Aims: To determine the presence or absence of human papillomavirus (HPV) DNA in oral inverted duc... more Aims: To determine the presence or absence of human papillomavirus (HPV) DNA in oral inverted ductal papillomas (IDPs) using in situ hybridisation (ISH), and to analyse all cases for histological features of HPV infection. Methods: Six cases were retrieved from archival material and paraffin wax blocks were submitted for the detection of HPV DNA by means of ISH. A wide spectrum probe for HPV subtypes 6, 11, 16, 18, 31, 33, 45, 51, and 52 was used initially. Cases that were positive using this wide spectrum probe were further subtyped using HPV type specific probes (6/11, 16/18, and 31/33). The histological features of all tumours were analysed using routine microscopy. Results: Of the six cases of oral IDP identified, three were positive for HPV subtypes 6/11. All positive cases showed histological features of HPV infection (koilocytosis, papillomatosis, binucleated keratinocytes, and abnormal mitosis) in both the surface and the inverted epithelium. The three cases that tested negative for HPV DNA also exhibited focal histological features of HPV infection (two in the surface epithelium and one in the endophytic epithelium). Conclusions: These are the first documented cases of oral IDP to demonstrate positivity for HPV DNA and also to show histological features of HPV infection.

Normal keratinocytes plated on such a surface expressed p16, arrested growth, and exhibited enhanced directional motility. Two cell lines cultured from neoplastic epithelium, one p16-deleted and the other expressing mutant p16, became directionally motile when plated on such Lam5 surfaces. In contrast, precoating culture dishes with serum, which contains fibronectin, produced no effect. We conclude that contact with endogenous or exogenous Lam5 induces p16 expression and hypermotility in normal keratinocytes, but that p16 is not essential for the hypermotile phenotype. These results suggest that p16-Lam5g2 coexpression is an important feature of normal epidermal wound healing, causing growth arrest of the motile cells. Loss of p16 during neoplastic progression permits motile cells to continue proliferating, thereby resulting in invasive growth.

Histologically, all lesions were multicystic and 9 cases displayed a thin non-keratinized uniform lining composed of squamous and cuboidal/columnar epithelium. Other features noted were focal epithelial nodular swirls (60%), microcysts (40%), minimal inflammation (90%), and presence of mucus cells (40%). Because of IRB restrictions of the study, clinical behavior of these cysts could not be determined. At this time it is unclear if these cysts are a distinct clinico-pathologic entity or a developmental stage (forme-fruste) of other variants of odontogenic cysts such as glandular odontogenic cyst. Careful follow-up and more longitudinal studies are needed to answer this question.

This study was designed to study a group of healthy individuals to compare with pathological conditions of oral mucosa. In this ongoing study, mean of cytoplasmic and nuclear área were 2,001.61 ¼m 2 and 47.55 ¼m 2 concluding that oral keratinocytes show a large cytoplasmic área compared with nuclear área. It would be considered in future comparative studies.

A Novel Histopathologic Finding in a Radiopaque Maxillary Lesion of a Patient with C3 Glomerulonephritis: A Case Report

We present a case series of localized juvenile spongiotic gingival hyperplasias (LJSGH), a lesion... more We present a case series of localized juvenile spongiotic gingival hyperplasias (LJSGH), a lesion of unknown etiology that typically affects the anterior maxillary gingiva in children and young adults. It presents as a bright red, raised, welldemarcated lesion with a slight papillary or granular surface. Lesional epithelium is characteristically positive for CK19, a marker of junctional epithelium. Methods: Cases of LJSGH from 2008 to present were retrieved from the University of Pittsburgh Oral Pathology Biopsy Service archives and reviewed. Clinical and demographic data from these cases were collected. Results: Seventeen cases were identified. No significant sex predilection was noted (M:F ¼ 1.13:1). The age range was from 8 to 53 years (mean, 19.1 years). All cases were confined to the anterior portions of the jaws as single lesions. Sixteen (94.1%) cases occurred in the maxilla, with only one mandibular case (5.9%). The most common clinical impression was pyogenic granuloma (58.8%). In two patients, aged 53 and 50, CK19 staining was utilized to confirm the histopathological findings, and it was strongly positive throughout the full thickness of the lesional epithelium in both cases. Three cases (17.6%) presented as recurrences (range, 17 months to 10.5 years). The case that recurred after 10.5 years had identical histomorphology to the initial case that was signed out descriptively in 2005, prior to the characterization of LJSGH. Conclusion: As in other series, LJSGH in our experience is most commonly seen in the anterior maxilla of children and young adults. We report two cases of LJSGH in middle-aged adults and demonstrate the diagnostic utility of CK19 in unique cases. Thus, this diagnosis should be considered in middle-aged adults, despite its current name.

Cartilagenous Metaplasia in Odontogenic Keratocyst (Okc): Case Series and Immunohistochemical Analysis

It is important to recognize this feature to avoid misdiagnosis as a chondroid tumor. We describe the largest series of cases with the objective to quantify the clinicopathologic features and to elucidate the nature of the cartilage. Fourteen lesions in 13 patients (1 recurrence) of OKCs exhibiting cartilaginous metaplasia in the wall were identified between 1997 and 2016. Immunohistochemical (IHC) analysis was performed on two cases. Eight of 13 cases occurred in the maxilla with 4 occurring anteriorly and 2 involving the maxillary sinus. Five cases occurred in the mandible, 3 anteriorly, including the recurrence. The average age was 55 with a range of 16 to 86 years old. Ten of 13 cases occurred in males. All 3 female cases occurred in the anterior maxilla. The chondroid areas were frequently identified in zones of inflammation near the OKC lining. The proportion of cartilage to cyst was variable. IHC for p63, GFAP, EMA, CK5/6, and actin were performed on two cases to elucidate the nature of the chondroid material. CK5/6, EMA, and p63 positivity was localized to the epithelial lining. GFAP and actin were negative in the cartilaginous areas. Our findings suggest that compared with conventional OKCs, there is a slight increase in age and predilection for the anterior jaws when cartilaginous metaplasia is observed. GFAP negativity suggests that the chondroid material may not represent actual cartilage. Cartilaginous metaplasia in the walls of OKCs should not be mistaken for a cartilaginous neoplasm.
