Background: Decreasing the rate of protein emptying from the stomach may improve efficiency of ut... more Background: Decreasing the rate of protein emptying from the stomach may improve efficiency of utilization of dietary amino acids for protein deposition. Some studies in rats and humans have shown casein to be more slowly released from the stomach than whey protein. To test if casein induces a slower rate of gastric emptying in cats than whey protein, L-[1-13 C]phenylalanine (Phe) was dosed orally into 9 adult cats to estimate gastric emptying and whole-body Phe flux. Results: Concentrations of indispensable amino acids in plasma were not significantly affected by dietary protein source. First-pass splanchnic extraction of Phe was not different between diets and averaged 50% (SEM = 3.8%). The half-time for gastric emptying averaged 9.9 min with casein and 10.3 min with whey protein, and was not significantly different between diets (SEM = 1.7 min). Phenylalanine fluxes were 45.3 and 46.5 μmol/(min • kg) for casein-and whey-based diets, respectively (SEM = 4.7 μmol/(min • kg)). Conclusions: In adult cats fed frequent small meals, the replacement of casein with whey protein in the diet does not affect supply or utilization of amino acids. These two milk proteins appear to be equally capable of meeting the dietary amino acid needs of cats.
The need for protein-rich plant-based foods continues as dietary guidelines emphasize their contr... more The need for protein-rich plant-based foods continues as dietary guidelines emphasize their contribution to healthy dietary patterns that prevent chronic disease and promote environmental sustainability. However, the Canadian Food and Drug Regulations provide a regulatory framework that can prevent Canadian consumers from identifying protein-rich plant-based foods. In Canada, protein nutrient content claims are based on the protein efficiency ratio (PER) and protein rating method, which is based on a rat growth bioassay. PERs are not additive, and the protein rating of a food is underpinned by its Reasonable Daily Intake. The restrictive nature of Canada's requirements for supporting protein claims therefore presents challenges for Canadian consumers to adapt to a rapidly changing food environment. This commentary will present two options for modernizing the regulatory framework for protein content claims in Canada. The first and preferred option advocates that protein quality not be considered in the determination of the eligibility of a food for protein content claims. The second and less preferred option, an interim solution, is a framework for adopting the protein digestibility corrected amino acid score as the official method for supporting protein content and quality claims and harmonizes Canada's regulatory framework with that of the USA.
Dietary starch is required for a dry, extruded kibble; the most common diet type for domesticated... more Dietary starch is required for a dry, extruded kibble; the most common diet type for domesticated felines in North America. However, the amount and source of dietary starch may affect digestibility and metabolism of other macronutrients. The objectives of this study were to evaluate the effects of 3 commercial cat diets on in vivo and in vitro energy and macronutrient digestibility, and to analyze the accuracy of the modified Atwater equation. Dietary treatments differed in their perceived glycemic response (PGR) based on ingredient composition and carbohydrate content (34.1, 29.5, and 23.6% nitrogen-free extract for High, Medium, and LowPGR, respectively). A replicated 3 × 3 Latin square design was used, with 3 diets and 3 periods. In vivo apparent protein, fat, and organic matter digestibility differed among diets, while apparent dry matter digestibility did not. Cats were able to efficiently digest and absorb macronutrients from all diets. Furthermore, the modified Atwater equation underestimated measured metabolizable energy by approximately 12%. Thus, the modified Atwater equation does not accurately determine the metabolizable energy of high quality feline diets. Further research should focus on understanding carbohydrate metabolism in cats, and establishing an equation that accurately predicts the metabolizable energy of feline diets.
Mannoheptulose (MH), a sugar found in avocados that inhibits glycolysis in vitro, has been prelim... more Mannoheptulose (MH), a sugar found in avocados that inhibits glycolysis in vitro, has been preliminarily investigated as a novel food ingredient for dogs. This study aimed to determine the effects of dietary MH, delivered as an extract of un-ripened avocado, on energy expenditure (EE) in healthy adult Labrador Retriever dogs (total of 12 dogs, 26.99 ± 0.634 kg, 4.9 ± 0.2 y). The study was a double-blind, cross-over with each dog receiving both dietary treatments, control (CON) and MH (400 mg/kg of diet; 6 mg/kg BW), in random order. Resting and post-prandial (10 h) EE and respiratory quotient (RQ) were determined by indirect calorimetry (d 42). The following day, body composition was assessed using dual X-ray absorptiometry. Continuous activity monitoring was conducted using an Atical1 accelerometer (d 43-47). A vastus lateralis muscle biopsy was obtained prior to the morning meal (d 49) and 4 h after consumption of their meal (d 56) to determine the protein content and phosphorylation of 5' adenosine monophosphate-activated protein kinase (AMPK). Diet did not affect body weight, resting EE or skeletal muscle AMPK phosphorylation. Dogs fed MH had significantly lower post-prandial RQ (p = 0.02) and ratio of fat to lean body mass (p = 0.02). Physical activity during light time periods (but not dark) was lower in dogs fed MH (p < 0.05) during weekends, but not on weekdays. These results suggest that MH affects energy balance of adult dogs, but that these effects are not dose dependent and not due to physical activity.
The objective of this study was to determine the standardized ileal digestibility (SID) of protei... more The objective of this study was to determine the standardized ileal digestibility (SID) of protein and amino acids (AA) and calculate the digestible indispensable AA score (DIAAS) of three varieties of Canadian grown pulses (faba bean, lentil, and yellow pea). Three steam-pelleted (80 • C) diets (faba/lentil/pea; 40% inclusion) and a nitrogen-free diet were fed to eight cannulated growing pigs in a 4 × 4 replicated Latin square design. The SID values were used to calculate DIAAS. There were no differences in SID of protein and AA when all diets were compared (P > 0.05). The SID of methionine was 95% for faba beans, 96% for lentils, and 96% for peas. The SID of lysine was 93% for faba beans, 92% for lentils, and 90% for peas. Tryptophan had the lowest SID across all indispensable AA of all pulses (59% faba, 61% lentil, and 41% pea). All other indispensable AA SID were above 80%. Tryptophan was the limiting indispensable AA for all pulses as determined by DIAAS, with scores of 30 (faba), 57.83 (lentil), and 27.27 (pea). These values will aid in optimal inclusion of steam-pelleted pulses in diets for swine and provide an insight on the protein quality of pulses for humans.

One adult male cat was used in two experiments. In each experiment, three isotope protocols were tested in triplicate using the same cat. During carbon oxidation study days, the cat was offered thirteen small meals to achieve and maintain a physiological fed state. In experiment 1, the isotope protocols tested (A, B and C) had a similar priming dose of NaH 13 CO 3 (0⋅176 mg/kg; offered in meal 6), but different priming [4⋅8 mg/kg (A) or 9⋅4 mg/kg (B and C); provided in meal 6] and constant [1⋅04 mg/kg (A and B) or 2⋅4 mg/kg (C); offered in meals 6-13] doses of L-[1-13 C]-Phe. In experiment 2, the isotope protocols tested (D, E and F) had similar priming (4⋅8 mg/kg; provided in meal 5) and constant (1⋅04 mg/kg; provided in meals 5-13) doses of L-[1-13 C]-Phe, but increasing priming doses of NaH 13 CO 3 (D: 0⋅264, E: 0⋅352, F: 0⋅44 mg/kg; provided in meal 4). Breath samples were collected using respiration chambers (25-min intervals) and CO 2 trapping to determine 13 CO 2 : 12 CO 2. Isotopic steady state was defined as the enrichment of 13 CO 2 , above background samples, remaining constant in at least the last three samples. Treatment F resulted in the earliest achievement of 13 CO 2 steady state in the cat's breath. This feeding and isotope protocol can be used in future studies aiming to study amino acid metabolism in cats.

Six adult male cats were used in a replicated 3 × 3 Latin square design. Three semi-synthetic diets were developed: a methionine (Met) and total sulfur AA (TSAA) deficient diet (T-BASAL; 0.24% Met+Cys − DM basis) and two Met and TSAA-sufficient diets in which either dl-Met (T-DLM) or 2-hydroxy-4-(methylthio)-butanoic acid (T-MHA) were supplemented, respectively, on an equimolar basis to meet the TSAA requirement (0.34%). After a 2-d diet adaptation, IAAO studies were performed. Cats were offered 13 small meals. The sixth meal contained a priming dose (4.8 mg/kg-BW) of l-[1-13C]-Phe and the remaining meals a constant dose (1.04 mg/kg-BW). Breath samples were collected every 25 min to measure 13CO2 enrichment. The following morning, fasted blood samples were collected. Cats returned to the T-BASAL top dressed with a dl-Met solution for 4 d prior to being fed a new dietary treatment. Isotopic steady state was evaluated through visual inspection. Data were analyzed using PROC GLIMMIX procedure in SAS 9.4. While 13CO2 enrichment was successfully captured in breath samples, cats failed to reach 13CO2 steady state. Thus, a non-steady state isotope model was developed and coded in ACSLX (V3.1.4.2) individually for each cat on each study day to predict 13CO2 enrichment, and then, calculate oxidation of l-[1-13C]-Phe (F13CO2). A higher predicted F13CO2 was observed for cats fed T-BASAL compared to the others (P &lt; 0.05), while no differences were observed between T-DLM and T-MHA (P &gt; 0.05). Cats fed T-DLM tended to have higher plasma Met concentrations compared to those fed T-BASAL with cats fed T-MHA intermediate (P = 0.0867). Plasma homocysteine concentrations were higher in cats fed T-BASAL compared to the others (P &lt; 0.05), while threonine concentrations tended to be higher in cats fed T-BASAL compared to those fed T-MHA (P = 0.0750). In conclusion, short-term provision of a semi-synthetic diet deficient in Met may elicit a metabolic response aiming to conserve Met. The successful quantification of 13CO2 enrichment in breath and the higher predicted F13CO2 in cats fed a Met deficient diet suggest that the IAAO technique may be used in cats. Adaptations in the isotope protocol should be made to achieve 13CO2 steady state in breath and avoid mathematical modeling to predict F13CO2.

Choline may work similarly in kittens at risk of obesity. Direct infusion MS (Di-MS) and NMR spectroscopy were used to investigate the metabolomic signatures of kittens supplemented with or without additional dietary choline for 12 weeks. Fifteen intact male kittens consumed a base diet (3310 mg choline/kg DM) to their daily metabolisable energy requirement (DER) over an 11-week acclimation. Kittens were gonadectomised and assigned, based on body weight, to the base diet (CONTROL, n 7) or the base diet with 300 mg/kgBW 0•75 additional choline as choline chloride (CHOLINE, n 8) and offered three times their individual energy requirement divided into three meals. At weeks −1 and 12, fasted blood was sampled and serum analysed for 130 metabolites via DiMS and fifty-one metabolites via NMR spectroscopy. Changes in fasted metabolites were assessed using a repeated-measures GLIMMIX procedure with time and group as fixed effects, and time as a repeated measure. Metabolites of one-carbon metabolism and lipids increased, and medium-chain acyl carnitines decreased from week −1 to 12 for CHOLINE (P < 0•05), but not CONTROL (P > 0•05). Increases in amino acid, biogenic amine and organic compound concentrations were observed in both groups (P < 0•05). The results suggest impacts of dietary choline at greater intakes than currently recommended on one-carbon metabolism and fatty acid oxidation, and these may promote healthy growth in postgonadectomy kittens.

Chemically defined diets are commonly used in amino acid (AA) requirement studies to allow for ti... more Chemically defined diets are commonly used in amino acid (AA) requirement studies to allow for tight control of AA delivery. However, those diets are not representative of commercial diets in the market and are unpalatable. Methionine (Met) is usually the first limiting AA in cat diets, but little is known about its requirement for adult cats. Thus, the objectives of this study were: 1) to develop a semisynthetic diet limiting in Met and evaluate its effect on acceptance and feeding behavior in cats; and 2) to evaluate the effect of different sources and inclusions of Met on preference in cats fed the semisynthetic diet. A semisynthetic diet deficient in Met and total sulfur AA (TSAA) was developed. Healthy adult male cats (n = 9) were fed (0800 and 1600 h) the semisynthetic diet top dressed with DL-Met solution (T-DLM), to meet 120% of the TSAA requirement, for 8 d. Feed intake was measured and a 30-min video recording was taken at the 0800 h feeding to evaluate feeding behavior of the cats. Following the acceptability trial, two bowl tests were performed where first choice was recorded and intake ratio was calculated as consumed food (A/A + B). Three combinations were tested: semisynthetic diet deficient in Met (T-BASAL) vs. T-DLM; T-BASAL vs. diet sufficient in Met provided 2-hydroxy-4-(methylthio)-butanoic acid (T-MHA); and T-DLM vs. T-MHA. Average feed intake remained high throughout the acceptability period (94.5% intake of total offered), but some cats decreased intake, resulting in a decrease in BW (≤2.5% of initial BW) over time (P &lt; 0.05). Behaviors were similar among days (P &gt; 0.05) with the exception of grooming the chest and body (P &lt; 0.05). No preferences were observed towards a specific treatment (Met source and level) during the two-bowl tests (P &gt; 0.05) and agreed with the cats expressing similar feeding behaviors during the preference tests (P &gt; 0.05). In conclusion, a semisynthetic diet deficient in Met was successfully developed and can be used in studies to evaluate the effects of low protein and AA supplemented diets. Cats seem to show no preference for Met source and/or inclusion level in a semisynthetic diet application, which is of benefit for future studies aiming to determine the Met requirement in this species.
Canine obesity is becoming an increasingly prevalent concern among companion animal veterinarians... more Canine obesity is becoming an increasingly prevalent concern among companion animal veterinarians and professionals alike. A number of sociodemographic, dietary, and exercise related variables have been shown to be predictive of a dog's bodyweight, however, all previous surveys designed to address these variables have been focussed on only one area of the world at a time. The objective of this survey was to investigate how an owner's exercise routine influences their dog's exercise routine and which of the owner's dietary and exercise habits influence their perception of their dog's body weight. The survey included respondents across France, Germany, the United Kingdom, Canada and the United States. The survey was distributed online via Qualtrics (Qualtrics XM, Utah, USA) and a total of 3,298 responses were collected, equally distributed across country and between sexes. Comparison of column proportions and multinomial logistic regression were performed in SPSS Statistics (Version 26, IBM Corp, North Castle, New York, USA). Respondents from Germany were more likely to exercise their dog for a longer amount of time, rank the importance of exercise as extremely important, report that their dog is an ideal body weight, and were less likely to report that someone (including a veterinarian) had told them their dog was overweight. Results from linear regression revealed that those who had been told their dog was overweight, those who restrict their dog's food intake to control weight, those who select a weight control diet and those who give their dog more other foods (treats, table scraps, fruits/vegetables) on a daily basis were all less likely to believe that their dog is an ideal body weight. In contrast, only those who reported doing more vigorous exercise themselves or those who reported that their dog performs vigorous exercise were more likely to believe that their dog is an ideal body weight. The results highlight owner's perceptions of healthy weight and the role of nutrition and exercise. Owner's intentions and attitudes towards the value of exercise and promoting an ideal body weight in their dog should be explored, but may require a One Health approach to ensure successful weight management among both dogs and their owners.
The ability of a diet or an ingredient to satisfy the indispensable amino acid (IAA) requirements... more The ability of a diet or an ingredient to satisfy the indispensable amino acid (IAA) requirements of an individual is a reflection of protein quality (PQ). The concept of PQ is gaining recognition in the pet food industry as a way to identify candidate ingredients for diet formulation. The objective of this report was to use IAA digestibility data from swine and cecectomized rooster assays to generate digestible IAA scores (defined herein as DIAAS-like values) to predict the PQ of ingredients used in dog diets. However, as PQ equation development relies on a reference IAA profile, which is intended to be based on the physiological requirements of a specific population, we sought to generate DIAAS-like values using IAA requirements established by the National Research Council (NRC) as well as practical IAA recommendations presented by the Association of American Feed Control Officials (AAFCO) and European Pet Food Industry Federation (FEDIAF), to assess how these profiles may affect PQ. In total, 30 animal (75 unique inputs) and 27 plant ingredients (94 unique inputs) satisfied all inclusion criteria to be used in the final data set. Ingredients were initially categorized as animal or plant, and further categorized based on AAFCO Official Common and Usual Names and Definitions of Feed Ingredients to allow for additional, more distinct comparisons to be made. Data were analyzed using PROC GLIMMIX in SAS, with ingredient reference as a random effect, and ingredient category, regulatory body, and life stage as fixed effects. As expected, differences were observed in DIAAS-like values for nearly all ingredients and ingredient categories when determined using NRC, AAFCO, or FEDIAF IAA requirements or recommendations as the reference pattern. Moreover, applying reference patterns based on NRC adult maintenance IAA requirements consistently produced the lowest DIAAS-like values. Ultimately, while future studies assessing PQ should utilize NRC minimal requirements, individual ingredient and ingredient category differences in DIAAS-like values when using AAFCO and FEDIAF recommendations underpin the different regulatory approaches to establishing dietary nutrient recommendations that exist globally and support the need for harmonization of dietary recommendations.

The lipotropic effects of choline have demonstrated benefits for growth and carcass composition in livestock. The consumption of supplemental choline on body weight (BW), body composition, lipid metabolism, energy expenditure (EE), and serum satiety hormones were evaluated in 15 gonadectomized male kittens. Kittens were offered a base diet formulated for growth (3310mg choline/kg dry matter [DM]) to daily energy requirements (DER) over an 11-week acclimation. Post-gonadectomy, kittens were assigned to a base diet (CONTROL, n = 7) or choline group (base diet with additional choline at 300mg/kg BW 0.75 as a top dress) (CHOLINE, n = 8). For 12-weeks post-neuter, kittens were offered three times their DER over three meals to mimic ad libitum feeding. At week-1 and 12, body composition was assessed using dual energy x-ray absorptiometry (DXA), 24-hour indirect calorimetry was performed for EE and respiratory quotients (RQ), and fasted serum samples were analyzed for lipid compounds and satiety hormones. Daily food intake (FI) and weekly BW were measured. Data was analyzed as a repeated measures of variance (ANCOVA) using the GLIMMIX procedure with time and group as fixed effects. CHOLINE had lower mean daily FI and lower rates of BW accretion (P<0.05) in contrast to CONTROL. All absolute body composition data increased over time for both groups, with lower increases in total tissue mass (P = 0.031) and fat mass (P = 0.005) in CHOLINE. Serum satiety hormones and lipid compounds did not differ (P>0.05) between groups, but both groups experienced a decrease in low-density lipoproteins and increase in high-density lipoproteins (P<0.05). Primary substrate utilization showed lipid use when fasted and use of protein or mixed macronutrients in the fed state. Fed state EE decreased post-gonadectomy (P = 0.004), however, CHOLINE did not affect total EE or RQ. These results suggest that supplemental dietary choline reduces FI, BW, and fat mass

This study investigated whether feeding adult cats with diets containing either 188 ppm of LC (LC188) or 121 ppm of LC (LC121) and feeding them 120% of maintenance energy requirement (MER) resulted in differences in total energy expenditure (EE), metabolic fuel selection, BW, body composition, and behavior. Cats (= 20, 4 ± 1.2 yrs) were stratified for BCS and randomly assigned to one of two dietary treatments and fed for 16 weeks. BW was measured weekly, and indirect calorimetry, body composition, physical activity, play motivation, and cognition were measured at baseline and throughout the study. A mixed, repeated measures, ANCOVA model was used. Cats in both treatments gained BW (< 0.05) throughout the study, with no differences between treatments at any time point (> 0.05). There were no differences in body composition between groups at baseline; however, body fat (g) and body fat : lean mass ratio were greater in cats fed LC121 in contrast to cats fed LC188 (< 0.05) on week 16. No other outcomes differed between treatments (> 0.05). Supplying dietary LC at a dose of at least 188 ppm may be beneficial for the health and well-being of cats fed above MER.
The effects of dietary carbohydrate and fat on feline health are not well understood. The effects... more The effects of dietary carbohydrate and fat on feline health are not well understood. The effects of feeding diets moderately high in fat (HF; n 10; 30 % fat, 26 % carbohydrate as fed) or carbohydrate (HC; n 10; 11 % fat, 47 % carbohydrate), for 84 d, were investigated in healthy, adult cats (3•5 (SD 0•5) years). Data on indirect calorimetry, blood biomarkers, activity, play and cognition were collected at baseline, and at intervals throughout the study. Body composition was measured by dual-energy X-ray absorptiometry at baseline and on day 85. There were no significant main effects of diet on body weight and composition. When data were analysed over study day within diet, cats fed HF diets experienced a significant increase in body fat (P = 0•001) and body weight (P = 0•043) in contrast to cats consuming the HC diet that experienced no change in body fat or body weight (P = 0•762) throughout the study. Overall, energy expenditure was similar between diets (P = 0•356 (fasted), P = 0•086 (postprandial)) and respiratory quotient declined with exposure to the HF diet and increased with exposure to the HC diet (P < 0•001; fasted and postprandial). There was no difference in insulin sensitivity as an overall effect of diet (P = 0•266). Activity declined from baseline with exposure to both diets (HC: P = 0•002; HF: P = 0•01) but was not different between diets (P = 0•247). There was no effect of diet on play (P = 0•387) and cats consuming either the HF or HC diet did not successfully learn the cognitive test. Overall, cats adapt to dietary macronutrient content, and the implications of feeding HC and HF diets on risk for adiposity as driven by metabolic and behavioural mechanisms are discussed.

High-fat (HF) or high-carbohydrate (HC) diets (30% fat, 18.9% carbohydrate; HF and 10% fat, 46.3%... more High-fat (HF) or high-carbohydrate (HC) diets (30% fat, 18.9% carbohydrate; HF and 10% fat, 46.3% carbohydrate; HC) and lengths of adaptation were investigated in cats (Felis catus; 10 ± 2 months, 3.6 ± 0.3 kg). Cats randomly received each treatment for 14 days in a crossover design with a 14-day washout period between each diet. Three 22-h indirect calorimetry studies were conducted after acute (day 0), semichronic (day 4) and chronic (day 13) dietary exposure. Blood samples were collected after a 24-h fast on days 1, 5 and 14. When cats consumed the HC and HF diet, oxidation of the restricted nutrient exceeded intake while oxidation of the nutrient in excess matched intake. Mean max energy expenditure (EE) of cats consuming the HF and HC diet were 107 and 102 kcal/kg(0.67)/day and occurred at a mean of 4 and 12 h post-feeding respectively. Maximal fat (0.90 g/h) and carbohydrate (carbohydrate; 1.42 g/h) oxidation were attained at 26 min and 10.4 h post-feeding respectively. The changes observed in macronutrient oxidation and EE suggest that cats adapt whole-body nutrient metabolism in response to changes in dietary macronutrient content, but may require longer than 14 day to adapt to a macronutrient that is present at a lower concentration in the diet.

The half-time for gastric emptying averaged 9.9 min with casein and 10.3 min with whey protein, and was not significantly different between diets (SEM = 1.7 min). Phenylalanine fluxes were 45.3 and 46.5 μmol/(min • kg) for casein-and whey-based diets, respectively (SEM = 4.7 μmol/(min • kg)). Conclusions: In adult cats fed frequent small meals, the replacement of casein with whey protein in the diet does not affect supply or utilization of amino acids. These two milk proteins appear to be equally capable of meeting the dietary amino acid needs of cats.
The need for protein-rich plant-based foods continues as dietary guidelines emphasize their contr... more The need for protein-rich plant-based foods continues as dietary guidelines emphasize their contribution to healthy dietary patterns that prevent chronic disease and promote environmental sustainability. However, the Canadian Food and Drug Regulations provide a regulatory framework that can prevent Canadian consumers from identifying protein-rich plant-based foods. In Canada, protein nutrient content claims are based on the protein efficiency ratio (PER) and protein rating method, which is based on a rat growth bioassay. PERs are not additive, and the protein rating of a food is underpinned by its Reasonable Daily Intake. The restrictive nature of Canada's requirements for supporting protein claims therefore presents challenges for Canadian consumers to adapt to a rapidly changing food environment. This commentary will present two options for modernizing the regulatory framework for protein content claims in Canada. The first and preferred option advocates that protein quality not be considered in the determination of the eligibility of a food for protein content claims. The second and less preferred option, an interim solution, is a framework for adopting the protein digestibility corrected amino acid score as the official method for supporting protein content and quality claims and harmonizes Canada's regulatory framework with that of the USA.

However, the amount and source of dietary starch may affect digestibility and metabolism of other macronutrients. The objectives of this study were to evaluate the effects of 3 commercial cat diets on in vivo and in vitro energy and macronutrient digestibility, and to analyze the accuracy of the modified Atwater equation. Dietary treatments differed in their perceived glycemic response (PGR) based on ingredient composition and carbohydrate content (34.1, 29.5, and 23.6% nitrogen-free extract for High, Medium, and LowPGR, respectively). A replicated 3 × 3 Latin square design was used, with 3 diets and 3 periods. In vivo apparent protein, fat, and organic matter digestibility differed among diets, while apparent dry matter digestibility did not. Cats were able to efficiently digest and absorb macronutrients from all diets. Furthermore, the modified Atwater equation underestimated measured metabolizable energy by approximately 12%. Thus, the modified Atwater equation does not accurately determine the metabolizable energy of high quality feline diets. Further research should focus on understanding carbohydrate metabolism in cats, and establishing an equation that accurately predicts the metabolizable energy of feline diets.
Mannoheptulose (MH), a sugar found in avocados that inhibits glycolysis in vitro, has been prelim... more Mannoheptulose (MH), a sugar found in avocados that inhibits glycolysis in vitro, has been preliminarily investigated as a novel food ingredient for dogs. This study aimed to determine the effects of dietary MH, delivered as an extract of un-ripened avocado, on energy expenditure (EE) in healthy adult Labrador Retriever dogs (total of 12 dogs, 26.99 ± 0.634 kg, 4.9 ± 0.2 y). The study was a double-blind, cross-over with each dog receiving both dietary treatments, control (CON) and MH (400 mg/kg of diet; 6 mg/kg BW), in random order. Resting and post-prandial (10 h) EE and respiratory quotient (RQ) were determined by indirect calorimetry (d 42). The following day, body composition was assessed using dual X-ray absorptiometry. Continuous activity monitoring was conducted using an Atical1 accelerometer (d 43-47). A vastus lateralis muscle biopsy was obtained prior to the morning meal (d 49) and 4 h after consumption of their meal (d 56) to determine the protein content and phosphorylation of 5' adenosine monophosphate-activated protein kinase (AMPK). Diet did not affect body weight, resting EE or skeletal muscle AMPK phosphorylation. Dogs fed MH had significantly lower post-prandial RQ (p = 0.02) and ratio of fat to lean body mass (p = 0.02). Physical activity during light time periods (but not dark) was lower in dogs fed MH (p < 0.05) during weekends, but not on weekdays. These results suggest that MH affects energy balance of adult dogs, but that these effects are not dose dependent and not due to physical activity.
The objective of this study was to determine the standardized ileal digestibility (SID) of protei... more The objective of this study was to determine the standardized ileal digestibility (SID) of protein and amino acids (AA) and calculate the digestible indispensable AA score (DIAAS) of three varieties of Canadian grown pulses (faba bean, lentil, and yellow pea). Three steam-pelleted (80 • C) diets (faba/lentil/pea; 40% inclusion) and a nitrogen-free diet were fed to eight cannulated growing pigs in a 4 × 4 replicated Latin square design. The SID values were used to calculate DIAAS. There were no differences in SID of protein and AA when all diets were compared (P > 0.05). The SID of methionine was 95% for faba beans, 96% for lentils, and 96% for peas. The SID of lysine was 93% for faba beans, 92% for lentils, and 90% for peas. Tryptophan had the lowest SID across all indispensable AA of all pulses (59% faba, 61% lentil, and 41% pea). All other indispensable AA SID were above 80%. Tryptophan was the limiting indispensable AA for all pulses as determined by DIAAS, with scores of 30 (faba), 57.83 (lentil), and 27.27 (pea). These values will aid in optimal inclusion of steam-pelleted pulses in diets for swine and provide an insight on the protein quality of pulses for humans.
The present study aimed to develop an isotope protocol to achieve equilibrium of 13 CO 2 in breat... more The present study aimed to develop an isotope protocol to achieve equilibrium of 13 CO 2 in breath of cats during carbon oxidation studies using L-[1-13 C]-Phenylalanine (L-[1-13 C]-Phe), provided orally in repeated meals. One adult male cat was used in two experiments. In each experiment, three isotope protocols were tested in triplicate using the same cat. During carbon oxidation study days, the cat was offered thirteen small meals to achieve and maintain a physiological fed state. In experiment 1, the isotope protocols tested (A, B and C) had a similar priming dose of NaH 13 CO 3 (0⋅176 mg/kg; offered in meal 6), but different priming [4⋅8 mg/kg (A) or 9⋅4 mg/kg (B and C); provided in meal 6] and constant [1⋅04 mg/kg (A and B) or 2⋅4 mg/kg (C); offered in meals 6-13] doses of L-[1-13 C]-Phe. In experiment 2, the isotope protocols tested (D, E and F) had similar priming (4⋅8 mg/kg; provided in meal 5) and constant (1⋅04 mg/kg; provided in meals 5-13) doses of L-[1-13 C]-Phe, but increasing priming doses of NaH 13 CO 3 (D: 0⋅264, E: 0⋅352, F: 0⋅44 mg/kg; provided in meal 4). Breath samples were collected using respiration chambers (25-min intervals) and CO 2 trapping to determine 13 CO 2 : 12 CO 2. Isotopic steady state was defined as the enrichment of 13 CO 2 , above background samples, remaining constant in at least the last three samples. Treatment F resulted in the earliest achievement of 13 CO 2 steady state in the cat's breath. This feeding and isotope protocol can be used in future studies aiming to study amino acid metabolism in cats.

Six adult male cats were used in a replicated 3 × 3 Latin square design. Three semi-synthetic diets were developed: a methionine (Met) and total sulfur AA (TSAA) deficient diet (T-BASAL; 0.24% Met+Cys − DM basis) and two Met and TSAA-sufficient diets in which either dl-Met (T-DLM) or 2-hydroxy-4-(methylthio)-butanoic acid (T-MHA) were supplemented, respectively, on an equimolar basis to meet the TSAA requirement (0.34%). After a 2-d diet adaptation, IAAO studies were performed. Cats were offered 13 small meals. The sixth meal contained a priming dose (4.8 mg/kg-BW) of l-[1-13C]-Phe and the remaining meals a constant dose (1.04 mg/kg-BW). Breath samples were collected every 25 min to measure 13CO2 enrichment. The following morning, fasted blood samples were collected. Cats returned to the T-BASAL top dressed with a dl-Met solution for 4 d prior to being fed a new dietary treatment. Isotopic steady state was evaluated through visual inspection. Data were analyzed using PROC GLIMMIX procedure in SAS 9.4. While 13CO2 enrichment was successfully captured in breath samples, cats failed to reach 13CO2 steady state. Thus, a non-steady state isotope model was developed and coded in ACSLX (V3.1.4.2) individually for each cat on each study day to predict 13CO2 enrichment, and then, calculate oxidation of l-[1-13C]-Phe (F13CO2). A higher predicted F13CO2 was observed for cats fed T-BASAL compared to the others (P &lt; 0.05), while no differences were observed between T-DLM and T-MHA (P &gt; 0.05). Cats fed T-DLM tended to have higher plasma Met concentrations compared to those fed T-BASAL with cats fed T-MHA intermediate (P = 0.0867). Plasma homocysteine concentrations were higher in cats fed T-BASAL compared to the others (P &lt; 0.05), while threonine concentrations tended to be higher in cats fed T-BASAL compared to those fed T-MHA (P = 0.0750). In conclusion, short-term provision of a semi-synthetic diet deficient in Met may elicit a metabolic response aiming to conserve Met. The successful quantification of 13CO2 enrichment in breath and the higher predicted F13CO2 in cats fed a Met deficient diet suggest that the IAAO technique may be used in cats. Adaptations in the isotope protocol should be made to achieve 13CO2 steady state in breath and avoid mathematical modeling to predict F13CO2.

Choline may work similarly in kittens at risk of obesity. Direct infusion MS (Di-MS) and NMR spectroscopy were used to investigate the metabolomic signatures of kittens supplemented with or without additional dietary choline for 12 weeks. Fifteen intact male kittens consumed a base diet (3310 mg choline/kg DM) to their daily metabolisable energy requirement (DER) over an 11-week acclimation. Kittens were gonadectomised and assigned, based on body weight, to the base diet (CONTROL, n 7) or the base diet with 300 mg/kgBW 0•75 additional choline as choline chloride (CHOLINE, n 8) and offered three times their individual energy requirement divided into three meals. At weeks −1 and 12, fasted blood was sampled and serum analysed for 130 metabolites via DiMS and fifty-one metabolites via NMR spectroscopy. Changes in fasted metabolites were assessed using a repeated-measures GLIMMIX procedure with time and group as fixed effects, and time as a repeated measure. Metabolites of one-carbon metabolism and lipids increased, and medium-chain acyl carnitines decreased from week −1 to 12 for CHOLINE (P < 0•05), but not CONTROL (P > 0•05). Increases in amino acid, biogenic amine and organic compound concentrations were observed in both groups (P < 0•05). The results suggest impacts of dietary choline at greater intakes than currently recommended on one-carbon metabolism and fatty acid oxidation, and these may promote healthy growth in postgonadectomy kittens.

Chemically defined diets are commonly used in amino acid (AA) requirement studies to allow for ti... more Chemically defined diets are commonly used in amino acid (AA) requirement studies to allow for tight control of AA delivery. However, those diets are not representative of commercial diets in the market and are unpalatable. Methionine (Met) is usually the first limiting AA in cat diets, but little is known about its requirement for adult cats. Thus, the objectives of this study were: 1) to develop a semisynthetic diet limiting in Met and evaluate its effect on acceptance and feeding behavior in cats; and 2) to evaluate the effect of different sources and inclusions of Met on preference in cats fed the semisynthetic diet. A semisynthetic diet deficient in Met and total sulfur AA (TSAA) was developed. Healthy adult male cats (n = 9) were fed (0800 and 1600 h) the semisynthetic diet top dressed with DL-Met solution (T-DLM), to meet 120% of the TSAA requirement, for 8 d. Feed intake was measured and a 30-min video recording was taken at the 0800 h feeding to evaluate feeding behavior of the cats. Following the acceptability trial, two bowl tests were performed where first choice was recorded and intake ratio was calculated as consumed food (A/A + B). Three combinations were tested: semisynthetic diet deficient in Met (T-BASAL) vs. T-DLM; T-BASAL vs. diet sufficient in Met provided 2-hydroxy-4-(methylthio)-butanoic acid (T-MHA); and T-DLM vs. T-MHA. Average feed intake remained high throughout the acceptability period (94.5% intake of total offered), but some cats decreased intake, resulting in a decrease in BW (≤2.5% of initial BW) over time (P &lt; 0.05). Behaviors were similar among days (P &gt; 0.05) with the exception of grooming the chest and body (P &lt; 0.05). No preferences were observed towards a specific treatment (Met source and level) during the two-bowl tests (P &gt; 0.05) and agreed with the cats expressing similar feeding behaviors during the preference tests (P &gt; 0.05). In conclusion, a semisynthetic diet deficient in Met was successfully developed and can be used in studies to evaluate the effects of low protein and AA supplemented diets. Cats seem to show no preference for Met source and/or inclusion level in a semisynthetic diet application, which is of benefit for future studies aiming to determine the Met requirement in this species.
Canine obesity is becoming an increasingly prevalent concern among companion animal veterinarians... more Canine obesity is becoming an increasingly prevalent concern among companion animal veterinarians and professionals alike. A number of sociodemographic, dietary, and exercise related variables have been shown to be predictive of a dog's bodyweight, however, all previous surveys designed to address these variables have been focussed on only one area of the world at a time. The objective of this survey was to investigate how an owner's exercise routine influences their dog's exercise routine and which of the owner's dietary and exercise habits influence their perception of their dog's body weight. The survey included respondents across France, Germany, the United Kingdom, Canada and the United States. The survey was distributed online via Qualtrics (Qualtrics XM, Utah, USA) and a total of 3,298 responses were collected, equally distributed across country and between sexes. Comparison of column proportions and multinomial logistic regression were performed in SPSS Statistics (Version 26, IBM Corp, North Castle, New York, USA). Respondents from Germany were more likely to exercise their dog for a longer amount of time, rank the importance of exercise as extremely important, report that their dog is an ideal body weight, and were less likely to report that someone (including a veterinarian) had told them their dog was overweight. Results from linear regression revealed that those who had been told their dog was overweight, those who restrict their dog's food intake to control weight, those who select a weight control diet and those who give their dog more other foods (treats, table scraps, fruits/vegetables) on a daily basis were all less likely to believe that their dog is an ideal body weight. In contrast, only those who reported doing more vigorous exercise themselves or those who reported that their dog performs vigorous exercise were more likely to believe that their dog is an ideal body weight. The results highlight owner's perceptions of healthy weight and the role of nutrition and exercise. Owner's intentions and attitudes towards the value of exercise and promoting an ideal body weight in their dog should be explored, but may require a One Health approach to ensure successful weight management among both dogs and their owners.
The ability of a diet or an ingredient to satisfy the indispensable amino acid (IAA) requirements... more The ability of a diet or an ingredient to satisfy the indispensable amino acid (IAA) requirements of an individual is a reflection of protein quality (PQ). The concept of PQ is gaining recognition in the pet food industry as a way to identify candidate ingredients for diet formulation. The objective of this report was to use IAA digestibility data from swine and cecectomized rooster assays to generate digestible IAA scores (defined herein as DIAAS-like values) to predict the PQ of ingredients used in dog diets. However, as PQ equation development relies on a reference IAA profile, which is intended to be based on the physiological requirements of a specific population, we sought to generate DIAAS-like values using IAA requirements established by the National Research Council (NRC) as well as practical IAA recommendations presented by the Association of American Feed Control Officials (AAFCO) and European Pet Food Industry Federation (FEDIAF), to assess how these profiles may affect PQ. In total, 30 animal (75 unique inputs) and 27 plant ingredients (94 unique inputs) satisfied all inclusion criteria to be used in the final data set. Ingredients were initially categorized as animal or plant, and further categorized based on AAFCO Official Common and Usual Names and Definitions of Feed Ingredients to allow for additional, more distinct comparisons to be made. Data were analyzed using PROC GLIMMIX in SAS, with ingredient reference as a random effect, and ingredient category, regulatory body, and life stage as fixed effects. As expected, differences were observed in DIAAS-like values for nearly all ingredients and ingredient categories when determined using NRC, AAFCO, or FEDIAF IAA requirements or recommendations as the reference pattern. Moreover, applying reference patterns based on NRC adult maintenance IAA requirements consistently produced the lowest DIAAS-like values. Ultimately, while future studies assessing PQ should utilize NRC minimal requirements, individual ingredient and ingredient category differences in DIAAS-like values when using AAFCO and FEDIAF recommendations underpin the different regulatory approaches to establishing dietary nutrient recommendations that exist globally and support the need for harmonization of dietary recommendations.

The lipotropic effects of choline have demonstrated benefits for growth and carcass composition in livestock. The consumption of supplemental choline on body weight (BW), body composition, lipid metabolism, energy expenditure (EE), and serum satiety hormones were evaluated in 15 gonadectomized male kittens. Kittens were offered a base diet formulated for growth (3310mg choline/kg dry matter [DM]) to daily energy requirements (DER) over an 11-week acclimation. Post-gonadectomy, kittens were assigned to a base diet (CONTROL, n = 7) or choline group (base diet with additional choline at 300mg/kg BW 0.75 as a top dress) (CHOLINE, n = 8). For 12-weeks post-neuter, kittens were offered three times their DER over three meals to mimic ad libitum feeding. At week-1 and 12, body composition was assessed using dual energy x-ray absorptiometry (DXA), 24-hour indirect calorimetry was performed for EE and respiratory quotients (RQ), and fasted serum samples were analyzed for lipid compounds and satiety hormones. Daily food intake (FI) and weekly BW were measured. Data was analyzed as a repeated measures of variance (ANCOVA) using the GLIMMIX procedure with time and group as fixed effects. CHOLINE had lower mean daily FI and lower rates of BW accretion (P<0.05) in contrast to CONTROL. All absolute body composition data increased over time for both groups, with lower increases in total tissue mass (P = 0.031) and fat mass (P = 0.005) in CHOLINE. Serum satiety hormones and lipid compounds did not differ (P>0.05) between groups, but both groups experienced a decrease in low-density lipoproteins and increase in high-density lipoproteins (P<0.05). Primary substrate utilization showed lipid use when fasted and use of protein or mixed macronutrients in the fed state. Fed state EE decreased post-gonadectomy (P = 0.004), however, CHOLINE did not affect total EE or RQ. These results suggest that supplemental dietary choline reduces FI, BW, and fat mass

This study investigated whether feeding adult cats with diets containing either 188 ppm of LC (LC188) or 121 ppm of LC (LC121) and feeding them 120% of maintenance energy requirement (MER) resulted in differences in total energy expenditure (EE), metabolic fuel selection, BW, body composition, and behavior. Cats (= 20, 4 ± 1.2 yrs) were stratified for BCS and randomly assigned to one of two dietary treatments and fed for 16 weeks. BW was measured weekly, and indirect calorimetry, body composition, physical activity, play motivation, and cognition were measured at baseline and throughout the study. A mixed, repeated measures, ANCOVA model was used. Cats in both treatments gained BW (< 0.05) throughout the study, with no differences between treatments at any time point (> 0.05). There were no differences in body composition between groups at baseline; however, body fat (g) and body fat : lean mass ratio were greater in cats fed LC121 in contrast to cats fed LC188 (< 0.05) on week 16. No other outcomes differed between treatments (> 0.05). Supplying dietary LC at a dose of at least 188 ppm may be beneficial for the health and well-being of cats fed above MER.

The effects of feeding diets moderately high in fat (HF; n 10; 30 % fat, 26 % carbohydrate as fed) or carbohydrate (HC; n 10; 11 % fat, 47 % carbohydrate), for 84 d, were investigated in healthy, adult cats (3•5 (SD 0•5) years). Data on indirect calorimetry, blood biomarkers, activity, play and cognition were collected at baseline, and at intervals throughout the study. Body composition was measured by dual-energy X-ray absorptiometry at baseline and on day 85. There were no significant main effects of diet on body weight and composition. When data were analysed over study day within diet, cats fed HF diets experienced a significant increase in body fat (P = 0•001) and body weight (P = 0•043) in contrast to cats consuming the HC diet that experienced no change in body fat or body weight (P = 0•762) throughout the study. Overall, energy expenditure was similar between diets (P = 0•356 (fasted), P = 0•086 (postprandial)) and respiratory quotient declined with exposure to the HF diet and increased with exposure to the HC diet (P < 0•001; fasted and postprandial). There was no difference in insulin sensitivity as an overall effect of diet (P = 0•266). Activity declined from baseline with exposure to both diets (HC: P = 0•002; HF: P = 0•01) but was not different between diets (P = 0•247). There was no effect of diet on play (P = 0•387) and cats consuming either the HF or HC diet did not successfully learn the cognitive test. Overall, cats adapt to dietary macronutrient content, and the implications of feeding HC and HF diets on risk for adiposity as driven by metabolic and behavioural mechanisms are discussed.

High-fat (HF) or high-carbohydrate (HC) diets (30% fat, 18.9% carbohydrate; HF and 10% fat, 46.3%... more High-fat (HF) or high-carbohydrate (HC) diets (30% fat, 18.9% carbohydrate; HF and 10% fat, 46.3% carbohydrate; HC) and lengths of adaptation were investigated in cats (Felis catus; 10 ± 2 months, 3.6 ± 0.3 kg). Cats randomly received each treatment for 14 days in a crossover design with a 14-day washout period between each diet. Three 22-h indirect calorimetry studies were conducted after acute (day 0), semichronic (day 4) and chronic (day 13) dietary exposure. Blood samples were collected after a 24-h fast on days 1, 5 and 14. When cats consumed the HC and HF diet, oxidation of the restricted nutrient exceeded intake while oxidation of the nutrient in excess matched intake. Mean max energy expenditure (EE) of cats consuming the HF and HC diet were 107 and 102 kcal/kg(0.67)/day and occurred at a mean of 4 and 12 h post-feeding respectively. Maximal fat (0.90 g/h) and carbohydrate (carbohydrate; 1.42 g/h) oxidation were attained at 26 min and 10.4 h post-feeding respectively. The changes observed in macronutrient oxidation and EE suggest that cats adapt whole-body nutrient metabolism in response to changes in dietary macronutrient content, but may require longer than 14 day to adapt to a macronutrient that is present at a lower concentration in the diet.
