Tittle: Experiment 2: Staining Onion Epidermal Tissue

Objectives
i. To prepare wet mounted slide.
ii. To draw and identify the onion epidermal cell.

Introduction

The characteristic of living plant cell have cell wall, large central
vacuole and chloroplast. Next, the type of tissue for onion
epidermal cell is parenchyma. However, the function of onion

photosynthesis. The layers of onion contain simple sugars
(carbohydrates) some of which are stored as starch (starch
granules).

1. One drop of diluted iodine was put on the slide.


2. The inner epidermal layer from the scale of an onion was
stripped off and cut it into smaller pieces.
3. The onion epidermal layer was place on the slide. Make
sure the tissue was not folded.
4. One drop of diluted iodine solution was applied on the
onion epidermal tissue.
5. A cover slip was used to lower the cover slip gently.
6. The excessive iodine was remove using a filter paper.
7. The tissue was observed using 10x or 40x objective lens.
8. Observation at high magnification was drew.

Observation
Discussion

The function of iodine is to stain the starch. Besides, the shape of onion epidermal
cell is rectangular shape due to present of cell wall and the position of its nucleus
at peripheral of cell due to large central vacuole. Next, the arrangement of onion
epidermal cell is rectangular in shape, compactly arranged and without any
intercellular spaces. Furthermore, organelle is not visible in the cell under the light
microscope is chloroplast. It is because onion cell gets no light to carry out
photosynthesis and largely present in the leafy part on the onion, which is exposed
to the sun.

Comparison the onion epidermal cell with two other plant cells, mesophyll cell and
xylem based on the structures and organelles present:
