Purpose:To report the utilization of diagnostic intracytoplasmic sperm injection (D-ICSI), an ICS... more Purpose:To report the utilization of diagnostic intracytoplasmic sperm injection (D-ICSI), an ICSI cycle performed in the natural cycle, to obtain information about embryo development potential after sperm injection into zona pellucida (ZP)-free oocytes.Materials and Methods:We report the case of a couple with primary unexplained infertility with a history of previous failed, in vitro fertilization intracytoplasmic sperm injection (IVF-ICSI) cycles characterized by the presence of ZP-free oocytes. Whole exome sequencing (WES) was carried out to analyse the possible genetic basis of oocyte abnormality.Results:Diagnostic ICSI provided information about the embryo development potential from ZP-free oocytes and allowed better planning of the subsequent ICSI cycle. WES revealed that the absence of ZP was likely to be due to a new (ZP1) mutation. Evidence for association of either smoking or Schistosoma infection with carcinogenicity and/or mutagenicity has been reported. This study deals with the induction of chromosomal damage assessed by bone marrow micronucleus (MN) and chromosome aberration (CA) tests, as well as the analysis of chromosomal aberrations in diakinesis-metaphase I spermatocytes in male mice infected with Schistosoma mansoni cercariae and treated with nicotine. Schistosoma infection increased the percentage of micronucleated erythrocytes and total CAs in bone marrow cells. Analysis of meiotic chromosomes showed significant elevation in the Schistosoma-infected mice. Administration of nicotine to infected mice substantially increased the percentages of micronucleated cells and total CAs. The percentage of chromosomal abnormalities in spermatocyte metaphase-I cells increased significantly in Schistosoma-infected mice. The incidence of chromosomal aberrations of metaphase I spermatocytes showed a highly significant increase after nicotine treatment; the effect on infected mice was significantly higher than that noted on the non-infected group. The data obtained in this study suggest that animals afflicted with schistosomiasis with synergistic nicotine administration suffer from a higher genetic risk, and may serve as a warning to schistosomiasis patients.

Hepatitis B virus infection (HBV) is a non-cytopathic virus; therefore, liver damage leading to f... more Hepatitis B virus infection (HBV) is a non-cytopathic virus; therefore, liver damage leading to fibrosis, cirrhosis and hepatocellular carcinoma (HCC) is considered to be the direct result of the immune response to viral infection. Cytokines play critical roles in the pathogenesis of HBV. The balance between Th1/Th2 has a great impact on the outcome of HBV infection. Interleukein (IL-and transforming growth factor (TGFΞ²) are important regulators of the immune response due to their anti-inflammatory action on the immune system. Given the importance of these cytokines in the complicated immune response against HBV, it is advantageous to assess their levels in the plasma of HBV patients and comparing them with normal healthy controls. Measuring IL-10 and TGF-Ξ² levels were achieved by enzyme-linked immunororbent assay (ELISA) in 118 Egyptian HBV patients and 119 normal healthy controls. Our results showed a significant (p<0.001) sharp reduction in the level of IL-10 in comparison to controls. Hepatitis B was significantly correlated with reduction in plasma IL-10 level (r=-0.864; p<0.001). On the other hand there, results presented here indicated that there is a significantly elevated serum level of TGF-Ξ² (p<0.001) compared with that of controls. A direct correlation was reported between TGF-Ξ² and HBV infection (r= 0.157, p<0.05). Accordingly, high levels of TGF-Ξ² may be a mechanism by which immune response against HBV is suppressed. In conclusion, HBV might be associated with reduction in IL-10 coincides with elevation in TGF-Ξ². Our results give a shed of light about the importance in balance between different cytokines in pathogenesis of HBV and its role in suppressing the immune system during viral infection.

Both 2 nd and 4 th larval instars of S. littoralis were exposed to the different concentrations of flufenoxuron and SpliNPV. The biochemical effect of IGR and SpliNPV on both 2 nd and 4 th larvae instar show changes in protein electrophoretic pattern in the current study reveals differences between the untreated samples and treated one, some proteins were missed or expressed at different stages which may be responsible for all the obtained deformation. The effect of these compounds on biochemical activities were revealed after studying electrophoretic protein (fractionated protein) using PAGE in order to determine their effects on the vital system of S. littoralis, which may indicate the formation of new proteins responsible for stimulating the immune system of the insect as a result of entering foreign objects inside the body of the insect.

Embryo's quality is closely linked to miRNA regulation, which plays a critical role in gene expre... more Embryo's quality is closely linked to miRNA regulation, which plays a critical role in gene expression and protein synthesis. In this prospective cohort observational study, extracellular miRNAs were tested using the miRCURY Locked Nucleic Acid (LNA) system, which used culture media from twenty-two singly transferred embryos. Study samples were collected on day three and embryos were transferred on day five. Based on earlier publications, hsa-miR-29c-3p and hsa-miR-30c-5p, among other miRNAs (not published), were chosen and assessed. The negative-implantation group had higher expression of hsa-miR-30c-5p regarded as statistically significant (P.value <0.001) than the other tested miRNAs. Contrary, there was no significant link between pregnancy outcome and expressed hsa-miR-29c-3p. Study findings, in comparison with findings from other reports, reveal the absence of hsa-miR-29c-3p and hsa-miR30c-5p from day 3 implanting embryos in contrast with day 5 where they are abundant, suggesting specificity to the embryo's development from cleavage to morula, blastocyst, and implantation stages. These findings could open the gate toward potential biomarkers for embryo development and implantation outcome.

Purpose The application of natural products as well as biological control-specific bioagents, esp... more Purpose The application of natural products as well as biological control-specific bioagents, especially the nucleopolyhedrovirus, is considered a very important tool to ensure that pest control does not affect the environment. Methods Chemical constituents screening and determination of pharmacopoeial constants, and minerals and trace elements of the brown algae Sargassum asperifolium , Sargassum dentifolium , and Sargassum linifolium from the Red Sea (Hurghada, Egypt) were carried out. Results and conclusion It was found that Sargassum spp. have a high ash content and were positive for subliminal matter, volatile constituents, carbohydrate content, sterols, and/or triterpenes. In addition, they had high Ca, K, Mg, and Fe contents. The protein content as well as the amino acid composition of the three algae species mentioned were assessed using the Kjeldahl method and an amino acid analyzer, respectively. The hydroalcoholic extracts (70%) of Sargassum spp. under investigation were examined for insecticidal activities in vitro on Spodoptera littoralis (Sl52 cells) and Spodoptera frugiperda (Sf9 cells) and in vivo on S. littoralis nucleopolyhedrovirus replication. The results showed that the three algae species tested had various insecticidal and antiviral activities.

The strains were sampled from natural infestation of cotton field in Kafr El-Sheikh Governorate in may 1997. Electron microscopy observations revealed the presence of icosahedral particles with 22 nm in diameter for the viral capside related to the Densovirus (DNV), and 29 nm for the second one as a Picornavirus (PV). For the DNV four capsid proteins of 91, 63, 53 and 47 KDa have been separated, and were undifferentiated from that of the unique published Densovirus isolate from Egypt, the Mythimna lofeyi DNV(M/DNV). A complete replication of the Spodoptera littoralis granulosis virus (SpliGV) was obtained, in vi... more A complete replication of the Spodoptera littoralis granulosis virus (SpliGV) was obtained, in vitro by both virus infection and DNA transfection in the ORS-Pop-95 (Pop-95) cell line established from embryonic cells of the potato tuber moth, Phthorimaea operculella. SpliGV multiplied significantly during several passages in Pop-95 cells at 19 degrees C. When the cells were infected and kept at 19 degrees C for the first 4 hrs and then at 27 degrees C for the rest of the experiment (20 days), the viral multiplication proceeded at the same rate. Comparison of SpliGV progenies, multiplied either in vivo or in vitro, using electron microscopy and restriction profile analysis, showed their identity.

To date, the control of this pest has been mainly based on the use of insecticides. However, this control method can result in environmental, social and economic problems. To improve management options against the weevil, the efficacy of the entomopathogenic nematode Steinernema carpocapsae Weiser (Nematoda: Steinernematidae) and the potential of a strain of the entomopathogenic fungus Beauveria bassiana (Ascomycota: Clavicipitaceae) were evaluated in laboratory, semi-field and field assays. The use of these entomopathogenic microorganisms resulted highly efficient against R. ferrugineus.

The mitochondrial DNA viability ratio is significantly higher in embryos that implant. The score might help to select euploid embryos for single embryo transfer. Embryo euploidy is a critical factor for successful pregnancy outcomes. However, transfer of euploid embryos does not invariably result in implantation, thus indicating that other factors may play a role. Metabolic rates and adenosine triphosphate content vary significantly in oocytes and embryos and might affect embryo viability. CONCLUSIONS: Nickel disrupted blastocyst hatching in a dose-dependent manner. Cleavage stage embryo development was not impacted by exposure to Nickel at concentrations below 100mM. Down-regulation of pluripotent genes and repression of trophectoderm differentiation may mediate the detrimental effects of Nickel on embryo development. This work was aimed to evaluate a simplified semi-synthetic diet based on combination of starch/a... more This work was aimed to evaluate a simplified semi-synthetic diet based on combination of starch/agar for mass rearing of Spodoptera littoralis. Two modified semi-synthetic diets (A & B) proved successful in maintaining the culture of S. littoralis for five successive generations without apparent adverse effects throughout the insect stages. The mean values of tested biological parameters were determined on the reared insects fed on either of the modified diets. The results showed a higher larval weight, pupal weight, adult emergence, fertility and development index as compared to the agar-based standard diet of Shorey and Hale (1965). Whereas, percentage pupation, survival, fecundity and sex ratio were non-significantly different in the tested treatments. Moreover, the cost of ingredient reduced by 45.6% and 33.3% per one liter of diets A and B, respectively as compared to the agar-based standard diet. A Phthorimaea operculella cell line was used to study the cytotoxic effects of the entomopathogen... more A Phthorimaea operculella cell line was used to study the cytotoxic effects of the entomopathogenic fungi Metahrizium anisopliae, Beauveria bassiana and Verticillium lecanii. P. operculella. Cell treatment with fungi crude filtrate (in shake culture and solid medium) showed a clear cellular cytopathic effect. Cells treated with M.anisopliae crude filtrate revealed granular contents, irregular forms and alterations on the cellular membrane. Treated cells of P.operculella with B. bassiana and V. lecanii crude filtrates showed elongation in cell shape. P.opercullela treated cells with sonicated conidospores filtrate of each of the three fungi isolates showed irregular forms and intensive vacuoles. However, treated cells with filtrated and then sonicated blastosopres of the fungi showed associated crystals in the culture medium and fibroplast elongated cells filled with intensive vacuoles.
