In this review, we summarize the technologies and medicinal chemistry strategies for hit identification, hit-to-lead and lead-to-candidate optimization, and current challenges in PB2 inhibitor development, as well as offering insights for the fight against drug resistance.

METHODS DIBC specimens were manufactured and randomly assigned to the control groups and the DIBC groups. According to the corresponding ASTM/ISO standards, the static biomechanical strength and the fatigue limit were tested systematically. The particle size distribution of diphosphonate powder was analyzed with the laser light scattering method. The fatigue test results, given as number of cycles-to-failure, were analyzed using the linearized format of the two-parameter Weibull function. RESULTS With the drug load increased, there was a slight increase in static biomechanical strength and a moderate decrease in fatigue limit, both with statistical significance. When immersed in PBS before the tests, the DIBC specimens presented an overall significant decrease of static biomechanical strength and fatigue limit. OBJECTIVE To describe the characteristics of oldest-old Chinese with disability and their adult-c... more OBJECTIVE To describe the characteristics of oldest-old Chinese with disability and their adult-child caregivers, and the extent to which these characteristics were associated with caregiver burden. METHODS The study was based on 168 pairs of disabled oldest-old adults and their adult-child caregivers, derived from the Chinese Longitudinal Healthy Longevity Survey. Descriptive analyses of care recipients' and caregivers' characteristics were conducted respectively, in reference to caregiver burden. Statistically significant characteristics identified in these bivariate analyses were then jointly evaluated in multiple linear regression models with caregiver burden as the outcome. RESULTS Care recipients positive emotion status [(β = -0.227 (-0.412, -0.042)], multiple chronic disease [(β = 0.513 (0.081, 0.945)], and caregivers spent more caregiving time [(β = 0.225 (0.061, 0.389)] were main factors associated with caregiver burden. CONCLUSION Adult-children caregivers perceived heavier burden if care recipients had low positive emotions, had multiple chronic diseases, and caregivers spent more time caregiving.

These reactions proceed well under mild conditions without any additives. Plausible mechanisms for such a polycyclic ring system assembly were also proposed. Moreover, benzo[f]imidazo[2,1-a][2,7]naphthyridine 3g displayed a fluorescence effect, demonstrating the potential applications in organic optical materials.

Hydrogel‐based soft electronics (HSE) is promising as implantable devices due to the similarity o... more Hydrogel‐based soft electronics (HSE) is promising as implantable devices due to the similarity of hydrogel substrates to biologic tissues. Most existing HSE devices are based on conducting hydrogels that usually have weak mechanical properties, low conductivity, and poor patternability. Reported here is an HSE with good mechanical performance, high sensitivity, and versatile functions by stencil printing of liquid metal on a tough hydrogel, facilitating integration of multiple sensing units. Self‐shaping ability is imparted to the HSE by creating gradient structure in the hydrogel substrate. The resultant HSE actively deforms into 3D configurations with zero or nonzero Gaussian curvature to fix on objects or organs with sophisticated geometries and maintains the sensing functions. The versatilities and potential applications of this HSE are demonstrated by monitoring motions of a rice field eel and beatings of a rabbit heart. Such HSE based on morphing substrate should pave the way for implantable electronics with better fixation and interfacial contact with the organs. The concept of morphing hydrogel devices can be extended to other soft electronics with responsive polymer films or elastomers as the substrates.

The aim of this study was to investigate the associations between serum 25-hydroxyvitamin D and disability in activities of daily living (ADL), mobility, and objective physical functioning among Chinese older adults. METHODS We used cross-sectional data of 2225 older adults (≥65 years) who participated in the 2011/2012 main survey of the Chinese Longitudinal Healthy Longevity Survey (CLHLS) and the 2012 biomarker sub-study. Serum levels of 25-hydroxyvitamin D were measured by enzyme immunoassay. Outcomes included ADL disability, mobility disability, and disability in three objective physical examinations (standing-up from a chair, picking-up a book from the floor, and turning-around 360°). RESULTS The multiple regression models suggested that participants in the serum 25-hydroxyvitamin D deficiency group had higher odds of ADL disability (OR: 4.08; 95% confidence interval (CI): 2.81, 5.92), mobility disability (OR: 1.59; 95% CI: 1.05, 2.41), and disability in standing-up from a chair (OR: 2.43; 95% CI: 1.60, 3.69), picking-up a book from the floor (OR: 3.09; 95% CI: 2.07, 4.60), and turning-around 360° (OR: 3.09; 95% CI: 2.07, 4.60). Subgroup analyses revealed that some of the above associations (particularly those with mobility disability and disability in turning-around 360°) were only statistically significant among the oldest-old. CONCLUSIONS Among the oldest-old in China, serum 25-hydroxyvitamin D deficiency was associated with disability in ADL, mobility, and objective physical functioning, highlighting the importance of managing the serum 25-hydroxyvitamin D level to promote healthy aging.

Long lasting antimicrobial activity and low toxicity are essentials for hydrogels in biomedicine.... more Long lasting antimicrobial activity and low toxicity are essentials for hydrogels in biomedicine. However, most reported hydrogels cannot combine these characteristics. In this work, poly (hexamethylene guanidine) hydrochloride (PHMG), a cheap cationic polymer with two terminal amino groups, was first modified with methacrylic anhydride to give PHMG dimethacrylamide (PHMGDMAAm), which was further used to prepare hydrogels with acrylamide (AAm) under ultraviolet irradiation in the presence of aketoglutaric acid (a-KGA) as photoinitiator in aqueous medium. The resultant hydrogels showed strong antibacterial activity against both Gram-negative and Gram-positive bacteria due to the PHMG segments in the hydrogel backbone. Moreover, the antimicrobial activity of the hydrogels did not decrease significantly after being soaked in water for one month and washed by water frequently for many times. Hemolysis and cytotoxicity assays demonstrated the excellent biocompatibility of the PHMG-PAAm hydrogels. This kind of low cost cationic hydrogels with long lasting antimicrobial activity and low toxicity is expected to have potential applications in biomedicine. V
