Aim To evaluate in a laboratory setting the response of human mesenchymal stem cells (MSCs) to pu... more Aim To evaluate in a laboratory setting the response of human mesenchymal stem cells (MSCs) to pulp capping materials with and without Resveratrol (RSV). Methodology Five materials, Calcimol LC, Life, TheraCal LC, ProRoot MTA, and Biodentine, were prepared according to the manufacturers' instructions. Human MSCs were then exposed to these materials, with and without RSV, for 24 h (n=8). Cell viability was evaluated using the MTT assay, and total cell death was quantified by annexin V-FITC staining with flow cytometry. The expression levels of the IL-8, IL-10, HBD-2 and BCL-2 genes were investigated using real-time polymerase chain reaction (RT-PCR). Data obtained from MTT test were analyzed using one-way ANOVA, and Tukey's multiple-comparison test. The Paired Student t test was employed to compare the effects of materials on gene expression (significance level of 5%). Results The group cell viabilities were Calcimol LC 53%, Life 43%, TheraCal LC 78%, ProRoot MTA 75%, and Biodentine 78%. The Calcimol LC and Life exhibited significant differences compared with the control groups (p <0.05). The percentages of necrotic/late apoptotic cells in Calcimol LC and TheraCal LC were higher than in the other materials. However, when RSV was added to wells containing materials, cell viability increased to Calcimol LC 63%, Life 52%, TheraCal LC 82%, ProRoot MTA 91%, and Biodentine 96%, and the percentages of early apoptotic and late apoptotic/necrotic cells decreased. Calcimol LC+ RSV and Life+ RSV differed significantly from the control group (p<0.05). The expression of IL-8 gene was high for all materials, ProRoot MTA caused significant overexpression, and the addition of RSV reduced the expression of IL-8 in the Calcimol LC, TheraCal LC and ProRoot MTA groups and led to increased expression of IL-10 in the Calcimol LC, Life and Biodentine groups. HBD-2 and BCL-2 exhibited increased expression in ProRoot MTA with RSV (p<0.05). Conclusions The addition of RSV exerted a protective effect on MSCs and regulated the inflammatory process by altering the expression levels of pro-and anti-inflammatory genes.

Aim:To evaluate the shear bond strength (SBS) of 3 fissure sealants used alone and with ozone and... more Aim:To evaluate the shear bond strength (SBS) of 3 fissure sealants used alone and with ozone and laser pretreatment.Materials and Methods:For the SBS test, the flattened enamel surfaces of 90 healthy third molars were randomly divided into nine groups ( n = 10 for each groups), based on different sealants (BeautiSealant, Fuji Triage, and Grandio Seal) and pretreatment conditions (no pretreatment, pretreatment with ozone, and pretreatment with neodymium-doped yttrium aluminum garnet [Nd:YAG laser]). Ozone was applied for 30 s in caries superficial mode, and Nd:YAG laser was applied for 60 s. After the pretreatment procedures, sealants were placed inside a mold on the enamel surface. SBS was measured with a universal testing machine. Data were analyzed using analysis of variance. Materials and Methods: Three bulk-fill composite (Beautifil Bulk-Fill Giomer, Filtek Bulk-Fill, and X-Tra Fill) and a methacrylate-based composite (Filtek Z250) were used in this study. Thirty disc-shaped specimens, 4 mm thick, were prepared from each composite resin. Specimens were divided into three groups and polymerized with light-emitting diode (LED), Plasma arch curing unit (PAC), and quartz-tungsten halogen LCU. OBJECTIVE The aim of this study was to evaluate the effect of brushing with whitening toothpaste ... more OBJECTIVE The aim of this study was to evaluate the effect of brushing with whitening toothpaste on the color change of composite resins discolored with coffee. METHODS Disc-shaped samples were prepared using one nano-hybrid (Clearfil Majesty Esthetic), one micro-hybrid (GC G-aenial Anterior), and one nano-ceramic composite (Zenit) (n=6). After initial color measurements, the samples were colored in the coffee solution for 5 days. Color measurements were made again after the coloring process, and brushing simulation was applied for 7 days, 15 days, and 30 days with two different whitening toothpastes containing calcium carbonate+perlite (Signal whitening system,SWN) and hydrated silica+hydrogen peroxide (Colgate optic White, COW) and a conventional non-whitening toothpaste containing hydrated silica (Sensodyne pronamel, SPN). Color change (∆E00 ) after brushing was calculated using the CIEDE2000 system. Repeated measures variance analysis was used for the analaysis of data. RESULTS G-aenial anterior and Zenit showed significant coloration compared to Majesty esthetic after coloration with coffee (p<0.05). The color change in all composite resins was above the clinically acceptable limit (ΔE00 >1.8). Color change over time after brushing with both whitening toothpastes differed significantly (p<0.05). SWN and COW exhibited a significantly higher whitening effect than SPN (p<0.05). While there was no significant difference in composite groups brushed with the SWS (p>0.05), there was a significant difference between Majesty esthetic, G-aenial anterior, and Zenit in groups brushed with COW (p<0.05). CONCLUSIONS Color change was high in micro-hybrid and nano-ceramic composites which were discolored in coffee. The whitening toothpastes were effective in reducing the coloration caused by coffee.

Aim: The purpose of this in vitro study was to determine how pretreatment with desensitizers affe... more Aim: The purpose of this in vitro study was to determine how pretreatment with desensitizers affects micro-tensile bond strength between dentin and self-etch resin cement (SEC). Material and Methods: The occlusal surfaces of extracted teeth were abraded with a diamond bur to the dentin layer and divided into 5 equal groups (n = 20); G1-control (no desensitizer, only Clearfil aesthetic resin cement (CEC); G2-Gluma and CEC; G3-Bi-fluorid 12 and CEC; G4-FGM desensitizer and CEC; G5-D/Sense Crystal and CEC. Resin cement layers were added to a depth of 8 mm in the control group, and desensitizer was applied to the surfaces before resin cement application in the other experimental groups. A specimen from each group was examined with scanning electron microscopy. The specimens were sectioned to be 1 mm 2 perpendicular to the adhesive layer, and the microtensile test was applied. According to the results of the post hoc Tamhane test, there was a significant difference. (p<0.05). Results: Only the Gluma desensitizer had a significantly lower bond strength value when compared with the other desensitizers and the control group. Conclusions: While dentin with oxalate, fluoride, and strontium chloride-potassium nitrate contamination did not affect the bonding strength of SEC, dentin contamination with glutaraldehyde/HEMA had a negative effect. ÖZ Amaç: Bu in vitro çalışmanın amacı, dentine hassasiyet giderici ajan uygulamasının, self-etch rezin simanın (SEC) mikrogerilim bağlanma dayanımını nasıl etkilediğini araştırmaktır. Gereç ve Yöntem: Çekilmiş dişlerin oklüzal yüzleri elmas frez ile dentin tabakasına kadar aşındırıldı ve 5 eşit gruba ayrıldı (n = 20); G1-kontrol (hassasiyet giderici ajan kullanılmadı, sadece Clearfil estetik rezin siman (CEC); G2-Gluma ve CEC; G3-Bi-fluorid 12 ve CEC; G4-FGM ve CEC; G5-D/Sense Crystal ve CEC. Kontrol grubunda, dentin üzerine 8 mm yüksekliğinde rezin siman tabakası eklendi ve diğer deney gruplarına rezin simandan önce dentin hassasiyet giderici ajanlar uygulandı. Her gruptan bir örnek taramalı elektron mikroskobu ile incelendi. Örnekler dentin-siman bağlantısına dik olarak 1 mm 2 olacak şekilde kesildi ve mikrogerilim bağlanma dayanımı ölçüldü. Post-hoc Tamhane testinin sonuçlarına göre anlamlı bir fark vardı (p <0.05). Bulgular: Dentin hassasiyet giderici ajanlar arasında sadece Gluma, diğer gruplar ve kontrol grubu ile karşılaştırıldığında anlamlı şekilde daha düşük bir bağlanma dayanımına sahipti. Sonuç: Dentin üzerine oksalat, florür ve stronsiyum klorür-potasyum nitrat uygulaması, SEC ile arasındaki bağlanma dayanımını etkilemese de, glutaraldehit/HEMA uygulaması olumsuz etkilemiştir. Bu çalıĢmanın amacı, direkt veya indirekt polimerizasyon teknikleri ile polimerize edilmiĢ farklı... more Bu çalıĢmanın amacı, direkt veya indirekt polimerizasyon teknikleri ile polimerize edilmiĢ farklı kompozit rezinlerin mikrosertlik, bükülme dayanımı, su emilimi ve çözünürlüklerini değerlendirmektir. Materyal ve Metot: Rezin matriksine göre sınıflandırılan 3 kompozit rezin seçildi: U200 (3M ESPE), Grandio (VOCO), Xtrafil (VOCO). Kompozit rezinler polimerizasyon tekniğine göre üç alt gruba ayrıldı: LED (Eliapar, 3M ESPE), LED+ indirekt ıĢık (DI 500 Coltene) , indirekt ıĢık+ısı+basınç (Tescera ATL, Bisco Dental). Her bir alt grup için yedi dikdörtgen örnek (25mm x 2mm x 2mm) yapıldı ve bükülme dayanım değerleri için üç nokta büküm testleri yapıldı. Vickers sertliğini, su emme derecesini ve çözünürlüğünü belirlemek için yedi silindirik örnek (çap: 8mm, kalınlık: 2mm) yapıldı. Silindirik örnekler 37 ° C'de 24 saat boyunca distile suda saklandı ve mikrosertlik değerleri bir Vickers sertlik test cihazı ile ölçüldü. Su emilim testi için hazırlanan örnekler 24 saat aynı koĢullarda depolandı ve su emme değerleri ölçüldü. Aynı örnekler desikatörde sabit kütle elde edilinceye kadar kurutuldu ve çözünürlük ölçümleri yapıldı. Veriler iki yönlü ANOVA ve Tukey's post-hoc testi (α = 0.05) ile analiz edildi. Bulgular: Polimerizasyon teknikleri ile kompozit rezinler arasındaki etkileĢim yalnızca bükülme dayanım değerlerinde anlamlı idi (p <0.05).Bükülme dayanımı, mikrosertlik ve su absorpsiyonu için en yüksek değer LED+indirekt ıĢıkla polimerize edilen örneklerde görüldü. Çözünürlük için en yüksek değer ise, indirekt ıĢık+ısı+basınçla polimerize edilen örneklerde görüldü. Sonuç: Ġndirekt polimerizasyon yöntemi kompozit rezinlerin mikro sertlik ve bükülme dayanımı değerlerini geliĢtirdi ancak su emilimi ve çözünürlük değerlerini olumsuz etkiledi.

Objective: The release of components from dental materials may cause oxidative stress which is cr... more Objective: The release of components from dental materials may cause oxidative stress which is crucial factor for tissue damage and cell apoptosis or death. The aim of this study was to evaluate the cytotoxicity of different flowable composites and this materials effect on total antioxidant capacity (TAC) and total oxidant status (TOS) level in human gingival fibroblast cell culture. Materials and Methods: Gingival fibroblast cells obtained from healthy persons were used for evaluation the cytotoxicity and oxidant status. Six flowable composites used were: two bulk-fill flowable composites (SureFil SDR, X-tra base), a self-adhering flowable composite (Vertise Flow), a highly filled flowable composite (GrandioSO Flow), two conventional flowable composites (Filtek Ultimate, Clearfil Majesty). Specimens in 3 mm diameter, 2 mm height were prepared from each composite (n=6) and were transferred to 24 well plates. Wells without composite material were used as the control group. After 24 h incubation period, cytotoxicity was determined by using the 3-(4,5 dimetylthiazol-2-yl)-2,5 diphenlytetrazolium bromide (MTT) assay. Oxidative alterations were assessed using TAC and TOS assay kits. Data were analyzed using the ANOVA and least significant differences posthoc test. Results: Cytotoxicity of six materials was significantly different from the control group (p<0.05). Vertise flow was the most cytotoxic material. TAC levels of Vertise flow were significantly different from X-tra base and GrandioSO. TOS levels increased in SureFil SDR and Vertise flow groups but it was not statistically significant difference. Conclusion: All of the materials used in this study showed cytotoxic effect in human gingival fibroblast cell culture. These materials did not have a significant effect on TOS level. However, TAC level could not prevent the rise of TOS level in Vertise and sureFil SDR. The purpose of this study was to evaluate the relationship between the preparation techniques and type of polymerization techniques on microleakage of composite laminate veneers. Ninety-one same sized, caries-free human maxillary central incisors were randomly assigned to 13 groups (n = 7) and were designed with four different preparation techniques (window type, feather type, bevel type, and incisal overlap type). One group determined as control group and any preparation was applied. Nanohybrid resin composite was used for restoration. Composite laminate veneers polymerized with three different techniques (direct light curing, indirect polymerization with a combination of pressure, light and heat using a light cup and heat cup, direct polymerization, and additionally heat cured in an oven). The specimens were thermocycled, and then immersed in 5% basic fuchsine solution. Following 24 h, all specimens were immersed in 65% nitric acid solutions for volumetric dye extraction test. Samples diluted with distilled water and centrifuged and microleakage determined by a spectrophotometer. Data were analyzed with two-way ANOVA and Tukey honest significant difference post hoc multiple comparisons test (P &amp;amp;amp;amp;amp;amp;amp;amp;lt; 0.05). For comparing the microleakage value of preparation and polymerization techniques, Window type preparation showed a significant difference in direct polymerization + additional cured group (P &amp;amp;amp;amp;amp;amp;amp;amp;lt; 0.05). Control group was statistically different from the other groups (P &amp;amp;amp;amp;amp;amp;amp;amp;lt; 0.05). Window type laminate preparation can be preferred in indirect polymerization technique because it caused less leakage in this present study.

The etiology of talon cusp is not well understood , but appears to have both genetic and environmental components. Permanent dentition have been involved three times more often than the primary dentition. This anomalous cusp may or may not cantain pulpal tissue. Aim: This study evaluated the effect of different storage conditions on dentin shear bond strengt... more Aim: This study evaluated the effect of different storage conditions on dentin shear bond strength of a Self-Etch adhesive. Material and Methods: Forty extracted non-caries human molars were used in this study. Flat dentin surfaces were prepared on all teeth and embedded in acrylic resin blocks with the occlusal surfaces facing up. The samples were divided randomly into 5 groups (n=8). A Self-etch adhesive (Clearfil S 3 Bond, Kuraray Dental, Okayama, Japan) and a nanohybrid composite resin (Grandio; Voco, Cuxhaven, Almanya) was applied on dentin surface with 3 mm diameter and 4 mm height in a mold. In the first group, composite resin was applied after adhesive and polymerized with LED curing unit (3M Espe, Seefeld, Germany). Samples were stored in distilled water for 24 hours and then shear bond test was applied with a universal testing machine. After this, one of the adhesive was refrigerated and the other one was stored at room temperature. After 1 month and 6 months each adhesive was applied on one group and shear test was performed. Data were recorded and one-way ANOVA test was done with SPSS 20 software (α=0.05). Results: The highest bond strength was observed in samples prepared after 24 hours (18,35 MPa). After 1 month, according to the results, there was no significant difference between samples prepared with the adhesive stored at refrigerated temperature or room temperature (p>0.05), while bond strength values of room temperature after 6 months' group were significantly lower than the others groups (p<0.05). Conclusion: Storage conditions of bonding adhesives are important factors for the bond strength of composite resins to dentin. The adhesives should be stored in the refrigerator as recommended by the manufacturer's instructions.

AMAÇ: Bu çalışmanın amacı farklı içeceklerde belirli zaman aralıklarında bekletilen siloran ve di... more AMAÇ: Bu çalışmanın amacı farklı içeceklerde belirli zaman aralıklarında bekletilen siloran ve dimetakrilat esaslı kompozitlerin renk stabilitelerinin karşılaştırılmasıdır. GEREÇ VE YÖNTEM: Bu çalışma için bir siloran-esaslı kompozit rezin (Filtek Silorane; n=25) ve bir dimetakilat-esaslı kompozit rezin (Alert; n=25) kullanıldı. Her bir materyalden 2 mm kalınlığında ve 8 mm çapında 50 adet kompozit rezin örneği hazırlandı. Örnekler dört renklendirme solüsyonu (çay, kahve, kola, kırmızı şarap) ve kontrol olarak distile suda bekletildi (n=5). Renk ölçümleri, örnekler solüsyonlarda bekletilmeden önce ve bekletildikten 1, 7, 15 ve 30 gün sonra spektrofotometre ile yapıldı. Verilerin istatistik analizinde tek yönlü varyans analizi, Tukey HSD çoklu karşılaştırma testi ve bağımsız örneklemlerde t-testi kullanıldı. BULGULAR: Tüm zaman periyotlarında Alert grubunda çay, kahve ve kırmızı şarapta, Filtek-Silorane grubunda ise kırmızı şarapta bekletilen örneklerde renk değişim miktarları ∆E≥3.3'ten daha fazla bulundu. Tüm solüsyonlar içerisinde en fazla renklendirici solüsyonun kırmızı şarap olduğu ve kompozit materyaller açısından da Alert kompozit materyalin Silorane'den daha fazla renk değişikliği gösterdiği tespit edildi (p<0.05). SONUÇ: Bu çalışmada kullanılan rezin kompozitlerin özellikle kırmızı şarap karşısında renk değişimine hassas olduğu bulunmuştur. Siloran-esaslı kompozit rezinler dışsal renklenmelere karşı daha dirençlidir.
