We have shown previously that citP is included in the citQRP operon, which is mainly transcribed from the P1 promoter in L. lactis subsp. lactis biovar diacetylactis. Furthermore, transcription of citQRP and citrate transport are not induced by the presence of citrate in the growth medium. In this work, we analyzed the influence of the extracellular pH on the expression of citP . The citrate transport system is induced by natural acidification of the medium during cell growth and by a shift to media buffered at acidic pHs. This inducible response to acid stress takes place at the transcriptional level and seems to be due to increased utilization of the P1 promoter. Plasmid pCIT264 from Luctococcus lactis subsp. lactis biovar diacetylactis (L. diacetylactis) con... more Plasmid pCIT264 from Luctococcus lactis subsp. lactis biovar diacetylactis (L. diacetylactis) contains an insertion sequence (IS&like element located in the citrate utilization (citQRP) cluster. This 967-nucleotide long element is bounded by 17 bp perfect inverted repeats and contains an open reading frame (ORFl) composed of 296 codons, which could encode a transposase. Expression of the IS from pCIT264 generates two mRNAs of 2900 and 1900 nucleotides. The transcription is driven by the P3 promoter, composed of a-10 region located at the right end of the IS and of a-35 region positioned downstream of this element. The IS-like element (IS982) is present in seven copies in the L. diacetylactis genome. The copy present in pCIT264 is highly stable and does not promote rearrangements of the tit cluster. We suggest that the stable maintenance of the IS-like element in pCIT264 could be due to a translational control of the putative transposase by an antisense RNA.

This block accounts for 30% of the gas production for the entire country. An operator faced a unique challenge to reduce well construction costs for this well while drilling through a harsh Carboniferous formations sequence. The Carboniferous interval is extremely hard and abrasive with unconfined compressive strength (UCS) values ranging from 12,000 psi to 24,000 psi, averaging 20,000 psi with 32,000-psi peaks. This interval is a complex drilling environment consisting of interbedded, conglomeratic, hard, and abrasive lithologies that presents several drilling obstacles, such as formation heterogeneity and multiple geological uncertainties. Esta tendencia ha operado de forma muy positiva en el comportamiento de los resultados empresariales durante la crisis economica padecida en Espana, sobre todo gracias al aumento de la exposicion a paises emergentes. Historically woman's' health during pregnancy is in hands of doctors. Through monthly clinical vi... more Historically woman's' health during pregnancy is in hands of doctors. Through monthly clinical visits women are informed about their health. But not all woman have the opportunity to on-site clinical visits or/and woman need constantly support in this hard and unknown journey. This lack of support is worldwide rising the maternal-fetal morbidity and mortality. This paper shows a study towards the design of PREGLOG, a mobile application purely thought for pregnant women to help them overcome any situation which could complicate their pregnancy process, including anomaly logs and useful information about the symptoms they could present during their current pregnancy month. We interviewed specialists in charge of maternal control of women, including three gynecologists and a specialist in Family Medicine, and 10 pregnancies in order to understand pregnancy care and pregnancy doubts, fears, and needs. As a result we obtained valuable insights that will help us to design a prototype which fulfills the main needs for both the patients and the specialists who treat them.

Two main points were targeted as fundamental to accomplish this objective: reduce the number of runs to drill the Carboniferous section and improve borehole enlargement efficiency. A multidisciplinary group comprising a service company and Repsol team members conducted a detailed and thorough investigation of previous failure modes to identify an integrated approach to improve the performance. Along with offset history, the team was able to identify lithology characteristics to accurately diagnose the underlying root causes. Cyanobacteria are key microbes in topsoil communities that have important roles in preventing soi... more Cyanobacteria are key microbes in topsoil communities that have important roles in preventing soil erosion, carbon and nitrogen fixation, and influencing soil hydrology. However, little is known regarding the identity and distribution of the microbial components in the photosynthetic assemblages that form a cohesive biological soil crust (biocrust) in drylands of Europe. In this study, we investigated the cyanobacterial species colonizing biocrusts in three representative dryland ecosystems from the most arid region in Europe (SE Spain) that are characterized by different soil conditions. Isolated cyanobacterial cultures were identified by a polyphasic approach, including 16S rRNA gene sequencing, phylogenetic relationship determination, and morphological and ecological habitat assessments. El presente trabajo tiene como propósito presentar 3 casos clínicos pediátricos, diagnosticados d... more El presente trabajo tiene como propósito presentar 3 casos clínicos pediátricos, diagnosticados de toxocariasis, vistos en Lima-Perú, entre los años 1987 y 1989. Los tres pacientes son menores de 5 años de edad, con antecedente de comer tierra, que presentaron leucocitosis leve o severa con hipereosinofilia (1500 eosinofilos por mm3). El primer paciente tuvo además hipergammaglobulinemia y gran compromiso del estado general, requiriendo el uso de tiabendazol. El segundo paciente presentó dolor abdominal crónico y la biopsia hepática reveló granulomas con abundantes eosinófilos. El tercer paciente tuvo dolor articular crónico de rodilla derecha con sinovitis en los dedos de las manos y pies. Los tres pacientes tuvieron títulos en sangre para toxocariasis en la prueba de ELISA a 1/4096, (CDC de Atlanta). Se trata del primer reporte clínico de larva migrans visceral en el Perú.

A methodology for entropy production minimization in design of extractive diabatic distillation s... more A methodology for entropy production minimization in design of extractive diabatic distillation systems is proposed. The feasibility of the design is obtained by the boundary value method used by Doherty and coworkers, and the optimal design is attained using a genetic algorithm, this strategy allows determine the optimal heat load distribution, and the flows and temperatures for hot and cold utilities. For optimal design of diabatic distillation columns it was employed a multilevel strategy, the first level solves the optimal adiabatic design and the second one determines the optimal heat load distribution in the sequential heat exchangers (SHE), in both levels the feasibility of the designs is forced by a penalty in the objective function, it considers the separation feasibility and the energy balance consistency. The ethanol dehydration by extractive distillation with ethylene glycol is assessed in order to show our optimal design procedure and to compare the diabatic distillation process with the adiabatic one. Results show that this strategy allows to solve the problem, and that with SHE, the entropy production can be reduced expressively: 14% for the recovery solvent column and 16% for the extractive one, compared to the adiabatic columns.

When cultures were transferred from 37° C to 20° C, a chloramphenicol‐ and rifampicin‐sensitive synthesis of a C‐16 unsaturated fatty acid was observed. Synthesis of this compound reached a plateau after 5 h at 20° C, reaching levels of 20% of the total fatty acid content. [14C]‐labelled fatty acids attached as thioesters to acyl‐carriers compounds, such as coenzyme A (CoA) or acyl‐carrier protein (ACP) synthesized de novo by glycerol‐requiring auxotrophs deprived of glycerol to arrest phospholipid synthesis, could not be desaturated at 20° C. Desaturation of these fatty acids was readily observed when glycerol was restored to the cultures allowing resumption of transfer of acyl‐moieties from acyl‐thioesters to phospholipid. The method we describe here uses a deleted Mu phage (a mini-Mu) to transpose E. coli genes into cosmids during mini-Mu replication. The resulting cosmids clones are packaged invivo into 2 phage particles. Plasmids carrying a particular DNA sequence can be selectively recovered after infection of a new host with the in vivo constructed genomic cosmid library. This system was used succesfully to clone several E. coli genes.

The ethanol dehydration by extractive distillation with ethylene glycol is assessed in order to show our optimal design procedure and to compare the diabatic distillation process with the adiabatic one. Results show that this strategy allows to solve the problem, and that with SHE, the entropy production can be reduced expressively: 14% for the recovery solvent column and 16% for the extractive one, compared to the adiabatic columns.

When cultures were transferred from 37° C to 20° C, a chloramphenicol‐ and rifampicin‐sensitive synthesis of a C‐16 unsaturated fatty acid was observed. Synthesis of this compound reached a plateau after 5 h at 20° C, reaching levels of 20% of the total fatty acid content. [14C]‐labelled fatty acids attached as thioesters to acyl‐carriers compounds, such as coenzyme A (CoA) or acyl‐carrier protein (ACP) synthesized de novo by glycerol‐requiring auxotrophs deprived of glycerol to arrest phospholipid synthesis, could not be desaturated at 20° C. Desaturation of these fatty acids was readily observed when glycerol was restored to the cultures allowing resumption of transfer of acyl‐moieties from acyl‐thioesters to phospholipid. The enzymatic activity of one of these protein kinases, KinA, was found to be inhibited in vitro by certain fatty acids. The most potent inhibitors have at least one unsaturated double bond in the cis configuration and a chain length of 16–20 carbon atoms. Homologous isomers with a trans double bond are not inhibitory. Saturated straight‐ or branched‐chain fatty acids are either much weaker inhibitors or have no effect. The inhibitors prevent autophosphorylation of KinA and are non‐competitive with ATP. B. subtilis phospholipids were found to contain at least one as yet unidentified type of fatty acid that, when present in an unesterified form, inhibited KinA. T... more A general in vivo procedure for cloning Escherichia coli genes into cosmids has been developed. The method we describe here uses a deleted Mu phage (a mini-Mu) to transpose E. coli genes into cosmids during mini-Mu replication. The resulting cosmids clones are packaged invivo into 2 phage particles. Plasmids carrying a particular DNA sequence can be selectively recovered after infection of a new host with the in vivo constructed genomic cosmid library. This system was used succesfully to clone several E. coli genes.
