Whether PVD is a predictor of adverse outcomes in patients undergoing percutaneous coronary intervention (PCI) in the drug eluting stent era is not known. Methods: Data was analysed from a total of 9110 patients from the Melbourne Intervention Group (MIG) database, a registry of patients undergoing PCI in Melbourne, Australia. Data was collected between April 2004 and July 2007 and analysed for baseline demographic data, traditional risk factors, procedural data and thirty day and twelve month major adverse cardiovascular events (MACE). Results: Of the 9110 patients undergoing PCI, 632(6.9%) had PVD. Patients with PVD had an average age of 70.6 years, were more frequently diabetic (41.3% vs. 22.6%, p<0.01), were hypertensive (79.7% v 63.1%, p<0.01), and had higher lipid levels (82.5% v 70.6%). We hypothesize that regulatory T-cell (Treg)-deficient strains have an altered TCR repertoire in ... more We hypothesize that regulatory T-cell (Treg)-deficient strains have an altered TCR repertoire in part due to the expansion of autoimmune repertoire by self-antigen. We compared the Vb family expression profile between B6 and Treg-lacking B6.Cg-Foxp3 sf/Y (B6.sf) mice using fluorescent anti-Vb mAbs and observed no changes. However, while the spectratypes of 20 Vb families among B6 mice were highly similar, the Vb family spectratypes of B6.sf mice were remarkably different from B6 mice and from each other. Significant spectratype changes in many Vb families were also observed in Treg-deficient IL-2 knockout (KO) and IL-2Ra KO mice. Such changes were not observed with anti-CD3 mAb-treated B6 mice or B6 CD4 1 CD25 2 T cells. TCR transgenic (OT-II.sf) mice displayed dramatic reduction of clonotypic TCR with concomitant increase in T cells bearing non-transgenic Vb and Va families, including T cells with dual receptors expressing reduced levels of transgenic Va and endogenous Va. Collectively, the data demonstrate that Treg deficiency allows polyclonal expansion of T cells in a stochastic manner, resulting in widespread changes in the TCR repertoire.

We implement online PWM search algorithm over parallel architecture. A large PWM data can be processed on Graphic Processing Unit (GPU) systems in parallel which can help in matching sequences at a faster rate. Our method employs extensive usage of highly multithreaded architecture and shared memory of multi-cored GPU. An efficient use of shared memory is required to optimise parallel reduction in CUDA. Our optimised method has a speedup of 230-280x over linear implementation on GPU named GeForce GTX 280.
