Glioblastoma multiforme (GBM), the most aggressive primary brain tumor, displays an enrichment ge... more Glioblastoma multiforme (GBM), the most aggressive primary brain tumor, displays an enrichment gene expression related to a stem cell signature. Sox2, a major stemness factor involved in pluripotency and in the maintenance of neural stem cell identity, has been described as frequently overexpressed in poorly differentiated gliomas. However, the role that this molecule plays in brain tumors remains under investigation. In this study, we sought to ascertain the Sox2 potential function in the control of self-renewal of brain tumor stem cells (BTSCs) and the malignant phenotype of gliomas. Our results showed Sox2 expression in human surgical malignant gliomas. In addition, Sox2 expression was significantly higher in the BTSC lines than in established glioma cell lines. Downmodulation of Sox2 in BTSCs by using siRNA resulted in the loss of the self-renewal properties of this cell population, and modification in the pattern of expression of markers related with stem cells properties and differentiation. These results are of major importance since, in the TCGA, we observed Sox2 overexpression in more than 80% of the samples (n=188), gene amplification of SOX2 in 18% of the individuals, and hypomethylation of the promoter in about 90% of the samples when compared with normal tissue, suggesting the relevance of this transcription factor in the malignant phenotype of GBMs. In this regard, our results demonstrated that SOX2 plays a prominent role in the invasive and migratory capabilities of both BTSCs and established glioma cell lines. Thus, ectopic expression of Sox2 was sufficient to induce invasion of glioma cells and knockdown experiments demonstrated that Sox2 is essential for maintaining the invasive properties of glioma cells. Therefore, these results identify a multi-functional aspect of Sox2 as both, a stemness promoting molecule and an essential pro-invasive protein, elucidating the possibility of targeting Sox2 for glioma therapy. [1] Current treatment of GBM consists of maximal resection followed by radiotherapy and chemotherapy, [2] and has remained unchanged since 2005. Histone deacetylases (HDACs) regulate cellular processes such as cell cycle, proliferation, cell survival, and angiogenesis, which make them an interesting therapeutic target for the treatment of cancer. [3] An aberrant expression or dysfunction of these proteins is related to different types of tumors, such as colon cancer, [4] gastric cancer, [5] and GBM. [6] Accordingly, some HDAC inhibitors have been developed to treat cancer. These drugs can revert the acetylation status of proteins found in different kinds of tumors, and reactivate the expression of tumor suppressor genes. A 56-year-old woman was seen for an asymptomatic papular eruption of 23 years duration on the dor... more A 56-year-old woman was seen for an asymptomatic papular eruption of 23 years duration on the dorsa of both hands. She described an absence of lesions on the trunk and lower limbs. Pruritus occasionally accompanied the lesions. There were no accompanying systemic problems, and the patient was otherwise in good health. The patient stated that her father suffered similar lesions in the same areas. Complete physical examination revealed numerous translucent, white-ivory lesions 3 to 5 mm in diameter, with a smooth surface, firm consistency, and located exclusively on the dorsum of the hands and the wrists and on the radial side of the distal third of both forearms. There was no induration or thickening of the affected skin (Fig, 1). The remainder of the physical examination was not significant. The following laboratory studies were normal or negative: hemogram, hepatic enzymes, thyroid hormones, proteinogram, and immunoglobulins. Hematoxylin and eosin stained sections of the biopsy specimen revealed a cutaneous papule with a normal epidermis, and a material presence in mid and upper reticular dermis, apparently well circumscribed with an increase of space among collagen bands, and few fibroblasts (Fig. 2). This material was positive when stained with colloidal iron (pH 2.5) (Fig. 3).

Introduction/Background To develop an ultrasound-based score for predicting debulking surgery out... more Introduction/Background To develop an ultrasound-based score for predicting debulking surgery outcome in advanced stage ovarian cancer. Methodology Retrospective analysis of data from a prospective cohort study comprising 65 women (mean age: 59 y.o.) with surgically confirmed stage IIB–IV epithelial ovarian cancer and ECOG status performance 0–2. All women had undergone transabdominal and transvaginal ultrasound evaluation for assessing disease spread prior to surgery. 12 anatomical areas were evaluated for the presence/absence of disease (recto-sigma involvement, carcinomatosis of pelvic peritoneum, major omentum, carcinomatosis of upper abdomen peritoneum, root of mesentery, pelvic lymph nodes, aorto-cava lymph nodes, lesser omentum, liver, spleen, hepatic hilum and ascites). All women included underwent surgery at our institution (diagnostic laparoscopy followed by upfront surgery if patient deemed adequate for optimal (R<1 cm) or complete (R0) cytoreduction). Surgical findings and permanent frozen section histologic diagnosis were used as reference standard. The score was developed according to the odd ratio (for optimal or complete cytoreduction) for each anatomical area evaluated and the value of CA-125 serum levels. (table 1). Results Cytoreduction was complete or optimal in 51 patients (78.5%), suboptimal in 3 (4.6%) and upfront surgery was discarded in 11 women (16.9%). A score >3 predicted correctly 44 out 51 patients (86%) with complete/optimal cytoreduction in and 8 out of 14 (57%) of those with suboptimal or non-cytoreduction surgery. AUC was 0.76, with correct prediction in 80% of all cases. Conclusion Albeit the series is small, the developed ultrasound-based score has a good performance for predicting surgical results in IIB–IV stage epithelial ovarian cancer. However, only sporadic case reports of subcutaneous GA sonography have been published to date. The objective of this study was to evaluate the ultrasonographic patterns of the different clinical variants of GA: localized, generalized, subcutaneous, and perforating.MethodsIn this retrospective observational study, we analyzed and correlated the clinical, histopathological, and sonographic features of 15 patients diagnosed with GA.ResultsWe included 8 women and 7 men with a mean age of 48.4 years (8–77 years). We found three different sonographic patterns depending on the clinical variant of GA: poorly defined hypoechoic band including the dermis (dermal pattern), irregularly shaped hypoechoic hypodermal lumps (hypodermal pattern), and ill-defined hypoechoic dermal and subcutaneous lesions (mixed pattern). Five cases showed increased blood flow signal on Doppler interrogation.ConclusionAlthough our findings are broadly consistent with the previous reports of subcutaneous GA, the sonographic features in localized, generalized, and perforating GA have not been previously reported.
