The present study aimed to detect and characterize Borrelia spp. in ticks attached to dogs in Kor... more The present study aimed to detect and characterize Borrelia spp. in ticks attached to dogs in Korea. Overall, 562 ticks (276 pools) attached to dogs were collected and tested for Borrelia infection by PCR targeting the 5S-23S rRNA intergenic spacer region (rrf-rrl). One tick larva (pool level, 0.4%; individual level, 0.2%) was confirmed by sequencing Borrelia garinii, a zoonotic pathogen. For molecular characterization, the outer surface protein A (ospA) and flagellin genes were analyzed. Phylogenetic ospA analysis distinguished B. garinii from B. bavariensis, which has been recently identified as a novel Borrelia species. On the other hand, phylogenetic analysis showed that single gene analysis involving rrf-rrl or flagellin was not sufficient to differentiate B. garinii from B. bavariensis. In addition, the B. garinii-infected tick was identified as Ixodes nipponensis by sequencing according to mitochondrial 16S rRNA and the second transcribed spacer region. To our knowledge, this is the first study to report the molecular detection of B. garinii in I. nipponensis parasitizing a dog in Korea. Continuous monitoring of tick-borne pathogens in ticks attached to animals is required to avoid disease distribution and possible transmission to humans.
Taxonomy, species diversity, and geographical distribution of the marine brown algal genus <i&... more Taxonomy, species diversity, and geographical distribution of the marine brown algal genus <i>Padina</i> (Dictyotales, Phaeophyceae) from tropical South-East Asian waters were investigated through a combination of morphological and molecular analyses. The study presents a new schematic diagram showing a suite of morphological characters including thallus hairline structures that are useful for species delineation in the genus. This study also revealed two new species from Myanmar, <i>P. gracilis</i> sp. nov. and <i>P. lata</i> sp. nov. based on molecular and morphological data. <i>P. gracilis</i> is characterised by a 2-layered thallus, thin hairlines on both surfaces of the thallus at equal distances, non-indusiate reproductive sori distally very close to the hairlines on the inferior surface, and the presence of <i>Vaughaniella</i> stage. <i>P. lata</i> is characterized by a 2–4-layered tiny thallus, broad hai...

The marine algal flora of eastern Sorsogon has been intensively collected and is generally consid... more The marine algal flora of eastern Sorsogon has been intensively collected and is generally considered as the richest in the Philippines. A trend of species records in the area has been dominated by rhodophytes (red algae) with significantly fewer similar studies for other algal groups (green and brown algae). In this study we present an updated catalogue of the green seaweeds (Ulvophyceae) of eastern Sorsogon. A checklist, including notes on taxonomy, is given of the 103 species. Twenty-six species are newly recorded locally of which five represent new records for the Philippines: Avrainvillea amadelpha, Caulerpa buginensis, an unidentified Caulerpa species, Codium cf. latum, and one taxon new to science. The new species is described as Halimeda magnicuneata Verbruggen et Dumilag based on morpho-anatomy and DNA sequence data. It has a latitudinal Pacific range from south of Japan down to northern Australia, with the highest concentration in Southeast Asia. However, its TWP fishing grounds have long been a hotspot of unsustainable exploitations, thus threatening the remaining wild populations. Despite the species’ commercial significance, there is limited understanding of its genetic structure and diversity. Herein, the genetic structure of S. leptolepis was examined using mitochondrial COI and CytB sequences. Both markers denoted significant genetic structuring based on high overall FST values. Anaplasmosis is a tick-borne, non-contagious, zoonotic disease caused by Anaplasma spp., which in... more Anaplasmosis is a tick-borne, non-contagious, zoonotic disease caused by Anaplasma spp., which include Anaplasma marginale, A. centrale, A. phagocytophilum, A. platys, A. ovis, and A. bovis. Recently, in Korea, the prevalence of Anaplasma spp. has been investigated in some animals, such as dogs, horses, goats, cats, and Korean water deer. In cattle, A. marginale is the most virulent species and regarded as the typical type of species. However, data on the seroprevalence of Anaplasma spp. in cattle in Korea during the last decade is limited. This study was designed to investigate the seroprevalence of bovine anaplasmosis in Korea. From 2010 to 2013, blood samples were collected from 568 cattle. Forty animals (7.0%) tested seropositive for Anaplasma spp. by cELISA. Despite that current bovine anaplasmosis seropositivity rate in the Gyeongsangbuk-do is lower than those in tropical countries, anaplasmosis needs to be regarded as a concerning disease. The identification of the specific Anaplasma species infecting cattle in this province requires additional molecular studies. Moreover, further monitoring and control programs for bovine anaplasmosis is required, and the information from this study will be beneficial to develop these programs.

This study evaluated the anti-oxidant activity of avicularin and isovitexin from Lespedeza cuneat... more This study evaluated the anti-oxidant activity of avicularin and isovitexin from Lespedeza cuneata using 2,2-diphenyl-1picrylhydrazyl (DPPH) and hydroxyl (OH) scavenging assays. The results showed that among the four fractions, the highest OH radical scavenging activity was exhibited by the n-butanol fraction. The DPPH radical scavenging activities of avicularin and isovitexin were higher than 50% at a concentration of 100 μg/mL. Moreover, one hundred micrograms per liter of avicularin and isovitexin exhibited effective anti-oxidant activity, with the OH radical scavenging rates being 87.54 and 91.48%, respectively. These results suggest that Lespedeza cuneata would be useful anti-oxidant agents from natural sources.

Extremophilic microalgae are primary producers in acidic habitats, such as volcanic sites and aci... more Extremophilic microalgae are primary producers in acidic habitats, such as volcanic sites and acid mine drainages, and play a central role in biogeochemical cycles. Yet, basic knowledge about their species composition and community assembly is lacking. Here, we begin to fill this knowledge gap by performing the first large-scale survey of microalgal diversity in acidic geothermal sites across the West Pacific Island Chain. We collected 72 environmental samples in 12 geothermal sites, measured temperature and pH, and performed rbcL amplicon-based 454 pyrosequencing. Using these data, we estimated the diversity of microalgal species, and then examined the relative contribution of contemporary selection (i.e., local environmental variables) and dispersal limitation on the assembly of these communities. A species delimitation analysis uncovered seven major microalgae (four red, two green, and one diatom) and higher species diversity than previously appreciated. A distance-based redundancy analysis with variation partitioning revealed that dispersal limitation has a greater influence on the community assembly of microalgae than contemporary selection. Consistent with this finding, community similarity among the sampled sites decayed more quickly over geographical distance than differences in environmental factors. Our work paves the way for future studies to understand the ecology and biogeography of microalgae in extreme habitats.

Simultaneous determination of three methoxyflavones, namely, cirsimarin (1), hispidulin (2), and ... more Simultaneous determination of three methoxyflavones, namely, cirsimarin (1), hispidulin (2), and cirsimaritin (3) in selected Korean thistles was performed via reversed-phase high performance liquid chromatography system. Compound 1 was present in all the thistle species examined, whereas 2 and 3 were only detected in Cirsium japonicum and C. japonicum var. maackii (CJM). The concentration of compounds 1-3 in CJM varied according to the time of harvest. Plants collected in the spring (CJMS) and fall (CJMF) had the highest contents of 3 and 1, respectively. A lower content of 2 was observed in CJMF than in CJMS. This indicates that seasonal variation affects the flavonoid content of CJM. The results of this study show that CJM is an excellent source of compounds 1-3 and it can potentially be cultivated for industrial and pharmaceutical applications involving these compounds.

In our previous study, four new dammarane-type ginsenosides were isolated from Panax ginseng, which are ginsenoside Rg18 (1), 6-acetyl ginsenoside Rg3 (2), ginsenoside Rs11 (3), and ginsenoside Re7 (4). Accordingly, the goal of this study was to determine the distribution and content of these newly characterized ginsenosides in different ginseng products. The content of compounds 1-4 in different ginseng products was determined via HPLC-UV. The samples included ginseng roots from different ginseng species, roots harvested from different localities in Korea, and samples harvested at different cultivation ages and processed under different manufacturing methods. The four ginsenosides were present at varying concentrations in the different ginseng samples examined. The variations in their content could be attributed to species variation, and differences in cultivation conditions and manufacturing methods. The total concentration of compounds 1-4 were highest in ginseng obtained from Geumsan (185 µg/g), white-6 yr ginseng (150 µg/g), and P. quinquefolius (186 µg/g). The results of this study provide a basis for the optimization of cultivation conditions and manufacturing methods to maximize the yield of the four new ginsenosides in ginseng.

Abstract: Among the distinctive members of the brown algal family Scytosiphonaceae is the genus H... more Abstract: Among the distinctive members of the brown algal family Scytosiphonaceae is the genus Hydroclathrus, which is primarily distinguished by its clathrate (net-like) appearance. Hydroclathrus, despite being seasonal and species poor, is a conspicuous floristic component in many tropical to warm temperate coasts. However, compared with other closely related genera, such as Colpomenia and Rosenvingea, Hydroclathrus remains poorly known; its taxonomy remains problematic, as information on the molecular phylogenetic relationships of the species is still scarce. We attempted to bridge these gaps by describing the diversity and molecular phylogeny of Hydroclathrus based on morphological and genetic (mitochondrial cox1 and cox3 and plastidial psaA and rbcL genes) data. We recognised four Hydroclathrus species: H. clathratus, H. stephanosorus, H. tenuis and H. minutus sp. nov., which is morphologically cryptic to H. tenuis. A new clathrate genus, Tronoella ryukyuana gen. & sp. nov., is also erected based on specimens from southern Japan. Tronoella is primarily distinguished from Hydroclathrus in possessing (1) highly convoluted thalli with branches that are sometimes slightly twisted and/or interadhesive; (2) siphonous protrusions that grow randomly and in various directions, later on becoming perforated branches with revolute margins; and (3) plurangia that are firmly coherent and massive. Our results highlight the need to re-evaluate the identity of the species reported as H. clathratus and H. tenuis and underscore the need to re-examine the taxonomy and systematics of the family Scytosiphonaceae.

This study evaluated the prevalence of species, particularly , and that are pathogenic to cattle,... more This study evaluated the prevalence of species, particularly , and that are pathogenic to cattle, in faecal samples collected from cattle with diarrhoea reared in the Republic of Korea by using microscopy and PCR. In addition, the prevalence of species was analysed according to age, type of cattle, region, season and nature of diarrhoea. Overall, species were identified in 279 of the 1261 (22.1 per cent) faecal samples through microscopy, and statistical analysis revealed a lower prevalence in calves aged than three weeks or less and higher prevalence in cattle with haemorrhagic diarrhoea. Of the 279 microscopy-positive samples, , and were identified in 100 (7.9 per cent), 83 (6.6 per cent) and 27 (2.1 per cent) faecal samples, respectively, by using PCR. To the authors' knowledge, this study is the first to apply PCR for epizootiology of bovine coccidiosis.

Bovine anaplasmosis is a tick-borne, infectious, non-contagious disease caused by Anaplasma margi... more Bovine anaplasmosis is a tick-borne, infectious, non-contagious disease caused by Anaplasma marginale, A. centrale, A. bovis, and zoonotic A. phagocytophilum. Recently, Anaplasma capra detected in goats was identified as a novel zoonotic pathogen. To determine whether A. capra can infect bovines, we used PCR to differentially diagnose Anaplasma spp. in 1219 South Korean cattle by performing multilocus gene typing and restriction fragment length polymorphism (RFLP). Nucleotide sequencing and phylogenetic analysis detected the 16S rRNA gene of A. bovis and four genes from A. capra in 12 (1.0%) and five (0.4%) cattle, respectively. Supplementary discrimination between A. bovis and A. capra was accomplished by RFLP. The 16S rRNA, msp4, groEL, and gltA genes of A. capra identified in this study had much lower degrees of identity to those in A. centrale and other Anaplasma spp. A. phagocytophilum was not detected in any of the tested cattle. Although the prevalence was low, this study suggests the potential of cattle to serve as reservoirs of A. capra. Thus, further studies are needed to clarify the pathogenesis of A. capra in cattle and its possible involvement in transmission to humans.

In this study, we evaluated the inhibitory activity of Cirsium japonicum var. maackii pappus (CJP) against rat lens aldose reductase (RLAR). The ethanolic extract, fractions and isolated flavonoids were subjected to an RLAR assay. Isolation of chloroform (CHCl3) and ethyl acetate (EtOAc) fractions led to the identification of four flavonoids: hispidulin (1), cirsimaritin (2), apigenin (3), and cirsimarin (4). The RLAR assay results suggested that the EtOAc fraction and flavonoids 1 and 3 promoted better AR inhibition than did TMG (control). The half-maximal inhibitory concentration (IC50) of compounds 1 and 3 was 0.77 and 3.19 μM, respectively. A simultaneous determination of flavonoid content using HPLC–UV indicated that CJP contained large amounts of compounds 2 and 3 (1.65 and 1.84 mg/g, respectively). Flavonoids from Cirsium species have been widely reported to show various pharmacological activities. This study indicated that CJP has the potential to prevent diabetic complications and was a potential source of flavonoids.

Pterocladiella maribagoensis is distinguished by small, terete to compressed thalli with alternate or irregular branches, peg-like haptera, rhizines in medulla, and tetrasporangial sori without sterile margin at the terminal of branches. Analyses of both mitochondrial cox1 and plastid rbcL sequences revealed that P. maribagoensis was distinct from other species of Pterocladiella and clustered with P. australafricanensis, P. beachiae, P. caerulescens, P. phangiae, and P. psammophila. Morphological differences between P. maribagoensis and similar species are presented.
