Triclosan (TCS) is present in toothpaste and other cosmetic products as an antibacterial and anti... more Triclosan (TCS) is present in toothpaste and other cosmetic products as an antibacterial and antifungal agent. This manuscript highlights that TCS is a potential oxidative stress-causing agent, an estrogenic, mutagenic, cancer-causing agent, and genotoxic agent present in cosmetic products. This study also summarizes the therapeutic approach to overcome all of the harmful effects. It is a popular current topic, and a new research study is needed to find a new alternative as an antibacterial and anti-fungal agent instead of TCS. TCS causes oxidative stress when the dynamic balance in synthesizing and removing reactive oxygen species (ROS) within typical physiological circumstances is disturbed. The antioxidant defence system includes both enzymatic and non-enzymatic antioxidants produced by the organism to tackle the harmful effects of ROS. TCSs have estrogenic, proliferative, and apoptotic properties due to research on cell fate. The mutagenic potential of TCS has been examined using in vitro and in vivo research in prokaryotic, eukaryotic systems, and mammalian cells. It also induces carcinogenic, estrogenic, and mutagenic effects. TCS's therapeutic effects, especially against inflammatory skin conditions, have been demonstrated by many materials gathered from in vitro and in vivo experiments. The new findings suggest that TCS, a commonly used cosmetic product, may cause cancer, as shown by animal and human models and clinical trials. TCS is not effectively regulated, as evidenced by its presence in various environmental media, human bodies, and animals. Its irresponsible usage and disposal may endanger humans and wildlife. TCS has been found to damage a wide variety of cells in cell-based investigations. TCS's exact function in the environmental selection of antibiotic and multidrug resistance genes is still unclear. Comprehensive evaluations of these domains, especially to derive serious human health risk inferences from TCS outcomes, may help future research and legislation to better serve the public's health.

Apart from its natural sweetening property, honey also possesses therapeutic role in the treatment of various health ailments. Among these, chemoprotective properties are more evident. Honey is thought to have chemopreventive effect against different types of cancers since long time. In fact, tt has been widely used for its antioxidant activity, anti-apoptotic, anti-proliferative, anti- anti-mutagenic, inflammatory and immunomodulatory activities and as a Tumor Necrosis Factor (TNF). The role of honey in each of these activities is well established and is used extensively worldwide in different settings of the health care.

Improved dietary products such as milk and its products with antioxidant properties and functional foods of animal origin have been utilized to prevent chronic diseases. The designer milk contains low fat and less lactose, more protein, modified level of fatty acids, and desired amino acid profiles. The importance of milk and its products is due to the presence of protein, bioactive peptides, conjugated linoleic acid, omega-3 fatty acid, Vitamin D, selenium, and calcium. These constituents are present in milk product, play a key role in the physiological activities in human bodies, and act as anti-inflammatory, anti-tumor, antioxidant, hypocholesterolemic, immune boosting, and antimicrobial activities. Consumer awareness regarding benefits of designer foods such as milk and its products is almost non-existent worldwide and needs to be established to reach the benefits of designer food technologies in the near future. The main objective of this review was to collect data on the antioxidant properties of milk and its constituents which keep milk-derived products safe and preserved.

The central question addressed is the relationship between "normal" aging and agerelated pathologies such as osteoarthritis, cardiovascular diseases, emphysema, malignant tumors and cognitive decline, dementias. The mechanisms recognized as most important in cell and tissue aging are briefly outlined. Emphasis is laid on the importance of post-synthetic modifications of the macromolecules of the extracellular matrix and on cell matrix interactions. Loss of intercellular communication and cell-matrix interactions as a result of receptor decay and receptor uncoupling were recently recognized as key events. Unavoidable polypathology at advanced age may be the answer to the above question.

Abstract Natural products are the chemical compounds or substances produced naturally by a living... more Abstract Natural products are the chemical compounds or substances produced naturally by a living organism via synthetic process and/or semisynthetic that are present in the nature (includes small molecules and complex structure such as urea and Taxol, respectively). Phytochemicals are originated from Greek word “Phyto” means “Plant” and are basically chemical compounds produced via primary or secondary metabolism which have role in biological activities (growth and defense mechanisms against competitor, pathogen, or predators). Phytochemicals are plant-origin chemicals and some of these are required as essential nutrients, having a key role in the maintenance of the healthy status of organisms. Information on the health effects of natural products including phytochemicals is often obtained from animal experiments, cell and tissue culture, and human data. Some of the most important natural products obtained from plants, so-called phytochemicals, are terpenes, alkaloids, and phenolics such as tannins and flavonoids. Natural products can be extracted from matrices by different methods including solvent extraction, solid-phase extraction (SPE) method, supercritical fluid extraction (SFE), microwave-assisted extraction (MAE), ultrasonic-assisted extraction (UAE), pulsed electric field (PEF) extraction, enzyme-assisted extraction (EAE), and pressurized liquid extraction (PLE). The advanced and modern chromatographic techniques are used to isolate and purify natural products. Different procedures of spectroscopy like ultraviolet-visible (UV) spectroscopy, mass spectrometry (MS), nuclear magnetic resonance (NMR) spectroscopy, and infrared (IR) spectroscopy are used to determine the structure of molecules. A number of natural products, especially phytochemicals, have demonstrated significant biological properties like antiinflammatory and antioxidant activity, allowing to decrease the risk of cardiovascular disease, anticancer, type 2 diabetes, cortical and subcapsular cataract, and progression in Alzheimer’s disease. The purpose of this chapter is to give an overview of the analysis techniques of natural products, with special emphasis on phytochemicals.

Biochemical evidence on the potential role of methyl mercury in hepatic glucose metabolism through inflammatory signaling and free radical pathways

Methylmercury (MeHg) is an extremely important environmental toxicant posing serious health risks... more Methylmercury (MeHg) is an extremely important environmental toxicant posing serious health risks to human health and a big source of environmental pollutant. Numerous evidence available showing a link between nervous system toxicity and MeHg exposure. Other forms of mercury are reason of metabolic toxic effects and alteration of DNA in the human body. The sources of exposure could be occupational or other environmental settings. In the present study MeHg was orally gavaged to mice, at doses of 2.5, 5, and 10 mg/kg for 4 weeks. Fasting hyperglycemia, activity of hepatic phoshphoenolpyruvate carboxykinase and glucose 6-phoshphate were reported high as compared to control group. Inflammatory markers like, tumor necrosis factor α, the actual end product of inflammatory mediators' cascade pathway was also raised in comparison to control group. Hyperinsulinemia observed in serum showed clear understanding of mercury induced insulin resistance. Moreover, tissue damage due to increased oxidative stress markers like, hepatic lipid peroxidation, 8-deoxygunosine, reactive oxygen species, and carbonyl groups was significantly higher as compared to control group. MeHg caused a significant reduction in antioxidant markers like ferric reducing antioxidant power and total thiol molecules. The present study highlighted that activity of key enzymes involved in glucose metabolism is changed, owing to MeHg induced toxicity in the liver. Induction of similar toxic effects assumed to be stimulated by the production of high quantity free radicals.

Ochratoxin A (OTA) is a naturally occurring mycotoxin mostly found in food items including grains... more Ochratoxin A (OTA) is a naturally occurring mycotoxin mostly found in food items including grains and coffee beans. It induces DNA single-strand breaks and has been considered to be carcinogenic. It is recognized as a serious threat to reproductive health both in males and females. OTA is highly nephrotoxic and carcinogenic, and its potency changes evidently between species and sexes. There is a close association between OTA, mutagenicity, carcinogenicity, and genotoxicity, but the underlying mechanisms are not clear. Reports regarding genotoxic effects in relation to OTA which leads to the induction of DNA adduct formation, protein synthesis inhibition, perturbation of cellular energy production, initiation of oxidative stress, induction of apoptosis, influences on mitosis, induction of cell cycle arrest, and interference with cytokine pathways. All these mechanisms are associated with nephrotoxicity, hepatotoxicity, teratotoxicity, immunological toxicity, and neurotoxicity. OTA administration activates various mechanisms such as p38 MAPK, JNKs, and ERKs dysfunctions, BDNF disruption, TH overexpression, caspase-3 and 9 activation, and ERK-1/2 phosphorylation which ultimately lead to Alzheimer disease (AD) progression. The current review will focus on OTA in terms of recent discoveries in the field of molecular biology. The main aim is to investigate the underlying mechanisms of OTA in regard to genotoxicity and epigenetic modulations that lead to AD. Also, we will highlight the strategies for the purpose of attenuating the hazards posed by OTA exposure.
