Given its ubiquitous expression and potent influence on cell survival, cancer cell-directed targeting of the CK2 signal is an important factor for development of an anti-CK2 therapeutic. We previously reported on the malignant cell specificity and effect on CK2 signaling of a tenfibgen (TBG) based nanocapsule for delivery of the CK2 small molecule inhibitor 2-dimethylamino-4,5,6,7-tetrabromo-1H-benzimidazole (DMAT) in cultured prostate cancer cells. Here we tested the ability of TBG-DMAT to affect the growth of prostate xenograft tumors in mice. Our results show that treatment of PC3-LN4 xenograft tumors with TBG-DMAT caused loss of proliferative Ki-67 signal as well as Nuclear Factor-kappa B (NF-κB) expression in the tumors. Further, the TBG-DMAT nanocapsule was detected in tumors and not in liver or testis. In conclusion, TBG-b...

In humans, four RSK isoforms (RSK1-4) have been identified. Although they are 73-80% conserved in sequence homology, RSK isoforms are reported to have distinct functions. RSKs phosphorylate cyclic AMP response element binding protein (CREB) which is a regulator of hematopoietic proliferation and differentiation. RSK1 is hyperactivated in AML and is therefore a potential therapeutic target for acute myeloid leukemia (AML). Therefore, inhibition of RSK requires understanding of its role during normal hematopoiesis. Furthermore, the role of RSK isoforms during hematopoiesis and leukemogenesis has not been well characterized. To study the expression of RSK isoforms in hematopoiesis, we cultured human cord blood CD34+ (CBCD34+) hematopoietic stem and progenitor cells (HSPCs) in differentiating media containing SCF, Flt3L, TOP, IL-3, IL-6, GM-CSF, and EPO and performed q...

The optimization of a regression test depends on the size of the test suite to be executed. Regression testing helps to verify existing modifications (fixing bugs) or verify new features added to a software product. When regression testing is applied to database applications it is necessary to consider aspects of the database along with the product code in order to guarantee a proper verification of the product. In this paper we present an approach for conducting regression tests on database applications under an iterative and incremental development setting.

The objective is to develop a package capable of improving the organoleptic characteristics such as shelf-life of the product. To develop this study we chose to use in modified atmosphere packaging of filleted. For this we used different concentrations in each mixture. The final product was subjected to two tests, the first microbiological and sensory evaluation at the second, which established the difference between the sensory characteristics and packaging. Finally presents the proposal package which has better influence on the product packaging to the given conditions. The end product is a package that helps increase the shelf life as well as the organoleptic properties of choice for this type of product.

One well-known agile approach is eXtreme Programming (XP) where pair programming (PP) is a relevant practice. Although various aspects of PP have been studied, we have not found, under a traditional model of PP, studies that examine the impact of using an IDE tool support. In an attempt to obtain a better understanding of the impact of using an IDE, we present the results of a controlled experiment that expose the influence on quality, measured as the number of defects injected per hour, of pair and solo programmers with and without the use of an IDE. Our findings suggest that the use of an IDE results in significantly higher defect injection rates (for both pairs and solos) when the programming task is not very complicated. Nevertheless, defect injection rates seem to decrease when pairs work on more complicated programming assignments irrespective of the tool support used. Quality is affected in a similar manner when either pair or solo programming is used.

(VALENCIA., 2010.)

The methodology of this proposal takes into account legal requirements and constraints on the assignment of teachers and classrooms in public educational institutions in Colombia. In addition, this proposal provides a set of constraints focused on cognitive rhythms and subjects are scheduled at the most convenient times according to the area of knowledge. The genetic algorithm evolves through a process of mutation and selection and builds a total solution based on the best solutions for each group. Sixteen groups in a school are tested and the results of class schedule assignments are presented. The quality of the solution obtained through the established approach is validated by comparing the results to the solutions obtained using another algorithm.

The accumulation of CTH, specifically in HBC, was attributed to the overexpression of cystathionine beta synthase (CBS), its synthesizing enzyme, and the undetectable levels of its downstream metabolizing enzyme, cystathionine gamma lyase (CGL). Interestingly both CBS and CGL could not be detected in normal breast tissues. We further observed that CTH protected HBC cells against excess reactive oxygen species (ROS) and chemotherapeutic drug-induced apoptosis. Moreover, CTH promoted both mitochondrial and endoplasmic reticulum homeostasis in HBC cells. As both the mitochondria and the endoplasmic reticulum are key organelles regulating the onset of apoptosis, we reasoned that endogenous CTH could be contributing towards increasing the apoptotic threshold in HBC cells. An increased apoptotic threshold is a hallmark of all cancer types, including HBC, and is primarily responsible for drug resistance. Hence this study unravels one of the possible pathways that may contribute towards drug resistance in HBC.
