Pedersen. In this work, callus cultures of P. glomerata were established in order to study whether in vitro-cultured tissue could produce _-Ecdysone. To induce calli, nodal segments were cultured on Murashige and Skoog (MS) medium with various levels of 6-benzylaminopurine (BAP; 1.0, 5.0, or 10.0 ?M), _-naphthaleneacetic acid (NAA; 0.5, 1.0, 5.0, or 10.0 ?M) or 2,4-dichlorophenoxyacetic (2,4-D; 0.5, 1.0, 5.0, or 10.0 ?M). _-Ecdysone concentrations [on a (w/w) dry weight (DW) basis] in in vitro-cultured and field-grown tissues were measured by high performance liquid chromatography.The level of _-ecdysone was related to the friability and shoot regeneration capacity of the different calli. Organogenic, friable calli grown on media supplemented with 10.0 ?M BAP, plus either 5.0 ?M NAA or 0.5 ?M 2,4-D, resulted in the highest _-ecdysone concentrations [0.282% (w/w) and 0.211% (w/w), respectively]. In contrast, the _-ecdysone concentrations in field-grown plants were 0.194% (w/w) in roots and 0.333% (w/w) in the aerial parts. Low levels of _-ecdysone [0.007% (w/w)] were detected in plantlets cultured on hormone-free MS medium, indicating the importance of culture conditions for the in vitro production of _-ecdysone.This is the first report on the presence of ecdysteroids in calli of P. glomerata.

Fred Hough, oriundas do processo de multiplicação in vitro, foram inoculadas em meio MS e MS/2, testando-se os reguladores de crescimento: ácido indol-3-acético (AIA); ácido indolbutírico (AIB) e ácido naftaleno acético (ANA), nas concentrações de 0, 1, 3 e 5 miM com o objetivo de observar o efeito dessas auxinas sobre o enraizamento da cultivar. Foram acrescentadas aos meios as vitaminas MS mio-inositol (100 mg/L) e sacarose (30 g/L) em meio de ágar (6 g/L). O pH do meio foi ajustado para 5,8 e a cultura foi incubada a 25 ± 2º C e 16 horas de fotoperíodo a 2.000 lux, permanecendo por 30 dias. Due to the importance of sweet potato culture, this study aimed to optimize the production of the... more Due to the importance of sweet potato culture, this study aimed to optimize the production of the in vitro plants of different cultivars. Nodal segments of cultivars ‘Abóbora’, ‘Catarina’, ‘Biaze’, ‘Da Costa’, ‘Americana’ e ‘Morada Inta’ were cultured on Murashige and Skoog medium and they were evaluated for eight weeks in an in vitro culture. The cv. ‘Da Costa’ was also grown on a medium free of plant growth regulators, supplemented with benzylaminopurine (BA) and/or gibberellic acid (GA3) or in photoautotrophic culture system. It was found that the propagation rate is variable, depending on the cultivar and according to time of cultivation. The interval among subcultures was dependent on the rate of plant senescence. In cv. ‘Da Costa’, a higher production of plants was obtained on medium containing BAP or on sucrose-free medium and in bottles sealed with cotton. Plants originating from all cultivars showed a high survival rate during the acclimatization.

This species, native of southwest Africa, was accidentally introduced in Brazil in the 1950s and has since become a troublesome weed of natural pasture, due to its high spreading power. In order to assess lovegrass seeds behavior against different potentially stressful factors, we evaluated wide ranges of luminosity, temperature and osmotic potential on E. plana germination. After exposure to treatments, vigor and seed germination were analyzed through the following: germination speed index (GSI), first count and accumulated germination. All evaluated variables influenced E. plana germination, suggesting a relative susceptibility to environmental factors such as light, temperature, and water availability, with respect to in vitro germination. This information can be used as an aid to planning future strategies to control the spread of this invasive grass.

Thus, the objective of this work was to evaluate the efficiency of different substrates combined with base fertilization for Ilex paraguariensis seedlings, through morphophysiological parameters. The experiment was conducted in a completely randomized design, in a 5 x 3 factorial arrangement, and the treatments consisted of five substrate formulations combined with two fertilizers, and a control without fertilization, in November 2013. The I. paraguariensis seedlings produced in the substrate consisted of subsurface soil, cattle manure and charred rice hulls (2:2:1) presented the greatest heights (53.36 cm), stem diameter (6.80 mm), leaf area (692.52 cm2), root dry weight (6.2 g) and total dry weight (16.4 g). Nodal segments of two genotypes (BRA and JB-UFSM) of P. glomerata, originated from aseptically grown plants, were cultivated on hormone-free Murashige and Skoog medium. For the proliferation of P. tuberosa shoots, nodal segments, originated from aseptically grown plants, were either cultivated on hormone-free Murashige and Skoog (MS) medium or were supplemented with 1.0 µM thidiazuron (TDZ); the elongation and rooting of these plants were carried out on MS medium without TDZ. Plantlets of both species were acclimatized and transferred to field conditions. The β-ecdysone content in the plants was determined by high performance liquid chromatography. For the proliferation of P. tuberosa shoots, nodal segments, originated from aseptically grown plants, were either cultivated on hormone-free Murashige and Skoog (MS) medium or were supplemented with 1.0 µM thidiazuron (TDZ); the elongation and rooting of these plants were carried out on MS medium without TDZ. Plantlets of both species were acclimatized and transferred to field conditions. The β-ecdysone content in the plants was determined by high performance liquid chromatography.
