verklaringskloof en subjectiviteit natorp, husserl en lacan over reflectie door Jasper Feyaerts en Stijn Vanheule (Gent) In recente jaren zijn we getuige geweest van een terugkeer van het vraagstuk van subjectiviteit in diverse onderzoeksdomeinen, gaande van de neurocognitieve wetenschappen, de analytische 'philosophy of mind', de studie van psychopathologie tot de zogenaamde 'consciousness studies'. Een van de belangrijkste redenen voor een dergelijke terugkeer ontspringt aan wat we freudiaans zouden kunnen omschrijven als een "Unbehagen in die kognitive Neurowissenschaft": een groeiend onbehagen met wat algemeen beschouwd wordt als het dominante paradigma in het wetenschappelijk onderzoek naar de menselijke geest sinds de jaren 60, de cognitieve (neuro)wetenschap. Hoewel zij initieel in staat werd geacht om Humes befaamde ambitie te verwezenlijken de "Newton of mental phenomena" te worden, stellen onderzoekers vandaag dat er nog steeds fundamentele vragen onbeantwoord zijn gebleven. Background and Aim: African swine fever (ASF) is a highly virulent and contagious viral disease caused by the ASF virus (ASFV). It has a significant impact on swine production throughout the world, while existing vaccines and specific treatments remain ineffective. ASFV p30 is a potent antigenic protein that induces protective antibodies immediately after infection; however, most recombinant p30 is insoluble. This study aimed to improve the solubility, yield, and purity of recombinant p30 by tagging it with a small ubiquitin-like modifier (SUMO) and modifying the protein purification process. Materials and Methods: SUMO fused with ASFV p30 (SUMO-p30) and p30 alone were cloned and expressed in Escherichia coli. SUMO-p30 and p30 solubility and expression levels were analyzed by sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE). Protein purification was modified by combining ammonium sulfate precipitation method with affinity chromatography. In addition, large-scale production of all versions of p30 were compared using SDS-PAGE and western blotting, and the purified p30 was used to develop the indirect enzyme-linked immunosorbent assay (ELISA). Results: The solubility and expression levels of SUMO-p30 were dramatically enhanced compared with that of p30. Modification of the purification process significantly increased purified and soluble SUMO-p30 and p30 yields by 6.59 and 1.02 μg/mL, respectively. Large-scale production confirmed that this procedure increased the quantity of recombinant p30 while maintaining protein purity and immunogenicity. The p30-based indirect ELISA was able to discriminate between positive and negative serum samples with statistically significant differences in mean optical density 450 values (p < 0.001). Conclusion: This study demonstrates the enhancement of solubility, purity, and yield of ASFV p30 expressed in E.coli by SUMO fusion tagging and combining ammonium sulfate precipitation with affinity chromatography for protein purification. These positive effects were sustained in large-scale production. Cleavage and removal of hexahistidine-SUMO tag from the fusion protein by protease may not be suitable when handling a large amount of the protein. However, the SUMO-fused p30 retained strong immunoreactivity to convalescent swine serum, indicating its application in immunization and diagnostic purposes. The expression and purification procedures in this study could be applied to increase solubility, quality, and quantity of other recombinant proteins as well. Habermas’s only two writings on the theme of civil disobedience date to the 1980s. The background to these pieces were the massive pacifist protests that took place in the fall of 1983 against Germany’s installation of cruise missiles (under the protection of NATO). The protestors, who violated multiple administrative norms, sought to justify their actions as acts of civil disobedience. In 1983, in one of his most combative essays, Habermas addressed the question of civil disobedience as a nonconventional, though legitimate, tool for the establishment of the collective political will. In Habermas’s valorization, this pacific form of dissidence represents an indispensable element for a democratic culture: an ideal instrument to realize the ends of a democratic state and secure the efficacy of constitutional principles and norms, that is, a useful means to attain the maturation of public opinion.

In our study, we attempted to identify systematically the use of Internet applications in the German population in order to derive risk factors for problematic use. In a representative survey of the German population, we queried 1,401 women and 1,111 men between the ages of 14 and 94 years by specific questions and standardized questionnaires on depression, anxiety (HADS), and depersonalization (CDS-2). The majority of the German population (55%) used the Internet in their leisure time. Users were younger and had a higher socioeconomic status (education, employment, income). Leisure-time use included e-mail and information search, as well as shopping. Chatting, online communities, games and sex were domains of young, mostly male adults. Overall, 9.3% reported at least one negative consequence of Internet use, especially neglect of recreational activities and problems with family/partner, work or education, and health. Problematic use was associated with longer average daily online times, avoidance of negative emotions, preference for certain applications (gaming, gam-bling, online sex) and an increased rate of depersonalization. The extent of Internet use per se is not sufficient as an addiction criterion and other negative consequences; rather, specific adverse consequences need to be identified. If the Internet is used excessively to cope with negative affect states and alternative means of coping (e.g., social support, health-promoting behavior) are diminished, a vicious cycle may ensue with increasing stress and reliance on the reinforcing properties of certain online activities that may finally lead to addictive behavior.

This paper seeks to interpret the Gospel of John in light of new innovations: utilizing the prologue to interpret the body of the gospel, seeking a theological background for Jesus’ ἐγώ eἰμί statements, and viewing Jesus’ farewell discourses in light of liberation theology. John 14:1-14 challenges us to look beyond our present circumstances to the God that does not change, by presenting a discourse on the unity of the Triune God who is incarnate in Jesus Christ. By seeking to ground this passage in its literary context, the significance of the tribulation that Jesus and his disciples are currently experiencing clearly becomes the primary lens that proper interpretation and application of John 14:1-14 must proceed through.

This past summer, I submitted a proposal to David Tanenbaum (president of the upcoming Second American Classical Guitar Congress to be held in Winston-Salem, North Carolina) requestingthat we holdaforum and panel discussion on standardizing guitar notation. Mr. Tanenbaum enthusiastically endorsed the idea and made it part of the congress agenda. Subsequently,the board ofdirectors for the Guitar Foundation of Americaagreed that G.F.A. would cosponsor and expand this important project by publishing a book illustrating preferred methods of notation according to the combined recommendations of an appointedpanel and numerous project consultants. When one method of notatingafingering, technique, or musical effect is considered by theirconsensus to be superior to alternative methods, it will be advocated as such in the G.F.A. publication. In some cases, however, more than' onemethodmay beendorsed with regard to its specific applications. For instance, the justification for a particular placement of left-hand fingerings might vary according to the priorities of individual editors (see example 1, next page). In discussing the proposal with several of my colleagues, it became apparent to me that some would object to the term "standardization," since it might suggest an attempt to impose upon the rights of individual expression. Let me assure you that this is not our intention. As always, an editor would be expected to use his or her best judgement in determining how to communicate musicaland technical ideas. The study looks at the Corporate Social Responsibility (CSR) of Indian firms from various sectors and its impact on the performance and market execution of firms. The study looks at the CSR contribution of various firms and its impact on profit after tax over a three year period from 2013 to 2015. The study utilizes substance investigation to study CSR revelation and characterizes and rates these exercises utilizing things from a setup scale took after by development of class shrewd CSR records. This study also analyzes the CSR activities of various sectors in India by covering a wide range of activities done by various leading companies of these sectors. The study finds that CSR reporting may have any critical effect on market performance and market execution of the firm in the fleeting yet environment situated CSR exposure may be adversely identified with the business sector execution of the firm. The concentrate additionally finds that organizations concentrate intensely on representative and client arranged CSR and the methods of CSR ventures are more contributory as opposed to participative in nature. This study also finds that various leading sectors in India are very focused and dedicated in terms of CSR activities and there is a kind of competitive environment among the companies for participating in CSR activities, which is actually a very good sign.
