(Acetyl Salicylic Acid)

GROUP B | PHARMACEUTICAL CHEMISTRY LAB

GROUP B | PHARMACEUTICAL CHEMISTRY LAB
EXPERIMENT NO. 6
To perform the assay of aspirin tablet by back titration method BP.

REQUIREMENTS
Apparatus: Analytical balance, Beaker, Spatula, Burette, Conical flask, Retort stand, Measuring
cylinder, Volumetric flask, Pipette.
Reagents: HCL , phenol red , methyl orange , disilled water ,sodium hydroxide, sodium carbonate

THEORY
Aspirin:
Aspirin is also known as acetylsalicylic acid.
IUPAC name: 2-acetoxy benzoic acid
It is a white crystalline solid at room temperature.

 Molecular Formula: C9H8O4

 Molar Mass: 241.158 g/mol
 Uses:
1. Aspirin is known as a salicylate and a NSAID.
2. Aspirin is used to reduce fever and relieve mild to moderate pain from conditions such as
muscle aches, toothaches, common cold and headaches.
3. It may be also used to reduce pain and swelling in conditions such as arthritis.
4. It is anti-inflammatory, so it is also used to treat inflammation.

PROCEDURE
1. Rinse all the glassware.
2. Prepare 0.5M HCL in 100ml and 0.5M NA2CO3 in 50ml.
3. Fill the burette with HCL solution up to the mark.
4. Take 10 ml of SODIUM CARBONATE solution in a conical flask. Add 2 drops of
methyl orange / methyl red as an indicator.
5. Standardize it against HCL solution until the color changes
6. Note the reading of burette. Repeat the process 3 - times, take concordant reading and
determine the correction volume
7. Take 20 tablets and weigh them correctly.
8. Transfer the weighted tablets to the mortar and pestle and crushed them into fine particles.
9. Add 30ml of 0.5 molar NaOH in the same flask and label the flask as flask A. Heat the
flask A for 10 mins until the mixture boils and cool up to the room temperature. In the
meanwhile, fill the burette with 0.5M HCl.

10. Take another flask add 0.5M NaOH 30ml in it. Add 2 drops of phenol red and mark the
flask B. Titrate it with HCl filled in burette.
11. When the flask A is cooled to room temperature, refill the burette with 0.5M HCl and
titrate the flask A after adding 2 drops of phenol red as an indicator.

WEIGHING THE ACTIVE INGREDIENT:

1. Amount of aspirin in 1 tablet = 0.3gm
2. Weight of 20 tablet = 5.966gm
3. Amount of aspirin in 20 tablet = 20 x 0.3 = 6gm
4. Therefore, we have 6gm of aspirin in gm of tablets
5. For 1gm aspirin =
6. For 0.5gm =
7. Thus, 0.5 gm of aspirin will be in gm of tablets

CALCULATION
1. Correction volume:

C.F: Volume of primary standard (Na2CO3) / volume of secondary standard (HCl)

2. To determine the burette reading :

B.R = [ Flask A(B.R) x C.V] – [ Flask B ( B.R) x C.V]

3. Titer factor:

4. Observed amount:
Observed amount: Titer factor x burette reading (assay)

5. Purity:
% Purity = Observed amount/ Standard amount x 100
