Dietitians play a key role in assessing food insecurity and in supporting individuals, families, and communities experiencing this problem. Yet, the detection of food insecurity is often missed during the routine nutritional assessment, and it is not clear why and to what extent this occurs. A grounded theory approach was used to understand the perceptions, knowledge and attitudes of Alberta dietitians from different areas of practice towards addressing household food insecurity in the dietetic practice. The findings revealed that complexity in addressing food insecurity was the central category associated with subcategories: perception of role, continuing education and training, availability of information, resources and support from health system, involvement in advocacy actions, and source of occupational stress. The central category referred to individual, community and organizational level barriers in screening and supporting clients living in food insecurity. These results point to dietitians' need for assistance in developing appropriate skills, and having access to training and resources for screening and discussing food insecurity with clients. Advocating for food security issues was perceived as an important but informal part of the dietitian's job, yet few dietitians reported being involved in food security advocacy. As well, feelings of constant discomfort, helplessness and inadequacy when screening clients for food insecurity were reported by dietitians. These findings will inform the gaps in knowledge for supporting dietitians in different settings to overcome barriers in incorporating food insecurity in routine nutritional assessments.

It begins by compiling thoughts from various theorists who emphasize the importance of opening real application avenues to assess students learning. The research took place in a bilingual college in Bogotá, Colombia, during the second semester of 2011 and the first semester of 2012. The data analysis was taken from a group enrolled in a sixth semester class. Results make clear that through accompanying students through the proper use of PBL ladder, as an application tool, they will be able to value their own process of learning.

Extracted from the mature heartwood of several Santalum species, sandalwood oil is comprised mainly of sesquiterpene olefins and alcohols. Four sesquiterpenols, a-, band nd epi-b-santalol and a-exo-bergamotol, make up approximately 90% of the oil of Santalum album. These compounds are the hydroxylated analogues of a-, band nd epi-b-santalene and a-exobergamotene. By mining a transcriptome database of S. album for candidate cytochrome P450 genes, we cloned and characterized cDNAs encoding a small family of ten cytochrome P450-dependent monooxygenases annotated as SaCYP76F37v1,

These quaternary protoberberines are elicitor inducible and, like other phytoalexins, are highly oxidized. The oxidative potential of these compounds is derived from a diverse array of biosynthetic steps involving hydroxylation, intra-molecular C-C coupling, methylenedioxy bridge formation and a dehydrogenation reaction as the final step in the biosynthesis. For the berberine biosynthetic pathway, the identification of the dehydrogenase gene is the last remaining uncharacterized step in the elucidation of the biosynthesis at the gene level. An enzyme able to catalyze these reactions, (S)-tetrahydroprotoberberine oxidase (STOX, EC 1.3.3.8), was originally purified in the 1980s from suspension cells of Berberis wilsoniae and identified as a flavoprotein (Amann et al. 1984). We report enzymatic activity from recombinant STOX expressed in Spodoptera frugiperda Sf9 insect cells. The coding sequence was derived successively from peptide sequences of purified STOX protein. Furthermore, a recombinant oxidase with protoberberine dehydrogenase activity was obtained from a cDNA library of Argemone mexicana, a traditional medicinal plant that contains protoberberine alkaloids. The relationship of the two enzymes is discussed regarding their enzymatic activity, phylogeny and the alkaloid occurrence in the plants. Potential substrate binding and STOX-specific amino acid residues were identified based on sequence analysis and homology modeling.

But only the opium poppy, Papaver somniferum, and Papaver setigerum are able to produce morphine. In this study, we started to investigate by gene expression analysis the molecular basis for this exceptional biosynthetic ability. A sequencing project from P. somniferum seedlings was initiated using a method based on the amplified fragment length polymorphism technique that resulted in 849 UniGenes. These cDNAs were analysed on macroarrays for differential expression between morphinecontaining P. somniferum plants and eight other Papaver species, which accumulate other benzylisoquinolines instead of morphine. Three cDNAs showing increased expression in P. somniferum compared to all the other Papaver species were identified. Whereas two showed no significant homology to any known protein, one putatively encoded an O-methyltransferase. Analysis of substrate specificity of the heterologously expressed protein and mass spectrometric identification of the enzymatic products identified this protein as S-adenosyl-L-methionine:(R,S)-3¢-hydroxy-N-methylcoclaurine 4¢-O-methyltransferase (EC 2.1.1.116). Unlike other Omethyltransferases of different positional specificities implicated in benzylisoquinoline metabolism, the enzyme only accepted tetrahydroxylated tetrahydrobenzylisoquinolines as substrates; methylation was tolerated only at the 6-hydroxy position.

Although formal syntheses of this alkaloid have been reported, the morphine molecule contains five stereocenters and a CC phenol linkage that to date render a total synthesis of morphine commercially unfeasible. The CC phenol-coupling reaction along the biosynthetic pathway to morphine in opium poppy is catalyzed by the cytochrome P450-dependent oxygenase salutaridine synthase. We report herein on the identification of salutaridine synthase as a member of the CYP719 family of cytochromes P450 during a screen of recombinant cytochromes P450 of opium poppy functionally expressed in Spodoptera frugiperda Sf9 cells. Recombinant CYP719B1 is a highly stereo-and regioselective enzyme; of forty-one compounds tested as potential substrates, only (R)reticuline and (R)-norreticuline resulted in formation of a product (salutaridine and norsalutaridine, respectively). To date, CYP719s have been characterized catalyzing only the formation of a methylenedioxy bridge in berberine biosynthesis (canadine synthase, CYP719A1) and in benzo[c]phenanthridine biosynthesis (stylopine synthase, CYP719A14). Previously identified phenol-coupling enzymes of plant alkaloid biosynthesis belong only to the CYP80 family of cytochromes. CYP719B1 therefore is the prototype for a new family of plant cytochromes P450 that catalyze formation of a phenol-couple.
