Our objective was to evaluate the effect of sonography-guided percutaneous microwave ablation of ... more Our objective was to evaluate the effect of sonography-guided percutaneous microwave ablation of high-grade dysplastic nodules in the cirrhotic liver. From July 1997 to May 2003, 49 histologically proven high-grade dysplastic nodules in 30 patients with liver cirrhosis were treated by microwave ablation. Three patients had concomitant small hepatocellular carcinomas (D < 3.0 cm), whereas another three had undergone liver segmentectomy for hepatocellular carcinoma 1 year earlier. The mean size of the nodules was 1.8 cm (range, 0.9-4.6 cm). Sixty-eight insertions with 78 applications were administered to the 49 nodules. The follow-up period was 12-82 months (mean, 45.1 +/- 19.0 months). Five patients died during this study: three from advanced hepatocellular carcinoma, one from bleeding in the upper gastrointestinal tract, and another from cerebral hemorrhage. All nodules showed decreased density on unenhanced CT and no enhancement on contrast-enhanced CT after microwave ablation. Posttreatment biopsy performed in 16 patients with 18 nodules 1-3 months after microwave ablation showed no evidence of viable tissue but replacement by fibrotic tissue in all nodules. Percutaneous microwave ablation as a minimally invasive therapy is effective for ablating high-grade dysplastic nodules, thus preventing their potential malignant transformation, which may improve survival. The preliminary data warrant further prospective, randomized studies.

Wikipedia, a killer application in Web 2.0, has embraced the power of collaborative editing to ha... more Wikipedia, a killer application in Web 2.0, has embraced the power of collaborative editing to harness collective intelligence. It can also serve as an ideal Semantic Web data source due to its abundance, influence, high quality and well-structuring. However, the heavy burden of up-building and maintain- ing such an enormous and ever-growing online encyclopedic knowledge base still rests on a very small group of people. Many casual users may still feel dif- ficulties in writing high quality Wikipedia articles. In this paper, we use RDF graphs to model the key elements in Wikipedia authoring, and propose an inte- grated solution to make Wikipedia authoring easier based on RDF graph match- ing, expecting making more Wikipedians. Our solution facilitates semantics reuse and provides users with: 1) a link suggestion module that suggests and au- to-completes internal links between Wikipedia articles for the user; 2) a catego- ry suggestion module that helps the user place her articles in correct categories. A prototype system is implemented and experimental results show significant improvements over existing solutions to link and category suggestion tasks. The proposed enhancements can be applied to attract more contributors and relieve the burden of professional editors, thus enhancing the current Wikipedia to make it an even better Semantic Web data source.

These all-female species can propagate by asexual reproduction modes, such as gynogenesis and hybridogenesis. However, the coexistence of sexuality and asexuality in a single species was recently noted only in a cyprinid fish silver crucian carp, Carassius auratus gibelio. This fish had been demonstrated to be capable of gynogenesis stimulated by sperm from other related species. To define the role of C-CAM in prostatic tumorigenesis, a tumorigenic human prostatic cancer cell line, PC-3, was transfected with an expression plasmid containing C-CAM1 (a C-CAM isoform). Transfected clones showed significantly lower growth rates, reduced anchorage-independent growth, and less tumorigenicity in vivo than control cells. Furthermore, transfection of an antisense vector into a nontumorigenic prostatic epithelial cell line, NbE, resulted in tumor formation in nude mice. Sublines derived from these NbE-induced tumors had lower levels of C-CAM than did control cells. These data suggest that C-CAM1 can function as a tumor suppressor in prostate tumorigenesis.

We recently purified and cloned a new prophenoloxidase-activating proteinase (PAP-2) from hemolymph of the tobacco hornworm Manduca sexta [J. Biol. Chem. 278, 3552-3561]. As the terminal component of a putative serine proteinase cascade, this enzyme activates prophenoloxidase (proPO) via limited proteolysis. To purify and study the activating proteinase for PAP-2 from this insect, we expressed the zymogen of PAP-2 (proPAP-2) in insect cells infected by a recombinant baculovirus that harbors the cDNA. To facilitate the purification of proPAP-2, we modified a commercial vector (pFastBac1) by inserting a synthetic DNA fragment encoding a hexahistidine sequence, allowing fusion of the affinity tag to the carboxyl terminus of a protein. The NOSs are a family of complex enzymes that catalyze the five-electron oxidation of L-arginine ... more The NOSs are a family of complex enzymes that catalyze the five-electron oxidation of L-arginine to form NO and L-citrulline. They are best characterized as cytochrome P-450-like hemeproteins that depend on molecular oxygen, NADPH, flavins, and tetrahydrobiopterin. The three human NOS isoforms identified to date, ecNOS, nNOS, and iNOS, are found on human chromosomes 7, 12, and 17, respectively. Regulation of NO synthesis and release occurs at the levels of enzyme activity and mRNA synthesis. The nNOS mRNA is structurally diverse as a consequence of alternative promoters and alternate splicing. The iNOS gene is predominantly regulated at the level of transcription by synergistic combinations of proinflammatory cytokines and bacterial wall products. Changes in mRNA levels of the ecNOS following endothelium activation are mediated by altered rates of transcription as well as by the intriguing process of changes in mRNA stability. Given the essential role of the NO pathway in a wide variety of physiological and pathophysiological process, it is possible that the three isoforms of NOS contribute to polygenic genetic diversity in neurological, immune, and cardiovascular biology. Further studies are needed to determine the mechanisms of gene regulation of NOS in health and disease.

Retinoic acid-related orphan receptor gamma thymus, the master transcription factor of T helper 17 cell differentiation, is selectively antagonized by digoxin. However, the effect of antagonizing retinoic acid-related orphan receptor gamma thymus on AAA has not been investigated. We used human aortic sample analysis and 2 different experimental AAA models: (a) Angiotensin II (Ang II)-induced ApoE(-/-) male mice (Ang II/APOE model) and (b) porcine pancreatic elastase perfusion C57BL/6 mice (porcine pancreatic elastase/C57 model). Recent transcriptome studies suggest that long noncoding RNAs (lncRNAs) are key components of the... more Recent transcriptome studies suggest that long noncoding RNAs (lncRNAs) are key components of the mammalian genome, and their study has become a new frontier in biomedical research. In fact, lncRNAs in the mammalian genome were identified and studied at particular epigenetic loci, including imprinted loci and X-chromosome inactivation center, at least two decades ago-long before development of high throughput sequencing technology. Since then, researchers have found that lncRNAs play essential roles in various biological processes, mostly during development. Since much of our understanding of lncRNAs originates from our knowledge of these well-established lncRNAs, in this review we will focus on lncRNAs from the X-chromosome inactivation center and the Dlk1-Dio3 imprinted cluster as examples of lncRNA mechanisms functioning in the epigenetic regulation of mammalian genes.

USP7 is a very important protein with an important role in regulating the p53 pathway, which is critical for genomic stability and tumor suppression. We here genotyped eight SNPs within the USP7 gene and conducted a case-control study in 312 CRC patients and 270 healthy subjects in the Chinese Han population. No significant associations were found for any single SNP and CRC risk. Our data eliminate USP7 as a potential candidate gene towards for CRC in the Han Chinese population.

Retinoic acid-related orphan receptor gamma thymus, the master transcription factor of T helper 17 cell differentiation, is selectively antagonized by digoxin. However, the effect of antagonizing retinoic acid-related orphan receptor gamma thymus on AAA has not been investigated. We used human aortic sample analysis and 2 different experimental AAA models: (a) Angiotensin II (Ang II)-induced ApoE(-/-) male mice (Ang II/APOE model) and (b) porcine pancreatic elastase perfusion C57BL/6 mice (porcine pancreatic elastase/C57 model). Recent transcriptome studies suggest that long noncoding RNAs (lncRNAs) are key components of the... more Recent transcriptome studies suggest that long noncoding RNAs (lncRNAs) are key components of the mammalian genome, and their study has become a new frontier in biomedical research. In fact, lncRNAs in the mammalian genome were identified and studied at particular epigenetic loci, including imprinted loci and X-chromosome inactivation center, at least two decades ago-long before development of high throughput sequencing technology. Since then, researchers have found that lncRNAs play essential roles in various biological processes, mostly during development. Since much of our understanding of lncRNAs originates from our knowledge of these well-established lncRNAs, in this review we will focus on lncRNAs from the X-chromosome inactivation center and the Dlk1-Dio3 imprinted cluster as examples of lncRNA mechanisms functioning in the epigenetic regulation of mammalian genes.

Colorectal cancer (CRC), now the third most common cancer across the world, is known to aggregate... more Colorectal cancer (CRC), now the third most common cancer across the world, is known to aggregate in families. USP7 is a very important protein with an important role in regulating the p53 pathway, which is critical for genomic stability and tumor suppression. We here genotyped eight SNPs within the USP7 gene and conducted a case-control study in 312 CRC patients and 270 healthy subjects in the Chinese Han population. No significant associations were found for any single SNP and CRC risk. Our data eliminate USP7 as a potential candidate gene towards for CRC in the Han Chinese population.
