Pasien biasanya datang pada stadium lanjut, sehingga pengobatan dengan kemoterapi tidak memberikan hasil yang diharapkan karena adanya potensi resistensi terhadap obat antitumor. Pada tingkat molekuler, microRNA-21 (hsa-miR-21) berperan sebagai post transcriptional regulator dalam mekanisme regulasi pasca transkripsi sehingga menekan atau menghambat translasi mRNA gen tumor supresor Phosphatase Tensin Homolog (PTEN). Penurunan atau hilangnya ekspresi PTEN berdampak meningkatnya proliferasi sel dan inhibisi jalur apoptosis yang akhirnya memicu tumorigenesis dan resistensi kanker payudara. Untuk memahami mekanisme tersebut, dilakukan eksperimen in vitro pada kultur sel lini MCF-7 resisten Doxorubicin guna mengetahui ekspresi hsa-miR-21 dan PTEN. Dilakukan analisis in silico untuk memprediksi interaksi hsa-miR-21 dan mRNA PTEN berdasarkan basis data bioinformatik. Eksperimen in vitro dilakukan dengan metode paparan Doxorubicin secara bertingkat pada sel MCF-7 sehingga diperoleh varian sel resisten Doxorubicin (MCF-7/Dox) berdasarkan karakteristik peningkatan nilai IC50 melalui MTT assay. Ekspresi protein P-gp dan PTEN dideteksi secara imunositokimia dengan antibodi monoklonal. Adapun analisis ekspresi hsa-miR-21 dilakukan menggunakan qRT-PCR dengan U6SnRNA sebagai reference gene dan UniSp6 sebagai kontrol positif. Hasil menunjukkan bahwa terdapat peningkatan nilai IC50 pada sel MCF-7/Dox (5,78 I¼g/ml) dibandingkan MCF-7 parental (0,68 I¼g/ml) dengan indeks resistensi sebesar 8,5. Analisis qRT-PCR membuktikan bahwa hsa-miR-21 mengalami overekspresi pada sel lini MCF-7/Dox dengan fold change expression 7,94. Meningkatnya regulasi hsa-miR-21 seiring dengan penurunan ekspresi protein PTEN pada sel MCF-7/Dox. Secara imunositokimia terdeteksi ekspresi P-gp, namun protein PTEN tidak terekspresi pada sel MCF-7/Dox. Penelitian ini berhasil membuktikan bahwa hsa-miR-21 mengalami overekspresi pada sel MCF-7/Dox sebagai model resistensi kanker payudara. Overekspresi hsa-miR-21 diduga kuat berperan dalam represi atau inhibisi translasi protein melalui mekanisme regulasi pascatranskripsi mRNA gen PTEN sehingga protein PTEN tidak terekspresi pada sel MCF-7/Dox. Breast cancer (BC) is epithelial carcinoma with high incidency and mortality worldwide, including in Indonesia. In clinical case, most of the patient having chemotherapy on the late stadium of BC which is costly ineffective and contribute to resistance. At molecular level, it is proposed that microRNA-21 (hsa-miR-21) plays important role in post transcriptional regulation by repressing or inhibiting translation of mRNA tumor suppressor gene Phosphatase Tensin Homolog (PTEN). Decreasing or loss of PTEN expression, which is code by PTEN gene, has impact on increased cell proliferation and aberrant apoptosis pathway which lead to tumorigenesis and chemotherapy resistance. To understand these molecular mechanism, in vitro experiment conducted in Doxorubicinresistant MCF-7 cell line to investigate the expression of hsa-miR-21 and PTEN. Interaction between hsa-miR-21 and mRNA PTEN were predicted by in silico analysis based on bioinformatics database. Parental MCF-7 cell culture has periodically Doxorubicin exposured to obtain resistant MCF-7 cell line with IC50 value determined by MTT assay. P-gp and PTEN protein expression were analyzed by immunocytochemistry using monoclonal antibody. qRT-PCR was performed to detect the expression of hsa-miR-21 as well as U6SnRNA served as reference gene and UniSp6 as positive control. Results showed that Doxorubicin resistant MCF-7 cell line had increased IC50 value (5,78 I¼g/ml) compared to parental MCF-7 (0,68 I¼g/ml) with resistance index value 8,5. Based on qRT-PCR analysis, hsa-miR-21 was overexpressed in MCF-7/Dox cell line with fold change expression 7,94. The upregulation of hsa-miR-21 in MCF-7/Dox cell line was concurrent with downregulation of PTEN protein. It was shown that P-gp expression was detected while PTEN found underexpression on MCF-7/Dox cell line. This study revealed that hsa-miR-21 was overexpressed in MCF-7/Dox cell line as Doxorubicin-resistance model in breast cancer. This finding suggest that overexpresion of hsa-miR-21 play important role in post transcriptional regulation by repressing or inhibiting translation of PTEN mRNA which affect the underexpression of PTEN protein in MCF-7/Dox breast cancer cell line.

New approaches to prevent and treat colon cancer has developed to more selective. Cell cycle and angiogenesis play an important role in tumor progression and metastasis then those contribute to patient prognosis.Active compound isolated from the leaves of T. diversifolia (TD) is most selective on WiDr cell line with IC50 = 0.585 ug/ml and selectivity index = 69, 015. Objective: To know the antiproliferative and antiangiogenesis potency of the active compound isolated from the leaves of TD through observation on WiDr cell cycle and VEGF expression. Methods: Isolation of TD is performed by preparative TLC method. WiDr cell cycle is analyzed by flow cytometry using propidium iodide (PI). Analysis of antiangiogenesis with immunocytochemistry method using anti-VEGF human monoclonal antibody. Results : Active compound isolated from the leaves of TD at 4 µg/mL inhibit the cell cycle at S (24,36, and 72 hours) and G1 phase (48 hours). It dose at 8 µg/ml inhibit at G2/M (24 hours), subG1 (36 and 48 hours), and S phase (72 hours). The expression of VEGF is significantly depleted from 6,29% (negative control group) to become 3,31% at 4 µg/mL. Conclusion: Active compound isolated from the leaves of TD at 4 µg/mL and 8 µg/ml inhibit the cell cycle in nonspecific phase and at 4 µg/ml deplete the VEGF expression on human colorectal cancer WiDr cells.

Perubahan iklim juga mempengaruhi penurunan imunitas sehingga akan mudah terserang penyakit. Terdapat beberapa tumbuhan herbal yang terbukti berpotensi sebagai imunomodulator. Desa Tanjungkulon Kajen, memiliki banyak tumbuhan yang dapat digunakan sebagai imunomodulator. Penelitian ini bertujuan untuk mengetahui tingkat pengetahuan masyarakat tentang tumbuhan herbal sebagai imunomodulator dengan menggunakan metode diskriptif kuantitatif. Populasi dalam penelitian ini adalah masyarakat Desa Tanjungkulon, Kecamatan Kajen, Kabupaten Pekalongan. Sampel yang diambil memenuhi kriteria inklusi dan eksklusi berjumlah 97 orang dengan teknik purposive sampling. Instrumen penelitian menggunakan kuisioner dari peneliti sebelumnya yang sudah tervalidasi. Hasil data yang diperoleh menunjukkan bahwa tingkat pengetahuan masyarakat Desa Tanjungkulon tentang tumbuhan herbal sebagai imunomodulator sebanyak 70 %.

Serabi is Indonesian traditional food and has been widely known in Surakarta. However, its sensor... more Serabi is Indonesian traditional food and has been widely known in Surakarta. However, its sensory profile has not yet been described. This study aims to characterize the sensory attributes of serabi products in Surakarta by combining quantitative descriptive analysis (QDA) and principal component analysis (PCA) methods. Ten samples of serabi produced by small medium enterprises (SMEs) in Surakarta were collected and evaluated. The sensory profile is categorized based on aroma, taste and texture. The focus group discussion (FGD) of eleven trained panellist agreed that the attributes of aroma are eggs, coconut milk, sweet, burnt and pandan leaf. Meanwhile the attributes of taste are coconut milk, sweet and salty, and softness, solid and crunchy particle, hardness, softness and creamy are the attributes of texture. The PCA result showed that two samples, J and I, were described as serabi that have intense burnt and hardness attributes while more shredded coconut texture were represented by sample H and A. Moreover, samples D, G, F fall in a quadrant that describes having dominant egg and flour taste, while the rest samples, E, C and B have more intense salty, sweet and pandan leaf aroma. This result can be used as a reference for describing the basic profile of traditional serabis in Surakarta.

This study evaluated differences in the clinical appearance of patients with hepatocellular carci... more This study evaluated differences in the clinical appearance of patients with hepatocellular carcinoma (HCC) based on plasma level and regulation of microRNAs (miRNA-29c, miRNA-21, and miRNA-155). The observational-analytical study with a cross-sectional design was conducted on 36 HCC patients and 36 healthy controls. The blood samples were collected from 2 Province Hospitals (Dr. Sardjito Hospital and Prof. Dr. Margono Soekarjo Hospital) for HCC and the Blood Bank Donor of the Indonesian Red Cross for 36 healthy controls. These blood samples were treated as follows: plasma isolation, RNA isolation, cDNA synthesis, quantification by qRT-PCR using a sequence-specific forward primer, and normalization of miRNA using housekeeping-stably miRNA-16. There were only 27 HCC patients with complete clinical variables (neutrophil to lymphocyte ratio (NLR), platelet to lymphocyte ratio (PLR), platelet count, albumin, C-reactive protein (CRP), and cholinesterase (ChE)) that were able to analyses for regulation miRNAs based on its fold change expression miRNA target. Background and aim: As a result of its rapid spread in various countries around the world, on Mar... more Background and aim: As a result of its rapid spread in various countries around the world, on March 11, 2020, WHO issued an announcement of the change in coronavirus disease 2019 status from epidemic to pandemic disease. The virus that causes this disease is indicated originating from animals traded in a live animal market in Wuhan, China. Severe Acute Respiratory Syndrome Coronavirus 2 can attack lung cells because there are many conserved receptor entries, namely Angiotensin Converting Enzyme-2. The presence of this virus in host cells will initiate various protective responses leading to pneumonia and Acute Respiratory Distress Syndrome. This review aimed to provide an overview related to this virus and examine the body's responses and possible therapies. Method: We searched PubMed databases for Severe Acute Respiratory Syndrome Coronavirus-2, Middle East respiratory syndrome-related coronavirus and Severe Acute Respiratory Syndrome Coronavirus. Full texts were retrieved, analyzed and developed into an easy-to-understand review. Results: We provide a complete review related to structure, origin, and how the body responds to this virus infection and explain the possibility of an immune system over-reaction or cytokine storm. We also include an explanation of how this virus creates modes of avoidance to evade immune system attacks. We further explain the therapeutic approaches that can be taken in the treatment and prevention of this viral infection. Conclusion: In summary, based on the structural and immune-evasion system of coronavirus, we suggest several approaches to treat the disease.

Objective: We determine the utility of Prostate Specific Antigen (PSA) for predicting the presenc... more Objective: We determine the utility of Prostate Specific Antigen (PSA) for predicting the presence of skeletal metastasis on Bone Scan (BS) in prostate cancer patients. Material & Methods: Retrospective analysis of medical records of 70 consecutive prostate cancer patients subjected to bone scan during the last 2 years was done. 5 cases were excluded due to the following reasons: Serum PSA not available, hormonal or other therapy given prior to serum PSA measurement, and/or Bone Scan, and symptomatic for bone metastasis. In remaining 65 cases, PSA value and bone scan were evaluated. Results: BS was found to be positive in 20/65 (31%) and negative in 45(69%) patients. 24 (37%) had serum PSA>100 ng/ml, 25 (38.5%) had PSA of 20-100 ng/ml and only 16 (24.5%) had PSA< 20 ng/ml. Conclusion: Serum PSA<20 ng/ml have high predictive value in ruling out skeletal metastasis. Our data are in corroboration with results from previous studies that BS should be performed only if PSA>20 ng/ml. Using this cutoff , unnecessary investigation can be avoided. Avoiding BS asymptomatic in this group of patients would translate into a significant cost-saving and reduction in their psychological and physical burden.

Digging and dissecting the formulation of predetermined problems using three theories are: (1) theory of existence, (2) perception theory, and (3) theory of communication barriers. This study uses research methods which include: research approaches using qualitative methods. The research location is in the District of Selatan Denpasar. Types and sources of data used are types of qualitative data, while data sources used are primary and secondary data. The technique of determining the informants used is purvitative sampling. Data collection techniques include: observation techniques, interviews, literature, and documentation. The results of the analysis obtained from this study are 1) the existence of Hinduism extensionists. In this task, the Hinduism Extension has 5 (five) functions, namely, administrative, educative, facilitative, consultative, and transformative. Potential This fostered area is classified as a densely populated urban area and a high level of religious heterogeneity. The need for worship facilities and infrastructure is sufficient. The number of Hinduism Religious instructors in the City of Selatan Denpasar is as many as 2 Hindu Religion Extension Servants PNS and 16 Non-PNS Hindu Religion Extension. 2) Community perception is categorized into, a). Perceptions of community leaders, b). Stakeholder Perception, c). sekaa perception, d). widyasabha perception. Perception is seen as the most important component in the delivery of information. Through the public perception can be realized, can understand about the condition of the community in 10 villages / villages in Selatan Denpasar. The community can reveal how the performance of the Hindu Religious Extension in each of the target areas 3) internal and external constraints that occur when coaching Hinduism.
