The applicability of a novel procedure for the disinfection of microbiologically polluted waters from fish-farming ponds, based on the combined action of visible light (including sunlight) and porphyrin-type photosensitising agents, has been investigated using (a) cell cultures of a Grampositive bacterium (meticillin-resistant Staphylococcus aureus), a Gram-negative bacterium (Escherichia coli) and a fungal pathogen (Saprolegnia spp.); (b) pilot aquaculture plants involving either spontaneously or artificially Saprolegnia-infected rainbow trout (Oncorhynchus mykiss). The results obtained by using two cationic porphyrins, namely a tetra-substituted N-methyl-pyridylporphine (C1) and its analogue where one N-methyl group had been replaced by a N-tetradecyl chain (C14), and low intensity visible light irradiation showed an extensive (up to 6-7 log) decrease in the bacterial/fungal population after short incubation and irradiation times in the presence of micromolar photosensitiser concentrations. Moreover, C14 showed some toxic effect also in the absence of light. Extension of these studies to the pilot plants indicated that both C1 þ light and C14 can prevent Saprolegnia infections or promote the cure of saprolegniasis in infected trout by treatments with submicromolar porphyrin doses. The procedure appears to be of low cost and to have a low environmental impact.

Lake Garda, the largest Italian lake, is suffering from the introduction of several non-indigenous species during the last decades and can now be considered one of the main European freshwater hotspots of xenodiversity. Among the Bivalvia (Veneroidea, Cyrenidae), Corbicula fluminea and Corbicula fluminalis were first recorded in 2002 and 2008 respectively, and are now widespread in the southern part of the basin. Recent observation of specimens that did not resemble either of these taxa, suggested that the populations of invasive Corbicula of Lake Garda could include some other taxa not previously recorded. With this aim, a thoroughly characterization of Corbicula shells found at Lake Garda was made. In this study, the production of polyhydroxyalkanoated PHA-rich microbial biomass as a novel feed additive in aquaculture was investigated at a lab-scale. Bio-based volatile fatty acids (VFAs), obtained from the acidogenic fermentation of agricultural residues in existing anaerobic digestion plants, were used as carbon and energy to cultivate the PHA-rich microbial biomass. The experimental activities were carried out using Thauera sp. Sel9 as pure strain, which was grown in a continuous stirred-tank reactor (CSTR) operated at three different hydraulic retention times (HRT). The highest productivity obtained of biomass cells was 0.69 g/L day, operating at one day HRT while the observed PHAs production yield was 0.14 gPHA/g soluble COD removed. Sustainability of aquaculture is tied to the origin of feed ingredients. In search of sustainable fish meal-free formulations for rainbow trout, we evaluated the effect of Hermetia illucens meal (H) and poultry by-product meal (P), singly (10, 30, and 60% of either H or P) or in combination (10% H + 50% P, H10P50), as partial replacement of vegetable protein (VM) on gut microbiota (GM), inflammatory, and immune biomarkers. Fish fed the mixture H10P50 had the best growth performance. H, P, and especially the combination H10P50 partially restored α-diversity that was negatively affected by VM. Diets did not differ in the Firmicutes:Proteobacteria ratio, although the relative abundance of Gammaproteobacteria was reduced in H and was higher in P and in the fishmeal control. Italy is the first producer of rainbow trout in Europe (EU member) and Trentino farmers represent roughly 8% of this production, but genetic information of the stocks employed is still lacking. This study aimed to estimate the genetic variability of the rainbow trout stocks farmed in Trentino and to reconstruct the related admixture history. Sixty hundred and thirty -two animals belonging to 23 different stocks were analysed using microsatellite loci. Seventeen stocks showed negative values of inbreeding coefficient within population (F is ranging from -0.06 to -0.107). NJ tree of genetic distances among stocks showed distinct clusters reflecting, at least partially, the information coming from known history. This study compared the nutrient-energy retention, digestive function, growth performance, and welfare of rainbow trout (ibw 54 g) fed isoproteic (42%), isolipidic (24%), fishmeal-free diets (CV) over 13 weeks. The diets consisted of plant-protein replacement with graded levels (10, 30, 60%) of protein from poultry by-product (PBM) and black soldier fly H. illucens pupae (BSFM) meals, either singly or in combination. A fishmeal-based diet was also tested (CF). Nitrogen retention improved with moderate or high levels of dietary PBM and BSFM relative to CV (p < 0.05). Gut brush border enzyme activity was poorly affected by the diets. Gastric chitinase was up-regulated after high BSFM feeding (p < 0.05). The gut peptide and amino acid transport genes were differently regulated by protein source and level. Carpione (Salmo carpio, Linnaeus 1758) is an endangered precious endemism of Lake Garda (Northern Italy), the largest Italian lake. To date, several bottlenecks about its culture remain unsolved, including the identification of a proper growth-out diet. The aim of the present study was to test four different grossly isolipidic, isoproteic, and isoenergetic diets in which the main ingredients had a different origin. Specifically, a diet currently used by local farmers for carpione culture, largely based on marine ingredients, was used as control (CTRL), while the other three diets were formulated by partially replacing marine ingredients with plant ones (VEG) or with different percentages of processed animal proteins (PAP1 and PAP2). This trial aimed to assess the growth performance of trout (Oncorhynchus mykiss) fed novel formulations, evaluate fish welfare status, and determine flesh quality as part of the evaluation of sustainable feeds. A control diet containing fish meal and soy products (CTRL) was compared to: a diet with processed animal proteins (PAP); a diet without PAP (NoPAP); a PAP diet lower in protein (PAP−); and a NoPAP diet higher in protein (NoPAP+). Groups of 50 fish, weighing 58.84 ± 1.39 g (IBW), were allocated to 20 tanks and fed with formulated diets ad libitum over 91 days. Better growth performance was observed after the experiment in fish fed the NoPAP+ diet when compared to other diets. Protein retention was higher in CTRL diets than in PAP and PAP− diets. Protein and phosphorous digestibility were lower in fish fed PAP− diet. Diets did not influence the texture analysis. The Precision Fish Farming (PFF) approach was applied to the estimation of fish oxygen consumption of rainbow trout in a raceway farm. A dynamic model, simulating the evolution of Dissolved Oxygen concentration, was identified: the daily oscillation of fish oxygen consumption rate was simulated by means of a sinusoidal function. The model was applied to the data set collected during a four-week field study, which was carried out in July 2019. Water temperature and Dissolved Oxygen concentration were measured with an hourly frequency in farm influent and effluent. Fish biomass was monitored on a daily basis by combining the data provided by a state-ofthe art system for non-invasive estimation of fish weight distribution with mortality counting. The monitoring period was partitioned into two time-windows, as fish was not fed during the first two weeks. These windows were further partitioned into a calibration and validation set. Three model parameters, i.e. the average daily respiration rate, the amplitude of its daily oscillation, and its phase were estimated by fitting the model output to the time series of DO concentration in the effluent. The results of the calibration show that: 1) the daily average oxygen consumption rate is consistent with the literature; 2) the amplitude of the daily oscillation when fish is regularly fed is more than twice that estimated for fasting fish. The results of the validation suggest that the model could be used to implement a cost-effective automatic control of oxygen supply, based on the short-term prediction of oxygen demand.
