Exercise of Solvent Extraction:

1. A 25.0 ml aliquot of a 0.4% (w/v) aqueous solution of a natural compound was


extracted with three 10.0 ml portions of ether. The combined ether extracts were
evaporated to dryness the residue was weighed. The ether/water partition
coefficient for the compound is 3.0. What was the weight of the residue?
2. The partition coefficient of an alkaloid between chloroform and water is 8 (it is
more soluble in the organic solvent). Fifty milliliters of a 0.01 M aqueous solution
of the alkaloid is extracted with one 10.0 ml portion of chloroform, and the
chloroform solution is evaporated to dryness. What is the weight of the residue?
The MW of the alkaloid is 210.
3. How many extractions are necessary to remove 99.9% of an enzyme from 50 ml
of an aqueous solution if it is extracted with 25 ml portions of PEG 1150 as
solvent and its partition coefficient is 1.0?
4. 100.0 ml of a 1% penicillin solution was extracted with 2 x 20 ml portions of
amyl acetate. The combined ether extracts were evaporated and the dry residue
was found to weight 865.5 mg. Calculate the partition coefficient.
5. A mixture of Ephedrine (pKa=9.5), Salicylic acid (pKa=3) and Phenobarbital
(pKa=8) was subjected to the following extraction shown in the picture. Tell in
which fraction (I, II, III, IV) each of the drugs will be predominantly found.
