Sodium Diclofenac is a non-steroidal anti-inflammatory drug (NSAID) taken to reduce inflammation ... more Sodium Diclofenac is a non-steroidal anti-inflammatory drug (NSAID) taken to reduce inflammation and, as an analgesic, reduce pain. Although this drug is widely used in the general population, properties such as the short half-time and some side effects restrict its clinical use. The most common side effects are: gastric irritation, gastritis, peptic ulcer and bleeding. Studies involving biomaterials as carrier for drug release have been proving their efficiency in overcoming those problems and better controling the release rate and targeting of the drug. Natural rubber latex (NRL) has been proven excellent for its biocompatibility and ability to stimulate angiogenesis, cellular adhesion and the formation of extracellular matrix, promoting the replacement and regeneration of tissue. In this work, a NRL membrane is used to deliver sodium diclofenac. Sodium diclofenac (NaDic) was found to be adsorbed on the NRL membrane, with little or no incorporation into the membrane bulk, according to energy dispersive Scanning Electron Microscopy with X-Ray microanalysis (SEM-EDS) spectroscopy. In addition, FT-IR shows that there is no molecular-level interaction between drug and NRL. Already, the X-Ray Diffraction (XRD) of NaDic-NRL shows a broader one spectrum than the sharper halo (amorphous characteristic XRD spectrum) of pure NRL. More importantly, the release time of diclofenac in a NRL membrane in vitro was increased from the typical 2-3 h for oral tablets to ca. 74 h. The kinetics of the drug release could be fitted with a double exponential function, with two characteristic times of 0.899 and 32.102 h. In this study, we demonstrated that the interesting properties provided by NRL membranes combined with a controlled release of drug is relevant for biomedical applications.

Co-Cr-Ni-Mo alloy samples were irradiated by pulsed Yb: YAG laser beam under air and atmospheric ... more Co-Cr-Ni-Mo alloy samples were irradiated by pulsed Yb: YAG laser beam under air and atmospheric pressure. Calcium phosphate coatings were deposited on the irradiated surfaces by the sol-gel method. The sol was prepared from the precursors Ca (NO 3) 2 .4H 2 O and H 3 PO 4 and the modified surfaces were submitted to heat treatment conditions at 400°C. The samples were characterized by the techniques of XRD, FTIR, SEM and contact angle measurements. The physicochemical and morphological analyzes showed the formation of a mixture of phases for the coating by the sol-gel method: calcium pyrophosphate and β-TCP. Thus, through the coatings, it was possible to transform the surface of the Co-Cr-Ni-Mo alloy into a bioactive surface constituted of calcium phosphate ceramics of biological interest. The hemolysis percentage of all Co-Cr-Ni-Mo alloy models were less than 5%, and no sign of thrombogenicity was observed. In this manner, the method of surface modification by laser beam irradiation and bioactive coatings of calcium phosphates of biological interest are promising and economically feasible for use in cardiovascular devices.

Abstract Natural latex serum (NLS) is one of the natural rubber latex fractions from Hevea brasil... more Abstract Natural latex serum (NLS) is one of the natural rubber latex fractions from Hevea brasiliensis tree, which is formed by centrifuged serum and is composed of proteins, acids, nucleotides, salts and carbohydrates. The proteins present in NLS have demonstrated several interesting biological properties, including angiogenic, healing, osteogenic, anti-inflammatory, antimicrobial, in addition to inducing neovascularization, bone formation and osseointegration. Thus, we proposed to characterize NLS by physicochemical techniques and to investigate the biocompatibility by toxicological assays and safety test in Galleria mellonella. Infrared spectrum showed vibrational bands characteristic of amide I, II and III that are linked to the protein content, which was confirmed by the High Performance Liquid Chromatography profile and by the Electrophoresis analysis. This material did not exhibit hemolytic (rate <0.5%) and cytotoxic effects (viability >70%) and was able to enhance the proliferation of fibroblasts (>600%) after 3 days. The pronounced proliferative effect observed in fibroblast cells can be explained by the presence of the fibroblast growth factor (FGF) like protein revealed by the Western blot test. Moreover, NLS did not provoke toxic effects (survival ∼ 80%) on the G. mellonella model, indicating that it is a biocompatible and safe material.

The incorporation of drugs and bioactive compounds in the natural rubber latex (NRL) matrix has b... more The incorporation of drugs and bioactive compounds in the natural rubber latex (NRL) matrix has been an alternative for the development of transdermal release membranes. Ibuprofen (IBF) is known to be used to treat inflammatory diseases, but when administered orally, high concentrations can cause some adverse problems. In this work, the incorporation of IBF in the NRL membranes was evaluated by physical-chemical, in vitro permeation, hemocompatibility and molecular modeling assays. In addition, the in vitro release profile of IBF in acid and basic media was analyzed during 96 h. The IBF-NRL membrane exhibited the absence of intermolecular bonding that could hinder drug release and presented compatible mechanical properties for applications as a cutaneous adhesive (0.58 and 1.12 MPa to Young's modulus and rupture tension, respectively). The IBF-NRL system did not present a significant hemolysis degree (1.67%) within 24 h. The release test indicated that in the first hours of the study, 48.5% IBF was released at basic pH and 22.5% at acidic pH, which is characteristic of a burst effect. Then, a stable release profile was observed until the end of the assay, with total IBF release of 60% in alkaline medium and 50% in acidic medium. The drug permeation results indicated that the IBF-NRL membranes can be used for the local skin treatment with permeation of 3.11% of IBF. Dynamic Molecular simulations indicated a pronounced electric dipole in the ionized form of IBF, which suggests a more effective interaction with water, explaining the efficient drug release in alkaline solutions. In general, the results demonstrate that the IBF-NRL membrane has great potential for a new adhesive that can be used for the treatment of inflammatory processes and injuries.

Background: Nipple pain is the second most common reason for early weaning, exceeded only by the ... more Background: Nipple pain is the second most common reason for early weaning, exceeded only by the insufficient milk supply. Nipple fissures can bring other problems, acting also as a portal for bacteria and leading to mastitis. This work proposes the breast protector composite development using materials with tissue repair and moisturizing properties, aligned with a low-cost procedure, aiming not only to relieve pain, but also to heal the nipple fissures caused by breastfeeding. Materials and methods: For the dressings, production was used Natural Latex extracted from the rubber tree and glycerol. The Samples were evaluated chemically and physically by the techniques of Scanning Electron Microscopy, Fourier transform infrared spectroscopy, mechanical traction, and contact angle. The samples were also biologically evaluated by the hemolytic and cytotoxic activity assays. Results: From the physical-chemical assays, the matrix with glycerol has high pore density; the natural latex and glycerol do not covalently interact, indicating that the glycerol can be released; the glycerol addition makes the matrix more elastic but fragile, and increase the wettability. From the biological assays, both materials showed no hemolytic effects; and the cytotoxicity results showed that glycerol did not present cytotoxicity in the fibroblasts, but show a dose-dependent influence in the keratinocytes. Conclusion: The material developed for application in breast fissures has mechanical properties similar to those found for materials for dermal applications, present high wettability and pore density. Furthermore, the material showed no cytolytic activity and the tests with skin cell cultures demonstrated the biocompatibility.

The biomembrane natural (NRL-Natural Rubber Latex), manipulated from the latex obtained from the ... more The biomembrane natural (NRL-Natural Rubber Latex), manipulated from the latex obtained from the rubber tree Hevea brasiliensis, has shown great potential for application in biomedicine and biomaterials. Reflecting the biocompatibility and low bounce rate of this material, NRL has been used as a physical barrier to infectious agents and for the controlled release of drugs and extracts. The aim of the present study was to evaluate the incorporation and release of peptides using a latex biomembrane carrier. After incorporation, the release of material from the membrane was observed using spectrophotometry. Analyses using HPLC and mass spectroscopy did not confirm the release of the antimicrobial peptide [W 6 ]Hylin a1 after 24 h. In addition, analysis of the release solution showed new compounds, indicating the degradation of the peptide by enzymes contained in the latex. Additionally, the release of a peptide with a shorter sequence (Ac-WAAAA) was evaluated, and degradation was not observed. These results showed that the use of NRL as solid matrices as delivery systems of peptide are sequence dependent and could to be evaluated for each sequence.

Natural rubber latex biomedical (NRLb) obtained from the rubber tree Hevea brasiliensis has shown... more Natural rubber latex biomedical (NRLb) obtained from the rubber tree Hevea brasiliensis has shown great potential in biomedicine and biomaterial applications. NRLb has been utilized as a physical barrier against infectious agents and in the controlled release of drugs and extracts. In the present work, NRLb was polymerized in a lyophilizer using different volumes of water to control the resultant membrane porosity and characterized regarding the surface morphology, water vapour permeability (WVP), mechanical properties, haemolytic activity and cytotoxicity. The release of bovine serum albumin protein from the latex membranes was evaluated. Drug release rates increased with porosity and membranes were able to control protein release up to 12 h. In addition, WVP increased with the quantity of pores. The cell viability observed for the porous membrane was higher than that noted for conventional membranes. In summary, the porosity control of natural latex membranes can be used to modulate properties and make them suitable for biomedical applications, such as wound dressings, modulated gas-exchange membranes and controlled drug delivery systems.

Este artigo buscou detalhar como ocorre a mediastinite pós-operatória em cirurgia cardíaca e quai... more Este artigo buscou detalhar como ocorre a mediastinite pós-operatória em cirurgia cardíaca e quais os impactos gerados em pacientes submetidos a este tipo de procedimento. A mediastinite possui uma alta taxa de mortalidade e morbidade, porém uma baixa prevalência, causada por infiltração e desenvolvimento ou proliferação de agentes infecciosos em resposta ao dano tecidual envolvendo o mediastino, causada por bactérias Gram positivas e Gram negativas. As diversas causas de infecção mediastinal envolvem fatores de risco antes, pré e após a cirurgia. O diagnóstico geralmente é clínico, se manifestando em um padrão sistêmico, além de apresentar achados em exames laboratoriais e imagens. A classificação é dividida em cinco tipos, realizada a partir do tempo do pós-operatório, os fatores de risco e tratamento. A infecção mediastinal é uma complicação grave da cirurgia, geralmente requer terapia antibacteriana de alto custo, cirurgia adicional com o intuito de controlar a infecção e estadias mais longas no hospital. Ainda que as técnicas venham sendo cada vez mais aprimoradas com auxílio do desenvolvimento de equipamentos e drogas farmacológicas, falta um maior investimento em estudos nesse contexto, a fim de diminuir o risco de infecções cirúrgicas e eventualmente a óbito.

Vehicle dynamics analyze the interactions among the vehicle, environment and driving style/profil... more Vehicle dynamics analyze the interactions among the vehicle, environment and driving style/profile applied. The tires are one of the most important components because all traction and braking forces are acting directly in the contact between the tire and ground. Therefore, this work focuses on the study of nonlinear Pacejka tire model, known as Magic Formula. The Pacejka equations are extensively applied in vehicle planar dynamics simulations. First, a vast bibliography review was performed aiming at the available tire models, considering its effects in the vehicle longitudinal and lateral dynamics. This paper presents the method to calculate the forces/efforts in planar behavior, necessary to determine torque, speed, power and others factors that can be used to design the vehicle control. The selected mathematical model was then implemented in Matlab/Simulink® environment, which enables an analysis of the slip and the tire forces according to the performed maneuvers. Finally, the resulting forces and vehicle dynamic response are compared, according to the variations of Pacejka parameters.

The method presented in this work intends to analyze drum brake design parameters of a light duty... more The method presented in this work intends to analyze drum brake design parameters of a light duty automotive drum brake system. The main objective of this work is to correlate brake materials and unstability parameters to identify which condition will effectively reduce squeal propensity. The methodology involves (a) the finite-element method of the brake components, namely, drum, shoes, and frictional linings, (b) static calculations to get a pre-stress state around which (c) is computed the complex eigenvalues of the system. Hence, positive real parts indicate dynamic instabilities which are explored by varying parameters, namely, the modulus of elasticity of the materials and the friction coefficient at the contact of the shoes with the drum. According to calculations, it was observed that there exist a given range of values for Young’s modulus and friction coefficient that are favorable to reduce drum brake squeal occurrence. In addition, the method proposed delivered results that match with brake squeal literature.
