CENTRIFUGE

Discovered by ANTONIN
PRANDTL , in 1864.

INTRODUCTION :
CENTRIFUGE is a mechanical
process that utilizes a spining
medium to separate one or more
components of a sample according
to density or size.
CENTRIFUGEL separations may be
classifled as either analytical or
preparactive.
E.g: molecular weight, shape, and
association.

TYPES OF CENTRIFUGE:

Benchtop centrifuge Continuous

flow centrifuge Gas centrifuge
Hematocrit centrifuge
High-speed centrifuge Low-
speed centrifuge
Microcentrifuge
Refrigerated centrifuge
Ultracentrifuge
CENTRIFUGE DEFINATION:

The centrifuge is commonly used

in laboratories for the separation of
biological molecules from a crude
extract.

In a centrifuge ,the sample is kept

in a rotor that is rotated about a flxed
point (axis) ,resulting in strong force
perpendicular to the axis.
There are different types of
centrifuge used for the separation of
different molecules ,but they all work
on the principle of sedimentation.

BENCHTOP CENTRIFUGE:

Also known as multipurpose

centrifuge .

USES:

pellet cells and bacteria.

flxed angle
rotor:17,000g/14,000rpm.
swinging bucket
rotor:3,800g/4,800rpm.

CONTINUOUS FLOW CENTRIFUGE:

Used industrially and for few lab

separations like recovery of bacteria
from litres of culture solution.

The sample is ejected through the

centre and separated at a
speciflc speed.

GAS CENTRIFUGE:

Uses physical principle of centri

petal force to separate U-235 from
U-238.

very high speed rotor generates

centripetal force.

separation increases with rotor

speed and length.
HEMATOCRIT CENTRIFUGE:

Micro : Hematocrit centrifuge

dsc-100MH-1.

Speed : 12,000rpm(flxed).

Motor : universal motor.

Timer : 0-15min . Mechanical

timer.

Weight (kg) : 4.5/ 5.0kgs.

HIGH-SPEED CENTRIFUGE :

15,000-20,000Rpm.

Lorge sample capacity depending

on rotor.

normally refrigerated.

research applications.

differentiation separation of
nucleus , mitochondrial , Protein
precipitate , large intact organclle
, cellular debris , bulky protein
aggregates.

LOW -SPPED CENTRIFUGE:

The low speed centrifuge has a

maximum speed of 4000-
5000rpm.

These instruments usually operate

at room temperature with no means
of temperature control.

Two types of rotor are used in it

flxed angle and swinging bucket.
MICRO CENTRIFUGE:

Microcentrifuges are used to

process small valumes of biological
molecules cells, or nuclei.

Microcentrifuges are small enough

to flt on a table -top and have rotors
that can quickly change speeds.

Maximum angular speeds of

12,000-13,000rpm.
REFRIGERATED CENTRIFUGE:

Refrigerated rotor champer.

capacity to change rotor

chambers for varying size

Spin speed upto 6500g and

sediment rapidly like erythrocytes
, yeast cell , nuclei and
chloroplast.

ULTRACENTRIFUGE:
It is an important tool in
biochemical research.

It is high speed vacuum- type

centrifuge.

It discovered by swedberg in 1920.

speed of more than 60,000rpm.

Temperature is not raised..
