic criteria in predicting the presence of complications. This was a retrospective study. We evaluated data from 231 patients (120 women and 111 men) affected with adrenal incidentalomas (AI). We studied the accuracy of different SH diagnostic criteria (cortisol after 1 mg overnight dexamethasone suppression test - 1mg-DST - at different cut-off such as 49.7, 82.8, 137.9 nmol/l, elevated urinary free cortisol, reduced adrenal corticotroph hormone (ACTH) levels alone or various combination of these parameters) in predicting the concomitant presence of the following three complications: hypertension, type 2 diabetes and vertebral fractures. The criterion characterized by the presence of two of 1mg-DST >82.8 nmol/l, elevated UFC and reduced ACTH struck the best balance between sensitivity and specificity, reaching a good accuracy in predicting the cluster of complications (61.9%; 77.1% and 75.8%, respectively). The presence of this cluster was associated with this criterion (OR 4.75, 95%CI 1.8-12.7, P = 0.002) regardless of gonadal status, body mass index (BMI) and age. The SH criterion characterized by the presence of two of 1mg-DST >82.8 nmol/l, elevated UFC and reduced ACTH seems the best in predicting the presence of chronic manifestations of subtle cortisol excess.
