Monoclonal Antibodies for the virus:
Zmapp
o Combination of 3 proteins
mAb114
o Derived from the Ebola survivor
Review
Filoviridae: causes
hemorrhagic fever
Zoonotic virus
Transmitted by direct
contact to bodily fluid
Death by bleeding
Coat
Cell attachment & Invasion
RNA polymerase
Makes copies of viral RNA
with VP35
• EBOV is make use of the process
called macropinocytosis.
• EBOV GP reacts with the protein
receptors (TIM proteins) of the
immune cells that detects debris.
Macropinocytosis
Immune cells
• EBOV is engulphed.
• EBOV GP is digested into GP2.
Lysosome
• Upon reaching cytoplasm,
EBOV GP2 interacts with the
Niemann-Pick C1 (NPC1) NPC1 proteins
proteins.
EBOV is able to detect its
location through NPC1
proteins.
• EBOV viral membrane fuses with the
endosomal membrane upon
reaching cytoplasm.
• Releases its viral genetical material.
• Replication of the genetic material.
• The genetic material will undergo
transcription and translation
process.
Viral budding (cell exit)
The virus exits the infected cell from:
Intracellular membrane
Multivesicular bodies
Plasma membrane
Target cells of EBOV
First targets for infection: Followed by:
Monocytes/Macrophage Fibroblasts
Dendritic cells Hepatocytes
Adrenal cells
Epithelial cells
EBOV induced pathogenic
events
Why so difficult for the cure?
• Too small (virus).
• Less target since it only makes few proteins.
• They evolve quickly.
• Making or studying EBOV require biosafety level 4
laboratories.
• Relatively few people are infected and fewer
survivors.
