However, the association between thyroid disease and breast cancer risk remains unclear. We evaluated the effect of thyroid status on breast cancer growth and dissemination in an immunocompetent mouse model. For this, hyperthyroid and hypothyroid Balb/c mice were orthotopically inoculated with triple-negative breast cancer 4T1 cells. Tumors from hyperthyroid mice showed an increased growth rate and an immunosuppressive tumor microenvironment, characterized by increased IL-10 levels and decreased percentage of activated cytotoxic T cells. On the other hand, delayed tumor growth in hypothyroid animals was associated with increased tumor infiltration of activated CD8+ cells and a high IFNγ/IL-10 ratio. Paradoxically, hypothyroid mice developed a higher number of lung metastasis than hyperthyroid animals. MSCs can be isolated from different sources, like adipose tissue or bone marrow, and have the capacity to differentiate, both in vivo and in vitro, into adipocytes, chondrocytes, and osteoblasts, making them even more interesting in the regenerative medicine field. Tumor associated stroma has been recognized as a key element in tumor progression, necessary for the biological success of the tumor, and MSCs represent a functionally fundamental part of this associated stroma. Exosomes represent one of the dominant signaling pathways within the tumor microenvironment. Lung is the most common metastatic site in osteosarcoma patients with a majority presenting lung ... more Lung is the most common metastatic site in osteosarcoma patients with a majority presenting lung micrometastasis at diagnosis time. MSCs compose a heterogeneous cell population with the ability to migrate and home into remodeling tissues, including tumors. To evaluate the participation of MSCs in lung metastasis formation, we intravenously injected LM7 osteosarcoma cells into immunosuppressed mice. Pre-labelled MSCs (DiR+/CMDiI+, at a dose of 0.5-1×10e6 cells) were intravenously administered 10 weeks after tumor cells injection, when microscopic metastasis become apparent. MSCs biodistribution was evaluated both through real time (DiR+) and microscopic (CMDiI+) analysis. MSCs levels in the lungs of tumor-bearing mice as compared to mice with no tumor burden was 1.15×10E5±0.07 p2/sec/cm2/sr vs. 5.44×10E4±0.5p2/sec/cm2/sr (one week post-inoculation, infrared signal, p<0.01). T-cell lymphoblastic leukemia (T-ALL) is a hematological hematopoietic T cell precursor disorder ... more T-cell lymphoblastic leukemia (T-ALL) is a hematological hematopoietic T cell precursor disorder with a not well defined etiology, although it would be linked to NOTCH signaling defects. CUTLL1 (Columbia University T-cell Lymphoblastic Lymphoma 1) is a human T-ALL cell line displaying biological responses to NOTCH inhibition, thus mimicking the disease´s etiology. The absence of human T-ALL cell lines with verifiably clinical aspects turns the establishment of an in vivo model an asset for preclinical assays; further, it serves to study the interaction between disease-mimicking cells and their surrounding niche. We have established an in vivo model for T-ALL in immunosuppressed NOD.CB17-Prkdc scid/J mice using CUTLL1 cells. Tumors developed after subcutaneous inoculation of a minimum dose of 2x10E7 cells and became visible 2.5 weeks post-inoculation into the right flank (809.1±67.33 mmE3).
