Penelitian ini dilaksanakan di Laboratorium Kultur Jaringan Kebun Raya Cibodas – Badan Riset dan Inovasi Nasional (BRIN) pada bulan Agustus sampai bulan November 2021. Metode penelitian yang digunakan adalah metode eksperimental menggunakan Rancangan Acak Lengkap (RAL) dengan sembilan kombinasi perlakuan 2,4-D (Dichlorophenoxyatic Acid) and Benzyl Adenine (BA) dengan tiga ulangan. Exotic plant invasion is one of the major causes of species extinction. In many contexts, mountai... more Exotic plant invasion is one of the major causes of species extinction. In many contexts, mountainous forests are the last refuge for native species. There are several inventory studies conducted in mountainous and tropical forests in Indonesia. However, there are no studies yet on the factors that explain the abundance and richness of surveyed naturalized alien species. This study investigated whether the number of individuals and abundance-weighted species richness (AWR) of naturalized alien plant species in the hiking-trail of Mount Gede-Pangrango National Park (MGPNP) forest area correlated with leaf traits (specific leaf area (SLA) and leaf thickness) and environmental factors (elevation, slope, and normalized difference vegetation index (NDVI)). We showed that leaf thickness and habitat elevation explained the AWR variations of naturalized alien species. We did not detect any important effect of leaf traits and environmental factors on the number of individuals per exotic species per plot. The influence of leaf thickness and habitat elevation indicates the important role of both biotic and abiotic factors on exotic species to develop a high species richness and become an invasive species in the tropical mountain forest ecosystem.

We had examined the seed variation of Rubus spp. collected in Cibodas Botanical Garden. The observation was conducted on 12 different species of Cibodas Botanical Garden’s collections. The variation of seed’s size was observed based on length, width, and sculpture of seed’s surface area. The results showed that R. pyrifolius seed was larger among others species, and the smaller one was R. fraxinifolius. Moreover, based on seed morphology, three morphological types of seed coats were identified, which are seeds with prominent reticulate sculpture and sharp back ridge (R. accuminatissimus, R. alpestris, R. ellipticus, R. lineatus, R. niveus, R. pyrifolius, R. rosifolius); seeds with prominent reticulate sculpture and broad back ridge (R. alceifolius, R. moluccanus, R. glomeratus); seeds with least prominent sculpture and blunt back ridge (R. chrysophyllus and R. fraxinifolius).

Percobaan ini ditujukan untuk mengetahui metode sterilisasi yang tepat pada eksplan R. rosifolius. Castanopsis argentea (Blume) A.DC., commonly known as saninten, is an endangered evergreen tree s... more Castanopsis argentea (Blume) A.DC., commonly known as saninten, is an endangered evergreen tree species native to Sumatra and Java Islands, Indonesia. This study was carried out to assess the genetic diversity of C. argentea in Mount Gede Pangrango National Park (MGPNP) based on random amplified polymorphic DNA markers. The samples of C. argentea were collected from six locations in MGPNP. The results showed that from 19 selected random primers used, 13 primers, i.e. OPA-4, OPA-7, OPA-9, OPB-1, OPB-7, OPB-8, OPC-5, OPC-8, OPE-1, OPE-4, OPE-6, OPE-7, and OPE-8 gave reproducible amplification banding patterns. The polymorphic information content was highest for the primer OPE-6 (0.49) followed by the primers OPB-7 (0.41), OPC-8 (0.41), OPE-4 (0.38), and OPE-7 (0.38). The similarity coefficient varied from 0.57 to 0.92, indicated a high level of genetic variation among the genotypes. The cluster analysis showed that the species of C. argentea in MGPNP were clustered into two groups: the first cluster consisted of samples from six locations (Bodogol, Tapos, Cisarua, Cibodas, Tegalega, and Selabintana) and the second consisted of samples from two locations (Selabintana and Cisarua). Considering the conservation status and genetic diversity of C. argentea, it is necessary to implement conservation strategies such as conserve the habitat and propagation using resources from several locations.

In order to cultivate loquat, the availability of high-quality seedlings is becoming an essential factor. This research was aimed to determine the response of loquat’s seed, stem, and shoot cutting on various types of medium. The seed, stem, and shoots cutting of loquat were collected from North Sumatra. Three types of medium, i.e., humus + roasted rice husk (1:1), raw rice husk + sand (1:1), and humus + raw rice husk (1:1), were used during the experiment in Cibodas Botanical Garden. The results show that the highest percentage of seed germination on the 10th week after the plantation was humus + roasted rice husk (91.18%) and followed by humus + raw rice husk (77.78%) and raw rice husk + sand (69.44%). Botanical garden routinely engaged in conservation, research, education, environmental services and ecotourism activities. Cacti and succulent are common ornamental plant which are introduced to CBG. This study aimed to re- inventory cacti and succulent collections in Cibodas Botanical Garden, also to determine its conservation status based on IUCN redlist. In order to manage cacti and succulent collections, CBG was established two green houses, one for cacti collections and the other one for succulent collections. Currently, CBG hold approximately 2.291 specimens of cacti and succulent. It consists of 17 family, 81 genus, and 206 species. Among those collections, there are nine species from Cactaceae, one species from Xanthorrhoeaceae, and one species from Asparagaceae are listed in IUCN redlist. Those species are Aloe jucunda, Cleistocactus winteri, Dracaena draco, Echinocactus grusonii, Eriosyce chilensis, Mammillaria parkinsonii, Mammillaria petrophila, Parodia concinna, Parodia magnifica, Rhipsalis dissimilis, and Turbinicarpus alonsoi.

Propagation is an important factor in developing a fruit cultivation. In vitro micropropagation is an important instrument to improve the quality of seedling. Our experiment was aimed to study the micropropagation of R. chrysophyllus and R. fraxinifolius. The shoot tips were cultured on MS medium supplemented with 10 mg/L indole-3-butyric acid (IBA) for enhancing roots of R. chrysophyllus and R. fraxinifolius in the in vitro condition. Then, the rooted plantlets were transplanted into cocopeat media for acclimatization. The results showed that R. chrysophyllus and R. fraxinifolius gave a different response on the medium contain 10 mg/L of IBA. The R. fraxinifolius shows the best results compared to R. chrysophyllus on 14 days after subculture. The roots length and roots numbers of R. fraxinifolius and R. chrysophyllus were 9.13 cm and 11.25; 2.20 cm and 2.00, respectively. Although R. fraxinifolius was growing better than R. chrysophyllus, but after acclimatization R. chrysophyllus were able to grow faster than R. fraxinifolius. Moreover, on the parameters of plant height shows that R. chrysophyllus (9.20 cm) were growth higher than R. fraxinifolius (4.05 cm) during acclimatization.

Flowers and flower buds were produced every month by Rubus spp., in different amount except Rubus pyrifolius. Furthermore, each species had a different response to the environment factors. Correlation analysis shows that maximum temperature and relative humidity influenced flower and fruit production. Rubus fraxinifolius and Rubus lineatus had more economic value due to its ability to produce fruit throughout the year.

It has a high potency to be cultivated plant. Although, some of the potential of Rubus has already known, information of about fruit nutrients of species that distribute in the mountain forest of Indonesia are not known yet. This study was aimed to determine the fruit nutrients of five species of wild raspberries that was collected in Cibodas Botanical Garden. The results of our experiments demonstrate that the fruits nutrients contents varied among five species of wild raspberries from Indonesian mountain's forests. Rubus fraxinifolius has the highest content of sugar (5.05 g sugar per 100 g fruits) compared to Rubus rosifolius, Rubus chrysophyllus, Rubus lineatus and raspberry. During the ripening and ripe period, there were a different value of vitamin C and iron contents. The highest vitamin C of R. fraxinifolius (83.65 mg/100 gram) on the ripening fruit stage II. On the other hand, the highest vitamin C of R. rosifolius (54.30 mg/100 gram) found on the stage of ripe fruit.

This experiment aimed to study the changes on the leaf traits of loquat seedling that treated with plant growth regulators (PGRs). Three types of PGRs, auxin (naphthalene acetic acid/NAA), gibberellin (gibberellic acid/GA3) and cytokinin (benzylaminopurine/BA) with four doses (0, 25, 50, 100 ppm) were sprayed onto the leaves of loquat seedling. We observed nine parameters, PGRs treatments were significantly affecting eight parameters, while there were one parameter is not significantly affected. In order to cultivate loquat, the availability of high-quality seedlings is becoming an essential factor. This research was aimed to determine the response of loquat’s seed, stem, and shoot cutting on various types of medium. The seed, stem, and shoots cutting of loquat were collected from North Sumatra. Three types of medium, i.e., humus + roasted rice husk (1:1), raw rice husk + sand (1:1), and humus + raw rice husk (1:1), were used during the experiment in Cibodas Botanical Garden. The results show that the highest percentage of seed germination on the 10th week after the plantation was humus + roasted rice husk (91.18%) and followed by humus + raw rice husk (77.78%) and raw rice husk + sand (69.44%). Botanical garden routinely engaged in conservation, research, education, environmental services and ecotourism activities. Cacti and succulent are common ornamental plant which are introduced to CBG. This study aimed to re- inventory cacti and succulent collections in Cibodas Botanical Garden, also to determine its conservation status based on IUCN redlist. In order to manage cacti and succulent collections, CBG was established two green houses, one for cacti collections and the other one for succulent collections. Currently, CBG hold approximately 2.291 specimens of cacti and succulent. It consists of 17 family, 81 genus, and 206 species. Among those collections, there are nine species from Cactaceae, one species from Xanthorrhoeaceae, and one species from Asparagaceae are listed in IUCN redlist. Those species are Aloe jucunda, Cleistocactus winteri, Dracaena draco, Echinocactus grusonii, Eriosyce chilensis, Mammillaria parkinsonii, Mammillaria petrophila, Parodia concinna, Parodia magnifica, Rhipsalis dissimilis, and Turbinicarpus alonsoi.

Propagation is an important factor in developing a fruit cultivation. In vitro micropropagation is an important instrument to improve the quality of seedling. Our experiment was aimed to study the micropropagation of R. chrysophyllus and R. fraxinifolius. The shoot tips were cultured on MS medium supplemented with 10 mg/L indole-3-butyric acid (IBA) for enhancing roots of R. chrysophyllus and R. fraxinifolius in the in vitro condition. Then, the rooted plantlets were transplanted into cocopeat media for acclimatization. The results showed that R. chrysophyllus and R. fraxinifolius gave a different response on the medium contain 10 mg/L of IBA. The R. fraxinifolius shows the best results compared to R. chrysophyllus on 14 days after subculture. The roots length and roots numbers of R. fraxinifolius and R. chrysophyllus were 9.13 cm and 11.25; 2.20 cm and 2.00, respectively. Although R. fraxinifolius was growing better than R. chrysophyllus, but after acclimatization R. chrysophyllus were able to grow faster than R. fraxinifolius. Moreover, on the parameters of plant height shows that R. chrysophyllus (9.20 cm) were growth higher than R. fraxinifolius (4.05 cm) during acclimatization.
Flowering and fruiting phenology were studied in five species of Rubus spp (Rubus ellipticus, Rub... more Flowering and fruiting phenology were studied in five species of Rubus spp (Rubus ellipticus, Rubus fraxinifolius, Rubus lineatus, Rubus moluccanus, and Rubus pyrifolius) collected from Cibodas Botanical Garden from April 2009 to March 2010. Flowers and flower buds were produced every month by Rubus spp., in different amount except Rubus pyrifolius. Furthermore, each species had a different response to the environment factors. Correlation analysis shows that maximum temperature and relative humidity influenced flower and fruit production. Rubus fraxinifolius and Rubus lineatus had more economic value due to its ability to produce fruit throughout the year.

It has a high potency to be cultivated plant. Although, some of the potential of Rubus has already known, information of about fruit nutrients of species that distribute in the mountain forest of Indonesia are not known yet. This study was aimed to determine the fruit nutrients of five species of wild raspberries that was collected in Cibodas Botanical Garden. The results of our experiments demonstrate that the fruits nutrients contents varied among five species of wild raspberries from Indonesian mountain's forests. Rubus fraxinifolius has the highest content of sugar (5.05 g sugar per 100 g fruits) compared to Rubus rosifolius, Rubus chrysophyllus, Rubus lineatus and raspberry. During the ripening and ripe period, there were a different value of vitamin C and iron contents. The highest vitamin C of R. fraxinifolius (83.65 mg/100 gram) on the ripening fruit stage II. On the other hand, the highest vitamin C of R. rosifolius (54.30 mg/100 gram) found on the stage of ripe fruit.

The objective of this study was to evaluate agronomic characters especially the size of pods and seeds of peanut lines in subspecies (ssp.) hypogaea and F 5 families derived from crossing between NC 7 and Kelinci. In this study, two experiments were reported. The first experiment evaluated 10 lines of ssp. hypogaea using completely randomized block design with six replications. The second experiment evaluated 22 F 5 families derived from crosses between NC 7 and Kelinci using completely randomized block design with three replications. Nine of ten lines ssp. hypogaea evaluated in Experiment I showed at least one superior trait in pod or seed size. The result showed that early Bunch was superior in 10-seed length, 100-seed weight, the number of mature pods, and pod weight per pod. NC 7 was superior in 10-pod width and 100-seed weight. Therefore, this research aimed to determine the effect of seed size on the germination and seedling growth of loquat. This research was compiled using a Randomized Complete Design with four different seed groups as treatments. The loquat seeds were obtained from Danau Toba region and grouped based on the weight of seeds, i.e. Grade I (<0.6 g), Grade II (0.6 - <0.9 g), Grade III (0.9 - 1.2 g) and Grade IV (> 1.2 g). This experiment aimed to study the changes on the leaf traits of loquat seedling that treated with plant growth regulators (PGRs). Three types of PGRs, auxin (naphthalene acetic acid/NAA), gibberellin (gibberellic acid/GA3) and cytokinin (benzylaminopurine/BA) with four doses (0, 25, 50, 100 ppm) were sprayed onto the leaves of loquat seedling. We observed nine parameters, PGRs treatments were significantly affecting eight parameters, while there were one parameter is not significantly affected. The results showed that either in mature or young leaves, PGRs treatments were significantly affecting in eight parameters the growth and development of leaves, such as leaf surface area, specific leaf area, fresh and dry weight leaf, water content, number of stomata, size of stomata, chlorophyll and transpiration rate compared to control.
