Anti cancer alkaloids

Pyrimidine is a heterocyclic aromatic organic compound containing two nitrogen atoms at positions 1 and 3 of the six-membered ring which shows wide range of biological activities. Numerous methods for the synthesis of pyrimidine and also their diverse reactions offer enormous scope in the field of medicinal chemistry. Pyrimidine posses wide spectrum of biological activities like including antitubercular, antibacterial, antifungal, antiviral, antiinflammatory, Antimalarial, anticancer, anti-HIV activity. The present review attempts to give a brief information about the synthesis and various biological activities of pyimidines and their derivatives.

The aim of the work was to develop methods to create fodder (sweet) varieties of Lupinus polyphyllus Lindi, with stable, low levels of alkaloids. New approaches were developed with the specific intention of avoiding the rapid restoration of alkaloid synthesis in this crosspollinated lupin species. We created sweet forage varieties of multifoliate lupin with sufficiently stable and low alkaloid content. Positive results were demonstrated with the first sweet commercial cultivar Pervenec, which was listed in the State Catalogue of Breeding Achievements in Russia. The major tasks carried out in Finland since 1996 were the breeding of cultivars with stable low alkaloid content, different types of pollination, non-dehiscent pods, winter hardiness and frost tolerance, capable of producing high yields of green matter (40 -50 Mg ha-1 ) and maintaining the shading of cover crops. One of our purposes of Washington lupin breeding in Finland is to create productive sweet forms with white flowers and seeds analogous to white flowered and white seeded varieties of narrow-leafed lupin (L angustifolius L.) in Australia.

Some of them are anticancer drugs with well-defined clinical uses, exerting their action on microtubules dynamics or DNA replication and topology. On the other hand, mitochondria have been recognized as an essential organelle in the establishment of tumor characteristics, especially the resistance to cell death, high proliferative capacity and adaptation to unfavorable cellular environment. Interestingly, many alkaloids exert their anticancer activities affecting selectively some functions of the tumor mitochondria by 1) modulating OXPHOS and ADP/ATP transport, 2) increasing ROS levels and mitochondrial potential dissipation by cross-talk between endoplasmic reticulum (ER) and mitochondria, 3) inducing mitochondria-dependent apoptosis and autophagy, 4) inhibiting mitochondrial metabolic pathways and 5) by alteration of the morphology and biogenesis of this organelle. These antecedents show the relevance of developing research about the effects of alkaloids on functions controlled by tumor mitochondria, offering an attractive target for the design of new alkaloid derivatives, considering organelle-specific delivery strategies. This review describes mitochondria as a central component in the anticancer action of a set of alkaloids, in a way to illustrate the importance of this organelle in medicinal chemistry.

Adhatoda vasica (L.) Nees (Acanthaceae) is a well known plant drug in Ayurvedic medicine, used for the treatment of different types of diseases and disorders. The tissue cultures of this plant species were established on Murashige and Skoog culture medium by using various concentrations of growth hormones (0.5-5.0 mg/L α-napththaleneacetic acid + 0.5-3.5 mg/L 6-benzylaminopurine). The concentration of vasicine acetate was found higher (0.51  0.519%) in six week old callus than in vivo (0.47  0.556%) plant material (roots). The antioxidant activity was evaluated by using DPPH-radical scavenging, reducing power and superoxide anion scavenging models. We found that 2-acetyl benzyl amine showed maximum antioxidant activity against various antioxidant models, including DPPH-radical scavenging (40.59 ± 0.774%), reducing power (461.55 ± 0.402%) and superoxide anion scavenging (52.54 ± 0.553%) than other isolated alkaloids at 10-30 μM concentrations. Our observed results suggest that the pyrroloquinazoline alkaloids of A. vasica demonstrated strong antioxidant activity, which could be used as potent drugs against different types of oxidative stresses.

The N.I. Vavilov Institute (St. Petersburg, Russia) holds more then 2500 accessions of lupin. Many years of observing the plants sown in various regions and biochemical analysis of seed and plant matter of the accessions using the specialized methodology of the Institute developed for lupin, have found new regularities in a variability of chemical structure of accessions. These range from genetic and ecological features to agronomic growing conditions. Among the rich specific diversity of lupins there are species, varieties and forms which accumulate large amounts of protein (up to 55% ), oil (up to 15% in Russia, and in the conditions of Uzbekistan, up to 20.8 % ) and other useful substances. Lupin is particularly valuable for the absence of anti-nutritive factors (inhibitors of proteinase and hemagglutinins) in its protein. Lupin seed (its low-alkaloid sweet forms), unlike other leguminous crops, is suitable for raw use without heat treatment. However, lupins are only grown on 1.5 million hectares, compared with soya on more than 70 million hectares.

Synthetic cannabinoid terpenes as potential anti-cancer substance.
                  Antitumor, anticancer cannabinoids terpenes, alkaloids, anthocyanins.

Many natural terpenes exhibit anti-cancer activity, among them can be considered as cyclic alcohols and ketones and active cannabinoids, directly or indirectly act as anti-cancer agents. Cannabinoids help reduce inflammation, regulate the immune system... Also cannabinoids and many terpenes anticancer is often cyclic or linear alcohols and ketones with a characteristic double groups like cannabinoids and terpenes. This allows to combine advantageously required for antitumor activity and for Pro-cannabinoid activity, the substituents in the same structure.
Thus it is possible to develop design terpene cannabinoids, bringing them closer to essential oils with anticancer activity. Agonists anandamide suggest the presence of the ketone group, double bonds, also active terpenes suggest the presence of the ketone group and double bonds, only need to combine the radicals so that their functions are duplicated. Examples include synthetic terpenes with the cannabinoid structure harmoniously introduced anticancerogenic radicals with some active as anti-cancer essential oils.
The work is aimed at promoting the synthetic physiologically active terpenes, such substances can be highly active and have a relatively diversified and complex antitumor activity.
Such structures can redirect the immune response, reduce inflammation, start the activity cell apoptosis and some others, if the experiment will yield positive results.
The work of theoretical, no experiments with these substances and result not received.
Because these substances are highly lipophilic structure it is possible that they will have a positive effect in brain tumors, and other organs and tissues composed of fats, or for the penetration in which requires the overcoming of the lipid barrier.
Also essential oils are active in small quantities and these cannabinoids can be very active, and in the end the result may be a balance in range of properties and synergy when used as a complementary agent in chemotherapy or as a prophylactic.
Also, among the cannabinoids presents a water-soluble substance and a Quaternary ammonium salt, surface active substances, which may be useful for tumors of the internal organs, tissues, or tumors of the skin... Among the cannabinoids are presented as Pro-CB1 and CB2.. substance.
Is also possible that the Quaternary ammonium bases do not pass the blood-brain barrier, it may minimize Central effects of cannabinoids. Structure approximate and not final, correction of the radicals can be achieved after synthesis and testing of substances.
Quaternary ammonium salts may also have independent chemotherapeutic activity and the activity induced Pro-terpene and phenolic radicals, immune response...
Cannabinoids like anthocyanins may have to be somewhat selective towards cancer of the stomach.
Also cannabinoids that do not pass the blood-brain barrier can be active against tumors of the brain, via activation of peripheral cannabinoid receptor system and the impact of active catabolites.
Examples of substance:
Not all substances have a clear route of synthesis and catabolism, not all substances are chemically stable, but are listed as an illustration of the design strategy of drugs.

  Substance cannabinoids having radicals contribute to high activity, such as alcoholic hydroxyl group, ketone group, five-membered unsaturated cycles, unsaturated and saturated carbon chains of the respective endogenous cannabinoids, as well as substance with no difficult chemical synthesis may take precedence over complex and less active Pro-terpene cannabinoids in the form of low cost and the ultimate cost of the drug. Thus, in the work by the substances shown on the model structures and nuclei of active substances, classes of substances, the targets of exposure, methods of administration, and ultimate affordability of the substance.

Drugs that not passing gematoentsefalichesky barrier, whether structurally opioid analgesics (methylnaltrexone bromide), cannabinoids.. are classified according to the type of peripheral actions. Accordingly, the peripheral actions of cannabinoids should be allocated to the group non-narcotic anti-tumor and anti-cancer drugs. Such drugs dispensed by prescription, but not belonging to list A or B drugs.

Fire ants are aggressive invasive insects spread around the world via ship cargo, mainly originating from the United States. They can cause severe impacts on human activities and the environment. This chapter presents an overview of what is known about fire ant venom, ending with different open possibilities for investigation in this topic. For decades, studies on fire ant venoms have been limited in scope because of the technical difficulties in obtaining enough samples for analysis and bioassays; yet now there is one simple, effective, published method for extracting venom from whole colonies. Fire ant venom is mainly composed of a mixture of>95%bioactive piperidine alkaloids and 0.01% of proteins, which comprise mainly allergens, phospholipases, and neurotoxins. The alkaloids of fire ants, generally named solenopsins, are well known for their antifungal and insecticidal properties; however, many have also been suggested as promising alternatives for other biomedical applications, such as the treatment of parasitemia and various neurological diseases. The venom proteins of fire ants remain only superficially studied, as most published literature focuses on just four allergens. Crucially, others may contain compounds of interest to immunotherapy or even play a central role in aspects of social organization.

Moringa oleifera (M. oleifera) has been considered as medicinal plant and food substance since ancient times. Nowadays various therapeutic effects of M. oleifera such as antimicrobial, anticancer, anti-inflammatory, antidiabetic, and antioxidant effects have been successfully investigated. In this study, the main objective was to optimize M. oleifera leaf-methanol extraction using Design of Experiment (DoE) software with response surface methodology in terms of temperature and incubation time by sonication. The other objective was to investigate anti-cancer activity (breast cancer cell: MCF-7) by M. oleifera methanol extraction incorporation with microtitrate tetrazolium (MTT) assay. In this research, the optimum condition for leaf extraction was found to be temperature 50 ⁰ C and incubation time 45 min with medium frequency at sonication and it obtained 88.39% cancer cell growth inhibition by this condition. Extracted leaves inhibited MCF-7 cell line with 87.13% in average at wavelength A570nm.

Quinine (cinchonan-9-ol, 6'-methoxy-, (8. alpha. 9R)-) is an antiprotozoal and an antimyotonic, and is known for the treatment of malaria caused by Plasmodium species, the treatment and prophylaxis of nocturnal recumbency leg muscle cramps, and the treatment of babesiosis caused by Babesia microti. Quinine is structurally similar to quinidine, which is also an antiprotozoal, but can function as an antiarrhythmic. The said Cinchona alkaloid isolated from the bark of several species of Cinchona trees, the medicinally active bark, which is stripped from the tree, dried and powdered, contains a variety of alkaloids having their antimalarial property. In 1820, quinine (( ?)-(6-methoxyquinolin-4-yl) ((2S, 4S, 8R) - 8-vinylquinuclidin-2-yl) methanol) was identified as an active compound. Since then, cinchona alkaloids especially, quinine have played a pivotal medicinal role. Approximately 700 metric tons of cinchona alkaloids are now extracted from the bark of Cinchona annually. Nearly half of this is used in the food and beverages industry as a bitter additive and much of the remaining quinine and quinidine is used as an important antimalarial drug and muscle relaxant compound and as a cardiac depressant (antiarrhythmic).
