The human iPSCs and their differentiated progenies provide a unique opportunity to identify these miRNA-mediated regulatory mechanisms. To identify miRNA–mRNA regulatory interactions in human nervous system development, well characterized NSCs were differentiated from six validated iPSC lines and analyzed for differentially expressed (DE) miRNome and transcriptome by RNA sequencing. Following the criteria, moderated t statistics, FDR-corrected p-value ≤ 0.05 and fold change—absolute (FC-abs) ≥2.0, 51 miRNAs and 4033 mRNAs were found to be significantly DE between iPSCs and NSCs. Devanagari script is being used in various languages, in south Asian subcontinent, such as Sanskr... more Devanagari script is being used in various languages, in south Asian subcontinent, such as Sanskrit, Rajasthani, Marathi and Nepali and it is also the script of Hindi, the mother tongue of majority of Indians. Recognition of handwritten characters of Devanagari alphabet set is an important area of research. The work done for the recognition of Devanagari handwritten script is negligible in literature despite it is being used by millions people in India and abroad and it has numerous applications. The feature extraction method(s) used to recognize hand-printed characters play an important role in ICR applications. There are many feature extraction methods available in literature. We have tested the recognition performance of about 5 feature extraction methods available in literature on Devanagari handwritten characters. A database of more than 25000 handwritten Devanagari characters is developed by collecting the samples from hundreds writers belonging to 43 Devanagari alphabets. The performance comparisons have been made using two classifiers MLP and SVM.

Nitric acid treated single and multi wall carbon nanotubes (SWNT and MWNT) have been dispersed in... more Nitric acid treated single and multi wall carbon nanotubes (SWNT and MWNT) have been dispersed in polypropylene using maleic anhydride grafted polypropylene (MA-g-PP) and butanol/xylene solvent mixture. SWNT exfoliation was characterized by Raman and UVe viseNIR spectroscopies. Evidence for hydrogen bonding between maleic anhydride grafted polypropylene and nitric acid treated nanotubes was obtained using infrared spectroscopy. Polypropylene/carbon nanotube composites were melt-spun into fibers. Dynamic mechanical studies show that for fibers containing 0.1 wt% SWNT, storage modulus increased by 5 GPa at À140 C and by about 1 GPa at 100 C, suggesting temperature dependent interfacial strength. The crystallization behavior has been monitored using differential scanning calorimetry and optical microscopy. Control fibers exhibited 27% shrinkage at 160 C, while the shrinkage in the composite fibers was less than 5%. Fibers heat-treated to 170 C show very narrow polypropylene melting peak (peak width about 1 C).

Characterization of Drosophila mutants for the InsP 3 R has demonstrated that InsP 3-mediated Ca 2+ release is required in Drosophila larvae for growth and viability. Methodology/Principal Findings: To understand the molecular basis of these growth defects a genome wide microarray analysis has been carried out with larval RNA obtained from a strong InsP 3 R mutant combination in which 1504 independent genes were differentially regulated with a log 2 of fold change of 1 or more and P,0.05. This was followed by similar transcript analyses from InsP 3 R mutants where growth defects were either suppressed by introduction of a dominant suppressor or rescued by ectopic expression of an InsP 3 R transgene in the Drosophila insulin like peptide-2 (Dilp2) producing cells. Conclusions/Significance: These studies show that expression of transcripts related to carbohydrate and amine metabolism is altered in InsP 3 receptor mutant larvae. Moreover, from a comparative analysis of genes that are regulated in the suppressed and rescued conditions with the mutant condition, it appears that the organism could use different combinations of pathways to restore a 'normal' growth state.

The interfacial morphology of carbon nanotube fiberepolypropylene matrix is investigated by means of polarized optical microscopy (POM), wide-angle X-ray diffraction (WAXD), scanning electron microscopy (SEM) and transmission electron microscopy (TEM). The supramolecular microstructures of polypropylene transcrystals induced by the nanotube fiber are observed in the range of isothermal crystallization temperatures from 118 C to 132 C. The dynamic process of transcrystallization is analyzed by using the theory of heterogeneous nucleation. Microstructure analysis shows that the nanotubes can nucleate the growth of both aand g-transcrystal, and a-transcrystals dominate the overall interfacial morphology. Close to the nanotube fiber surface, a cross-hatched lamellar microstructure composed of mother lamellae and daughter lamellae is observed.

The present/current diagnostic kits and methods to diagnose BTV are laborious, time consuming and expensive. In the present study, we have attempted to develop a novel approach to detect BTV antibodies in sera that in future can be harnessed for developing a pen side diagnostic test. Briefly, we identified the immunodominant regions of the VP7 protein of BTV and synthesized them in the multiple antigenic peptide (MAP) format with cysteine at C-terminal of the lysine mosaic, which elicited highly ordered conformation as well as ELISA reactivity. Finally, we coated the MAP peptides on the gold nanoparticles that can be used to detect BTV specific antibodies in the sera using a spot test.

Differentiating wild-type VZV strains from the Oka vaccine strain can be achieved only by molecular genotyping methods. The development of PCR methods for VZV strain differentiation has been hampered by the fact that the VZV genome is highly conserved. We used VZV ORF 62 PCR-RFLP analysis to identify and differentiate wild-type VZV strains in India from the Oka vaccine strain. Digestion of VZV ORF 62 amplicons with SmaI, enabled accurate strain differentiation; the Oka strain was positive for three SmaI sites, compared to two SmaI sites in the wild-type VZV strains that we tested.

SEVA TB ES-31 antigen has shown potential in detecting tuberculous IgG antibody in earlier studies from our laboratory. In the present study we have analysedSEVA TB ES-31 antigen specific immunoglobulinsIgM, IgA and IgG in clinically and bacteriologically confirmed pulmonary tuberculosis cases to determine the usefulness of specific immunoglobulin class in the diagnosis of patients attending the hospital.Of the 30 cases of pulmonary tuberculosis 25 (83.3%) were positive for IgG, 19 (63.3%) for IgM and 16 (53.3%) for IgA. On combining IgG and IgM positivity, sensitivity was increased to 93.3%. While combining IgG and IgA positivity, sensitivity increased to 90%. However specificity was decreased to 66.6% and 70% for both of these combinations respectively. It could be envisaged from this study that IgG antibody detection against ES-31 antigen showed acceptable sensitivity (83.3%) and specificity (86.6%) compared to IgM or IgA alone or in combination. When immune responses were analysed according to degree of sputum positivity, IgG response was observed to be predominant in all grades, compared to IgM or IgA antibody. The addition of IgM or IgA as an adjunct test increases the sensitivity but at the cost of specificity. Hence the detection of IgG alone is more useful compared to IgM or IgA assay, in detecting tuberculosis disease cases coming to the hospital.

Anin vitro released 31 kDa antigen ESAS-7F isolated fromM.tb H(37)Ra culture filtrate by salt precipitation, SDS-PAGE and cation exchange fast protein liquid chromatography (FPLC) was shown earlier to be a diagnostically important antigen fraction. In this report, we describe the isolation of ESAS-7F antigen using monospecific antibody coupled to sepharose CL-4B column. The percentage recovery of ESAS-7F antigen using affinity chromatography was approximately 8% of the total ES antigen proteins compared to 0.05% obtained by conventional purification steps using salt precipitation, SDS-PAGE and FPLC. Similar seroreactivity was observed by the antigen isolated by both the methods in indirect ELISA. Affinity chromatography helped in an increased recovery of ESAS-7F antigen and obviates the need for time consuming conventional purification steps.
