The Rehearsal-lesson: a proposal of rehearsal methodology to bands of Brazilian schools Abstract: This text presents a proposal of rehearsal methodology called Rehearsal-lesson which has as a goal to make bands' rehearsals in Brazilian schools more efficient. To elaborate the proposal of the rehearsal-lesson, the musical development of students from four bands was analyzed in activities involving composition, music appreciation and performance, using as a theoretical reference The Spiral Theory of Musical Development (Swanwick, 1988, from Swanwick and Tillman, 1986).

In solder joints on Cu, not only is Kirkendall voiding at the solder/Cu interface a concern, but also the growth of interfacial Cu-Sn intermetallic compounds (IMCs). In this work, evolution of microstructure in the interfacial region was studied after thermal aging at 100-150°C for up to 1000 h. Special effort was made during sample preparation to reveal details of the interfacial structure. Thickness of the interfacial phases was digitally measured and the activation energy was deduced for the growth of Cu 3 Sn. Kirkendall voids formed at the Cu/Cu 3 Sn interface as well as within the Cu 3 Sn layer. The thickness of Cu 3 Sn significantly increased with aging time, but that of Cu 6 Sn 5 changed a little. The interfacial Cu 3 Sn layer was found growing at the expense of Cu 6 Sn 5. Evolution of the interfacial structure during thermal aging is discussed.

We present a packaging system which makes use of an accessible rapid prototyping method, and facilitates the rapid and reliable implementation of polymeric microfluidic device designs. The packaging system uses a modular design and is based on an adhesive-free connection of a reusable and stiff polymeric interface plate with a disposable, soft microfluidic chip under com- pression. We characterised the system by numerically and experimentally studying the effect of compression and key dimensions on burst pressure and flow rate. All parts are fabricated with readily available low-cost materials and micro-milling technology. The presented approach is both facilitating and systematising the fabrication of devices with different degrees of complexity; keeping assembly and interconnection simple and straightforward. Furthermore, minimising the time...

The first enzyme layer was covalently immobilized to surface-functionalized solid supports using a bifunctional linker, while the second and third enzyme layers were assembled using the protein affinity method based on the strong "avidin-biotin" interaction. Each enzyme was biotinylated before incorporation into the system; avidin was used as the intermediate linker for the consecutive enzyme layer-by-layer process. The model of building multienzymatic systems was evaluated using alternating structures of glucose oxidase, horseradish peroxidase, and invertase. Mono-, bi-, and tri-enzyme arrangements have demonstrated biocatalytic activity when specific substrates were added to the systems. Furthermore, the biocatalytic response was dependent on the relative position of enzymes within the 3D architectures. Finally, the concept was tested using nonalcoholic beverages added to alternating bi-enzyme systems, which have effectively behaved as glucose bioanalytical sensors.

Using the Natural Neighbour Radial Point Interpolation Meshless Method, considering the two-dimensional stress plane approach, this study allows to evaluate the nonlinear behaviour of the bone in the presence of a prosthesis. The numerical model of the bone structure was created using medical imagens, through the segmentation of the bone in the cortical and trabecular types.

In this work, three different interpolation methods are studied and expanded. The first method is the conventional interpolation Spline. The second method is a subdivision based geometric algorithm. The third method interpolates a given set of points with additional point normal constraints. The last two methods were implemented with uniform B-Splines curves. In this work, both methods are expanded to use non uniform B-Spline curves. Three critical curves are used to test the developed methods: circle involute, bowditch and epitrochoid. The results show that the non uniform B-Spline implementations have better quality with smaller errors, once the value of the distance error of the curve is in the order of 10 −15 % of the bound box diagonal of the initial input data points and the normal error is around 10 −4 rad in the worst case.

Hydropower generators turbine blade is a one of kind product that requires specific maintenance. ... more Hydropower generators turbine blade is a one of kind product that requires specific maintenance. They are designed in very specific software tools based on features, which allow good CAD/CAE automation by simply tweaking parameters. However, such CAD/CAE automation has brought an undesired side effect: it's hard to add new features that are not considered in the original CAD. Specially in simulation-optimization applications, where parameters that are not considered in the design must be modified. This is the case in this work, where fluid-structure analysis is performed and a geometric feature that transcend the design activity (blending surface between the crown and the blade) is modified. This paper describes an ongoing project which involves the implementation of a module able to intercept the geometry generated by a CAD program using the IGES format, create/modify a new geometric feature and update the fluid volume and blade geometries. The surface boundary curves are extracted, and the adjacency relationship between the surfaces is determined by coordinate numerical comparison. The surfaces are coherently oriented and the solid models are completely determined. Using the de Boor's algorithm and the surface orientation a model with exclusive triangular faces coherently oriented is created, and then the blending surface is automatically created. The refined mesh for the models are created and fluid-structure analysis is performed for the simulation-optimization.

Solvent tolerant bacteria are particularly appealing for application in biocatalysis, since plenty of relevant educts and products are poorly water soluble. This feature critically limits process productivity in aqueous media, although an opposite picture results if bioconversions are performed in organic environment. Similarly, solvent tolerant bacteria can be of use in bioremediation. Given their potential and relevance, dedicated efforts have been made in order to get a significant insight on the mechanisms underlying solvent tolerance. This is mostly due to cell wall adaptation and rapid repair mechanisms, solvent efflux pumps or enzymatic pathways allowing the mineralization of the deleterious solvent. Although several Gram-positive bacteria (viz. Rhodococcus spp., Mycobacterium spp.) have been effectively used for biocatalysis and bioremediation in the presence of organic solvents, and the mechanisms for solvent endurance have been looked into, the information most widely disseminated on these matters relies on Gram-negative model systems. The present review aims to provide an updated perspective on the applications of Gram-positive bacteria in organic solvents environment, their mechanisms of tolerance and foreseen developments. © 2011 by Nova Science Publishers, Inc. LoC system's designs and development are simultaneously dependent upon the physicochemical properties of the analyte under consideration and governed by the fundamental laws of fluid dynamics, that are usually applied at the macroscale. This chapter provides a brief overview of fluid hydrodynamics and also transport phenomena at the microscale that are relevant to analyte-detection science. This chapter will provide a solid grounding in the possibilities and limitations inherent to operating at the small scale.

The present study reports on a novel approach for the development of enzymatic biosensor systems ... more The present study reports on a novel approach for the development of enzymatic biosensor systems using chemically functionalized supports. Silica glass slides were surface-modified by reacting with organosilanes at room temperature and a glucose oxidase (GOx) enzyme layer was covalently immobilized using the bi-functional linker glutaraldehyde (GA). The activities of enzymes in solution and immobilized on hydroxyl-, amine-and thiol-modified glass surfaces were tested by sensing β-D-glucose with the horseradish peroxidase (HRP) mediated oxidation of 3,3’,5,5’-tetramethylbenzidime hydrochloride (TMB) by H2O2 based on biochemical reactions. The results indicated that the intensity and overall kinetics of the enzymatic catalysis were dependent on solid support chemical functionality with the amine-modified support providing the highest enzymatic catalytic activity.

In the particular case of Leishmania infantum, a unicellular parasite that parasitizes macrophages, infection indexes are usually determined either by visual inspection of cells directly under the microscope or by counting digital images using appropriate software. In either case assessment of infection indexes is time consuming, thus motivating the creation of automatic image analysis approaches that allow large scale studies of Leishmania-infected macrophage cultures. We propose a fully automated method for automatic evaluation of parasite infection indexes through the segmentation of individual macrophages nucleus and cytoplasm, as well as the segmentation and co-localization of the parasites in the image. To perform such analysis with robustness and increased performance we propose the use of local image filters tuned to the specific size of the objects to detect, in conjunction with image segmentation approaches. The objects size estimation is then improved through a learning feedback loop. Cytoplasm is detected by seeded watershed segmentation. Our approach obtains, for 86 images from 4 experiments, an average parasite infection index evaluation error of 2.3%.

This field of research is of acknowledged relevance, since the rational use of small scale reactors is contributing to speed up process development in several areas of biotechnology, in particular due to the possibility of carrying out high throughput assays. Small scale reactors are available in different working volumes, configurations and architectures. These bioreactors are currently used in a wide array of applications, from cell screening to process optimization. The present work is focused on three major application areas, namely steroid biotransformations, siderophore production and screening of antibacterial agents. Results have shown the reliability of these platforms as suitable alternatives of traditional reactors and as screening tools. Fully automated platforms with monitoring and control devices are on the pipeline to further take advantage of the high throughput capability.

These small-scale reactors are available in different working volumes and configurations, being useful in a wide array of applications, from cell screening to process optimization. The present work was focused on the development of a high-throughput strategy, combining microtiter plates and analytic methodologies, to screen an in-house library of environmental bacteria in order to identify good siderophore producers. From a library of roughly 500 marine microorganisms, it was possible to ultimately obtain 11 bacterial strains with high production capabilities. Two of them had not been previously identified as siderophore producers. The bioprocess was scaled-up from microtiter plates to a 5 L stirred tank reactor, while maintaining the overall volumetric productivity, using the k L a similarity as scale-up criterion. This novel approach is a suitable alternative to traditional screening tools.
