In this research, we evaluated the combined effect of static magnetic field and cis -diamminedich... more In this research, we evaluated the combined effect of static magnetic field and cis -diamminedichloroplatinum(II) (CDDP) on cell death, apoptosis and cell cycle in normal dermal fibroblast (Hu02) and HeLa cells. The cells were exposed to CDDP in the presence of 10 mT SMF for 24 and 48 h. By using MTT assay the IC 50 concentrations of CDDP were determined. The cytotoxic effects of this simultaneous application were studied using flow-cytometric measurements for cell cycle and apoptosis. Based on the obtained results, the simultaneous application of CDDP and static magnetic field for 48 h led to a significant increase in arrest of cell cycle progression of cells in G 2 /M phase, necrosis and late apoptosis in HeLa cells compared to the CDDP-treated group. In contrast, this combination led to a decrease in G 2 /M phase arrest, necrosis cells in Hu02 compared to drug-treated cells. Studies in recent years demonstrated that lipoxygenase (LOX) might play an important role in the plant defence response. In the present work we studied the role of LOX in Taxol biosynthesis pathway in Corylus avellana suspension cultures under ultrasound elicitor. The cells were grown in LS media and were exposed to ultrasound at power density of 4 mW/cm2 for 4 to 40 min. Changes of LOX activity, expression of 1-Deoxy-D-Xylulose-5-Phosphate Reductoisomerase (DXR) and phenylalanine ammonialyase (PAL) were measured. The aim of our research was the improvement of nucleic-acid binding function prediction. Methods: In the current study, nine machine-learning algorithms were used to predict RNA- and DNA-binding proteins and also to discriminate between RNA-binding proteins and DNA-binding proteins. The electrostatic features were utilized for prediction of each function in corresponding adapted protein datasets. The mechanism of Ca2+ transfer through TRPV2 channel is not well known. Here, we investigated the reaction coordination and energy fluctuation of Ca2+ transition in TRPV2 channel by steered molecular dynamics (SMD) simulations and potential of mean force (PMF) calculation. Results showed that electrostatic interactions between Ca2+ and residues of the first and second gates had main roles in ions transfer through the channel, and also, we recognized important amino acids in this path. Moreover, results indicated that enter and exit of calcium ions needed to overcome barrier energies in first and second gates.

Aflatoxins are a kind of mycotoxins that are mostly produced by a group of molds such as Aspergil... more Aflatoxins are a kind of mycotoxins that are mostly produced by a group of molds such as Aspergillus flavus and Aspergillus parasiticus. The studies on different parts of Iran showed that AFM 1 infection of milk is higher than European Union's standard. In this study, the specific aptamer with the carboxylic group at the end of 5′ and Fe 3 O 4 nanoparticles with amine groups was synthesized. Morphological and structural qualities of Fe 3 O 4 were determined by the Fourier-transform infrared spectrograph (FTIR), dynamic light scattering, scanning electron microscope, and X-ray diffraction devices. The specificity of the aptamer to AFM 1 was investigated in adjacent of AFM 1 and aflatoxin B 1. Then, aptamers were attached to nanoparticles to improve synthetic qualities and to ease of its detachment. The attachment was approved by FTIR method. The complex (Fe 3 O 4-APT) was then added to infected milk and after the proper time was detached from the milk using a magnet. The remained amount of AFM 1 was attained in milk using high-performance liquid chromatography. Our result showed that this method for aflatoxin detoxification is much more effective than conventional methods based on recognition of AFM 1 and their concentration in infected milk. This method is more applicable, faster, and cheaper highlighting its application in milk factories.

Abstract Purpose: In this study, the effects of different intensities of Static Magnetic Fields (... more Abstract Purpose: In this study, the effects of different intensities of Static Magnetic Fields (SMFs) (10, 15 and 25 mT) and different concentrations of cisplatin drug were investigated on the viability percent and IC50 of the A2780 and A2780-CP cell lines at 24, 48 and 96 h to show useful potential of SMF as a physical agent to enhance the effectiveness of common therapeutic approaches and decrease of drug resistance to cisplatin anticancer drug. Materials and methods: Magnetic field exposure was performed using a locally designed generator. The cell viability percent, IC50 and cisplatin uptake in treated cells were evaluated by MTT assay and inductively coupled plasma (ICP), respectively. Results: Increasing of concentration and time of cisplatin drug showed a noticeable decrease in viability percent in sensitive and resistant cell lines compared with control group. These decreases were more significant in resistant cells compared with sensitive cells. The obtained IC50 values for resistant were greater than the values obtained for A2780 cells. ICP analysis demonstrated an increased uptake of cisplatin after treatment for 48 and 96 h relative to untreated groups in both resistant and sensitive cells. Conclusion: Results showed that A2780 cells were more sensitive to cisplatin than A2780-CP. Studies have shown that SMF can increase the effect of cisplatin on cell viability percent and decrease the resistance of A2780-CP cells by producing large, verruca shaped structures at the surface of the cell membrane.

Studies have confirmed that on the ground, the plant cells must expend energy to maintain positio... more Studies have confirmed that on the ground, the plant cells must expend energy to maintain positional homeostasis against gravity. Under microgravity conditions, such energy may be saved for other process such as biosynthesis of beneficial metabolites for growth. This hypothesis was examined on a cell line of tobacco (Nicotiana tabacum cv. Burley 21). The cells were continuously treated with 2-D clinostat for 1 week. Exposure to clinorotation conditions increased biomass and total protein. Total content of soluble sugar also increased which may provide more precursors for Krebs cycle and adenosine triphosphate (ATP) production. In the case of 2-D clinorotation, the expression and activity of glutamate producing enzyme, glutamate dehydrogenase (GDH) increased, whereas the activity of glutamate decarboxylase (GAD) decreased. Regarding the role of GAD in initiation of gamma amino butyric acid (GABA) shunt, it is plausible that under clinorotation condition, the tobacco cells directed their metabolism toward saving energy for Krebs cycling and more production of ATP rather than shifting to side paths such as GABA shunt. Improvement of radical scavenging enzymes activity and increase of the contents of phenolic compounds and certain peroxide neutralizing amino acids, e.g., His, Pro, Ser, and Asp under clinorotation conditions decreased membrane lipid peroxidation and maintained the growth potential of tobacco cells.

Early detection of breast cancer plays an important role in reducing the mortality rate of the di... more Early detection of breast cancer plays an important role in reducing the mortality rate of the disease. Dynamic contrast-enhanced magnetic resonance imaging (DCE-MRI) of the breast is especially robust for the diagnosis of cancer in high risk women due to its high sensitivity. Despite excellent sensitivity of DCE-MRI, there is still some difficulty in the prediction of malignancy because of the lack of the optimum guideline for the interpretation of breast MR studies as well as the reported overlap in T1 and T2 relaxation times. In this research, we utilized chaos analysis and nonlinear dynamics in the interpretation of breast masses on DCE-MRI. The analysis is performed after injecting 11 patients with a contrast agent and 16 mass lesions were extracted from these patients. After pre-processing and segmentation stages, time series of the tumor has been generated for every slice image. The time series were further analyzed by extracting the largest Lyapunov exponent (LLE) for each mass lesion. Chaos features such as LLE are significant parameters which can improve the performance of classifiers in the early detection of breast cancer.
