El uso de mapas conceptuales tiene como objetivo conseguir, con el apoyo de los profesores-tutore... more El uso de mapas conceptuales tiene como objetivo conseguir, con el apoyo de los profesores-tutores, que los estudiantes de Veterinaria lleguen a alcanzar conocimientos significativos y perdurables de los contenidos de una asignatura de notoria complejidad como es la Farmacologia, Farmacia y Terapeutica. Los alumnos inscritos voluntariamente (n= 11) en el Seminario desarrollado durante el curso 2011/2012, dentro de las actividades propuestas por el Aula Virtual de Farmacologia en el Campus Virtual de la UCM, participaron activamente en el desarrollo de las siguientes actividades. En primer lugar, un seminario o sesion tutorial de adiestramiento en las tecnicas de elaboracion de mapas conceptuales. A continuacion, se formaron grupos de estudiantes que, con el uso de esta herramienta de estudio, desarrollaron mapas tematicos sobre las distintas secciones que conforman el programa de la asignatura. Thus, in the presence of hydrogen peroxide (H2O2), AR (a nonfluorescent compound) is converted to resorufin (a strongly fluorescing product) by the action of HRP. In the last years, multiple assays have been developed using this reagent to quantify a diverse assortment of analyses and also to detect the activity of many different enzymes. We recently showed that BzATP, agonist of several ionotropic nucleotide receptors (P2X), interfered with Amplex Red oxidation catalyzed by HRP. In the present work we reported that glutamate determinations using Amplex Red Glutamic Acid/Glutamate Oxidase Assay Kit are also strongly interfered by BzATP. The identification of acetylcholine nicotinic receptors (nAChRs) formed by α9 and α10 subunits in... more The identification of acetylcholine nicotinic receptors (nAChRs) formed by α9 and α10 subunits in the sensory cells of the vestibular and auditory systems, prompted us to investigate their presence in adrenomedullary chromaffin cells, in which they could mediate membrane hyperpolarization through the activation of small-conductance Ca2+-dependent K+ channels (SK channels). The aim of the current study has been to pharmacologically identify α9α10 nAChRs and initiate their functional characterization in isolated chromaffin cells from the rat adrenal medulla. We have employed the patch clamp technique to record either the ionic currents generated by the activation of nAChRs or the associated changes in membrane potential. We took advantage of the specificity of α-conotoxin RgIA for the nAChRs formed by α9 and α10 subunits. Our pharmacological results suggest that the rat chromaffin cells express functional α9α10 nAChRs that would influence the electrical behaviour of these cells.

En este trabajo de tesis doctoral hemos estudiado la expresion de la proteinas de choque termico ... more En este trabajo de tesis doctoral hemos estudiado la expresion de la proteinas de choque termico de 70 kDa de expresion constitutiva, llamada Hsc70, en la retina de varias especies de vertebrados. Concretamente hemos estudiado su expresion y distribucion en la retina de pollo, a lo largo de todo el desarrollo embrionario y en estadios postnatales, y en la retina postnatal de rata y conejo. Para ello hemos empleado tecnicas de inmunoblot, inmunohistoquimica. Nuestro resultados muestran que Hsc70 se encuentra en la retina de pollo desde los primeros estadios del desarrollo (al menos desde el dia embrionario 4), y que se expresa en todos los tipos celulares. Si bien la cantidad de proteina detectada por inmunoblot no varia a lo largo del desarrollo ni en los estadios postnatales estudiados, los resultados obtenidos empleando tecnicas inmunohistoquimicas sugieren una diferente regulacion de la expresion de Hsc70 en las capas externas e internas de la retina a lo largo del desarrollo. La expresion de esta proteina tambien esta regulada en estadios postnatales (P), de modo que en animales recien nacidos (P5) se observa un descenso en la expresion de Hsc70 en todas las capas de la retina, mientras que hacia el final del primer mes de vida (P21, P29) se expresa con algo mas de intensidad y de manera similar en todas las capas. El estudio inmunocitoquimico sobre celulas disgregadas, empleando un metodo de disgregacion celular desarrollado en el laboratorio en el que se ha realizado esta tesis doctoral, nos confirma que Hsc70 se expresa en todas las celulas de la retina de pollo durante el desarrollo embrionario y en estadios postnatales: tanto en celulas en ciclo, como en neuroblastos y gliobastos, asi como en neuronas, en celulas de glia y en celulas de retina pigmentaria. Achondroplasia is a pathology due to a mutation in the receptor for the fibroblast growth factor ... more Achondroplasia is a pathology due to a mutation in the receptor for the fibroblast growth factor type 3 (FGFR3). This alteration produces problems in individuals’ stature as well as other muscle-skeletal problems. The application of nucleotides and dinucleotides permit achondroplasic cells (chondrocytes) to recover, aparently, from this pathology. In particular, the application of the dinucleotide Ap4A, permits to restore the correct calcium levels in achondroplasic cell. Moreover, this dinucleotide permits the right degradation of the FGFR3 receptor, which does not downregulate properly in achondropasic chondrocytes, by facilitating the proteosomal and lysosomal pathways alter the dinucleotide application. Also we have discovered that the pyridoxal phosphate derivative PPADS can dramatically reduce the activation of ERK casacade which is abnormaly elevated by the achondroplasic FGFR3 receptor raising the pathology. In summary, we wish to introduce a series of new pharmacological strategies for the treatment of achondroplasia in clear contrast with the current surgery ones. The subcellular distribution and early signalling events of P2X7 receptors were studied in mouse ... more The subcellular distribution and early signalling events of P2X7 receptors were studied in mouse cerebellar granule neurons. Whole-cell patch-clamp recordings evidenced inwardly directed non-desensitizing currents following adenosine 5'-triphosphate (ATP; 600 µM) or 2'-3'-o-(4-benzoylbenzoyl)-adenosine 5'-triphosphate (BzATP; 100 µM) administration to cells bathed in a medium with no-added divalent cations (Ca(2+) and Mg(2+)). Nucleotide-activated currents were inhibited by superfusion of 2.5 mM Ca(2+), 1.2 mM Mg(2+) or 100 nM Brilliant Blue G (BBG), hence indicating the expression of ionotropic P2X7 receptors. Fura-2 calcium imaging showed [Ca(2+)]i elevations in response to ATP or BzATP at the somas and at a small number of axodendritic regions of granule neurons. Differential sensitivity of these [Ca(2+)]i increases to three different P2X7 receptor antagonists (100 nM BBG, 10 μM 4-[(2S)-2-[(5-isoquinolinylsulfonyl)methylamino]-3-oxo-3-(4-phenyl-1-piperazinyl)propyl] phenyl isoquinolinesulfonic acid ester, KN-62, and 1 μM 3-(5-(2,3-dichlorophenyl)-1H-tetrazol-1-yl)methyl pyridine hydrochloride hydrate, A-438079) revealed that P2X7 receptors are co-expressed with different P2Y receptors along the plasmalemma of granule neurons. Finally, experiments with the fluorescent dye YO-PRO-1 indicated that prolonged stimulation of P2X7 receptors does not lead to the opening of a membrane pore permeable to large cations. Altogether, our results emphasise the expression of functional P2X7 receptors at both the axodendritic and somatic levels in mouse cerebellar granule neurons, and favour the notion that P2X7 receptors might function in a subcellular localisation-specific manner: presynaptically, by controlling glutamate release, and on the cell somas, by supporting granule neuron survival against glutamate excytotoxicity.

Here, the expression of this subunit has been detected in wild type mice midbrain, by quantitative real time-polymerase chain reaction, immunocytochemistry and Western blot assays. The functionality of this P2X 7 subunit has been confirmed using microfluorimetric experiments in isolated synaptic terminals from mice midbrain.

The first group corresponds to those responding to Ap 5 A or Gp 5 G at nanomolar concentration, representing 16% and 12%, respectively, and the second one responds to micromolar concentration and represents, respectively, 17% and 14%, of the total functional synaptosomal population in rat midbrain.
