Numerous genetic factors play a crucial role in the progression of prostate cancer. This study was undertaken to reveal the correlation of TP53 rs1042522 and CDH1 rs16260 polymorphisms with the risk of prostate cancer in the Bangladeshi population. Materials and methods We recruited 210 prostate cancer patients and 210 healthy controls for the investigation. Genotyping was conducted using the PCR-RFLP technique. Results In case of TP53 gene rs1042522, the association analysis revealed that all genetic models were linked with significantly increased risk for prostate cancer development (CG vs. CC: OR = 1.99, p = 0.001; GG vs. CC: OR = 4.82, p Conclusions Our study indicates a significant association of TP53 rs1042522 and CDH1 rs16260 polymorphisms with increased prostate cancer risk in the Bangladeshi population. However, replication studies are required in different populations to validate our findings.

An attempt has been made to investigate thrombolytic and antimicrobial activities of ethanolic ex... more An attempt has been made to investigate thrombolytic and antimicrobial activities of ethanolic extracts of Andrographis paniculata whole plant. Phytochemical constituents of A. paniculata were assessed by human erythrocyte and the results were compared with standard streptokinase (SK). Moreover, the plant extracts were compared with the antibiotic kanamycin to investigate antibacterial activity against several microorganisms. Glycosides, steroids, phenols, alkaloid and tannins were found in the ethanol extract of whole plant. Crude ethanol extract (P<0.05) and soluble fraction of ethanol extract (P<0.05) have shown thrombolytic properties. Crude ethanol extract, n-hexane soluble fractions and carbon tetrachloride soluble fraction of ethanol extract of the whole plant have shown antimicrobial activities against common gram positive and gram negative microorganisms. The results of current study justify thrombolytic and antimicrobial activities of A. paniculata.

Early-onset AD (EOAD) and late-onset AD (LOAD, more common) are major identified types of AD. The genetics of EOAD is extensively understood, with three gene variants such as APP, PSEN1, and PSEN2 leading to the disease. Some common alleles, including APOE, are effectively associated with LOAD identified, but the genetics of LOAD is not clear to date. It has been accounted that about 5-10% of EOAD patients can be explained through mutations in the three familiar genes of EOAD. The APOE ε4 allele augmented the severity of EOAD risk in carriers, and the APOE ε4 allele was considered as a hallmark of EOAD. Exploring the genetic causes behind these inter-individual variabilities in platinumbased chemotherapeutic responses by investigating the effects of GSTP1 (rs1695), XRCC1 (rs25487), XPC (rs2228001) and ERCC1 (rs11615) genetic polymorphisms on toxicity and therapeutic response of this treatment among Bangladeshi advanced non-small cell lung cancer (NSCLC) patients was the aim of this study. 285 Clinically proven either stage IIIB or IV (advanced) NSCLC patients aging not less than 18 years old and receiving platinum-based chemotherapy were recruited to assess the influence of these four single nucleotide polymorphisms (SNPs) on peripheral leukocytes. Toxicity and response were evaluated by multivariate regression analyses using SPSS statistical software (version 17.0). XRCC1 (rs25487) polymorphism was found to act as a predictive factor for not only grade 3 and 4 anemia (p = 0.008), neutropenia (p = 0.010), thrombocytopenia (p = 0.025) and gastrointestinal toxicity (p = 0.002) but also for therapeutic response (p = 0.012) in platinum-based chemotherapy. Although GSTP1 (rs1695) polymorphism might serve as prognostic factor regarding grade 3 or 4 neutropenia, a significant (p = 0.044) improvement in response to platinum-based chemotherapy was observed. However, XPC (rs2228001) and ERCC1 (rs11615) polymorphisms could not establish any significant relation with toxicity or therapeutic response. XRCC1 (rs2228001) and GSTP1 (rs1695) polymorphisms might explain platinum-induced clinical outcomes in terms of both toxicity and therapeutic response variations among Bangladeshi advanced NSCLC patients.

Objective: To analyze the phytochemical constituents responsible for the plausible antioxidant ef... more Objective: To analyze the phytochemical constituents responsible for the plausible antioxidant effect of methanolic extract of the seed, pulp and peel of Baccaurea ramiflora Lour. Methods: Fresh seed, pulp, and peel of Baccaurea ramiflora fruits were extracted with methanol (MEBRse, MEBRpu, MEBRpe) and evaluated by phytochemical analysis for their content of innumerable metabolites (primary and secondary) viz. carbohydrates, alkaloids, glycosides, tannins, phenols, terpenoids, flavonoids, proteins, and fixed oils. The antioxidant efficacy was assessed through different assay methods viz. 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radical scavenging activity, total antioxidant capacity (TAC) and reducing power capacity (RPC). Estimation of total phenolic content (TPC), and total flavonoid content (TFC) was also done to confirm the presence of these phytochemicals. Results: It was revealed from the phytochemical analysis of MEBRse that alkaloids, glycosides, carbohydrates, phenols, and flavonoids were present, while that of MEBRpu showed the existence of carbohydrates, proteins, alkaloids, glycosides, phenols, saponins, flavonoids, and fixed oils. Presence of carbohydrates, alkaloids, phenols, tannins, flavonoids, and terpenoids were found in the MEBRpe. A significant antioxidant activity was revealed by the MEBRpu [EC50: (27.612 ± 1.375) μg/mL], compared to MEBRpe, and MEBRse in DPPH assay. The ranking order for RPC was MEBRpu > MEBRpe > MEBRse respectively. The EC50 value of TAC of the MEBRpu, MEBRpe, and MEBRse were (25.107 ± 0.744) μg/mL, (241.127 ± 7.463) μg/mL and (372.364 ± 11.030) μg/mL, respectively. Quantity of TPC and TFC were the highest in the MEBRpu (124.360 ± 2.078 mg GAE/g and 107.527 ±1.900 mg QRE/g extract) rather than MEBRpe and MEBRse extracts. Conclusions: This study suggests that MEBRpu has a significantly higher antioxidant property than MEBRpe and MEBRse. These extracts might be advantageous in prevention or decelerating the progress of different diseases related to oxidative-stress/damage. Moreover, detailed analysis of these extracts is required to identify the presence of promising compound(s) responsible for their antioxidant activity.

Currently, there is no drug that can reduce the pathological events of this degenerative disease but symptomatic relief is possible that can abate the disease condition. N-methyl-D-aspartate (NMDA) receptors exert a critical role for synaptic plasticity as well as transmission. Overstimulation of glutamate receptors, predominantly NMDA type, may cause excitotoxic effects on neurons and is recommended as a mechanism for neurodegeneration. Atypical activation of the NMDA receptor has been suggested for AD by synaptic dysfunction. NMDA receptor antagonists especially memantine block the NMDA receptor and can reduce the influx of calcium (Ca 2+) ions into neuron, thus, toxic intracellular events are not activated. This review represents the role of NMDA receptors antagonists as potential therapeutic agents to reduce AD. Moreover, this review highlights the repositioning of memantine as a potential novel therapeutic multitargeting agent for AD.

Melatonin, a pineal gland synthesized neurohormone is known as a multifunctioning pleiotropic age... more Melatonin, a pineal gland synthesized neurohormone is known as a multifunctioning pleiotropic agent which has a wide range of neuroprotective role in manifold age-related neurodegenerative disorders especially Alzheimer's diseases (AD). AD is a devastating neurodegenerative disorder and common form of dementia which is defined by abnormal and excessive accumulation of several toxic peptides including amyloid β (Aβ) plaques and neurofibrillary tangles (NFTs). The Alzheimer's dementia relates to atrophic changes in the brain resulting in loss of memory, cognitive dysfunction, and impairments of the synapses. Aging, circadian disruption, Aβ accumulation, and tau hyperphosphorylation are the utmost risk factor regarding AD pathology. To date, there is no exact treatment against AD progression. In this regard, melatonin plays a crucial role for the inhibition of circadian disruption by controlling clock genes and also attenuates Aβ accumulation and tau hyperphosphorylation by regulating glycogen synthase kinase-3 (GSK3) and cyclin-dependent kinase-5 (CDK5) signaling pathway. In this review, we highlight the possible mechanism of AD etiology and how melatonin influences neurogenesis by attenuating circadian disruption, Aβ formation, as well as tau hyperphosphorylation. Furthermore, we also find out and summarize the neuroprotective roles of melatonin by the blockage of Aβ production, Aβ oligomerization and fibrillation, tau hyperphosphorylation, synaptic dysfunction, oxidative stress, and neuronal death during AD progression.

This study aimed at exploring the cause behind this inter-individual alterations in consequences of 5-fluorouracil-based chemotherapy by investigating the effects of DPYD*2A and MTHFR C677T polymorphisms on toxicity and response of 5-FU in Bangladeshi colorectal cancer patients. Colorectal cancer patients (n = 161) receiving 5-FU-based chemotherapy were prospectively enrolled. DPYD and MTHFR polymorphisms were assessed in peripheral leukocytes. Multivariate analyses were applied to evaluate which variables could predict chemotherapy-induced toxicity and efficacy. Altered gene expressions due to genetic polymorphisms in the BRCA1, BRCA2, RAD51, and HER2 contribute toward the development of breast cancer, and yet, no such type of study has been conducted in the Bangladeshi population. This study was designed to evaluate the role of BRCA1rs80357713, BRCA1rs80357906, BRCA2rs11571653, RAD51rs1801320, and HER2rs1136201 polymorphisms as risk factors in the development of breast cancer in the Bangladeshi population. A total 310 patients with invasive breast cancers were recruited as cases from different public and private hospitals of Bangladesh, and 250 Bangladeshi healthy women matching age with the patients were recruited as controls. Polymerase chain reaction-restriction fragment length polymorphism method was used to analyze the genetic polymorphisms. Patients carrying BRCA1/2 mutations, GC and GC plus CC genotypes of RAD51rs1801320, and AG plus GG genotype of HER2rs1136201 polymorphisms were found to be associated with breast cancer. In subgroup analysis, AG plus GG genotype of HER2rs1136201 was found to be associated with the breast cancer risk in the patients younger than 45 years of age compared with the older patients having more than 45 years of age, and RAD51rs1801320 was related to the tumor size and tumor aggressiveness (higher graded tumor). Our results indicate that BRCA1/BRCA2, RAD51rs1801320 and HER2rs1136201 polymorphisms were associated with breast cancer in the studied population.

Various polymorphisms of tumor suppressor gene TP53 and E-cadherin (CDH1) have been found to be associated with increased breast cancer risk worldwide. This study aimed to analyze the contribution of TP53 and CDH1 gene anomalies in breast cancer risk in the Bangladeshi breast cancer patients. For risk determination, 310 patients with breast cancer and 250 controls from Bangladeshi women were recruited who are matched up with age and use of contraceptives with patients. Genetic polymorphisms were detected by using polymerase chain reaction restriction fragment length polymorphism. Birds (n=100) were randomly and equally distributed into four groups (T0, T1, T2 and T3) and fed on a diet containing 0, 2, 4 and 8 g Spirulina/kg feed respectively for 4 weeks. The body weight was significantly (P<0.05) increased in the treatment groups fed with Spirulina diet from 7 th days to 28 th days old. FCR was also significantly (P<0.05) decreased among the treatment groups. Hematological parameters were significantly (P<0.05) increased except ESR which was decreased significantly (P<0.05) in the treatment group. Aspartate aminotransferase (ALT) and alanine aminotransferase (AST) level were significantly (P<0.05) decreased in all the treatment groups. The study suggests that, Spirulina is a good natural feed additive which has a tremendous effect to improve the broiler production and thereby may reduce the production cost.

To verify possible associations between polymorphisms of glutathione S-transferase Mu (GSTM1), gl... more To verify possible associations between polymorphisms of glutathione S-transferase Mu (GSTM1), glutathione S-transferase θ (GSTT1) and glutathione S-transferase Pi (GSTP1) genes and susceptibility to lung cancer. Methods: A total of 106 lung cancer patients and 116 controls were enrolled in a case-control study. The GSTM1 and GSTT1 were analyzed using PCR while GSTP1 was analyzed using PCR-restriction fragment length polymorphism. Risk of lung cancer was estimated as odds ratio at 95% confidence interval using unconditional logistic regression models adjusting for age, sex, and tobacco use. Results: GSTM1 null and GSTT1 null genotypes did not show a significant risk for developing lung cancer. A significantly elevated lung cancer risk was associated with GSTP1 heterozygous, mutant and combined heterozygous+mutant variants of rs1695. When classified by tobacco consumption status, no association with risk of lung cancer was found in case of tobacco smokers and nonsmokers carrying null and present genotypes of GSTM1 and GSTT1. There is a threefold (approximately) increase in the risk of lung cancer in case of both heterozygous (AG) and heterozygous+mutant homozygous (AG+GG) genotypes whereas there is an eightfold increase in risk of lung cancer in cases of GG with respect to AA genotype in smokers. Conclusions: Carrying the GSTM1 and GSTT1 null genotype is not a risk factor for lung cancer and GSTP1Ile105Val is associated with elevated risk of lung cancer.

Standard streptokinase and water was used as positive and negative control which demonstrated 37.08% and 33.24% lysis of clot of human blood Preliminary phytochemical investigation suggested the presence of various compounds described later in the plants. The decoctions of Coccinea grandis (L.) and Centella asiatica (L.) leaves were evaluated for thei... more The decoctions of Coccinea grandis (L.) and Centella asiatica (L.) leaves were evaluated for their antidiabetic and antioxidant potentials in alloxan-induced rat model and by DPPH free radical scavenging assay, respectively. Graded doses of the decoctions administered to normal and experimental diabetic rats for two weeks showed significant reduction in fasting blood glucose levels in the normal as well as in treated diabetic animals. The experiment showed 188.42 ± 1.2 mg/dl glucose and 117.78 ± 11.2 mg/dl cholesterol levels for C. grandis, while 187.23 ± 0.8 mg/dl glucose and 116.14 ± 1.5 mg/dl cholesterol for C. asiatica. Although, the decoctions produced significant effect on serum urea level, the total protein and liver enzymes (SGOT and SGPT) were found to be normal. Acute toxicity studies showed that both the decoctions were safe at 2-fold high dose when compared with the commonly used antidiabetic dose. The most important cytotoxic drug namely, cyclophosphamide used in breast cancer along with epiru... more The most important cytotoxic drug namely, cyclophosphamide used in breast cancer along with epirubicin and 5-fluorouracil, is transported by ABCC transporters and detoxified by glutathione S-transferases (GSTs). The activities of these enzymes and transporters may vary in different population due to the presence of genetic polymorphisms. This study was aimed to evaluate the effects of GSTP1rs1695 and ABCC4rs9561778 polymorphisms on the response and toxicities produced by chemotherapy used in the treatment of Bangladeshi breast cancer patients. A total of 200 and 56 patients with invasive breast cancers were recruited from different public and private hospitals of Bangladesh of which 117 patients received neoadjuvant chemotherapy to examine the response as well as the toxicity, and another 139 patients received adjuvant chemotherapy to evaluate only the toxicity. Genetic polymorphisms of the mentioned genes were detected by using Polymerase Chain Reaction Restriction Fragment Length Polymorphism (PCR RFLP). Patients carrying AG and AG plus GG genotype of GSTP1rs1695 were more likely to have a good response, whereas no association of ABCC4rs9561778 was found with the chemotherapy response. Patients carrying GT and GT plus TT genotypes of ABCC4rs9561778 were found to be associated with anemia, neutropenia, leukopenia, and gastrointestinal toxicities when compared with GG genotype whereas no association was found with thrombocytopenia. GSTP1rs1695 was not associated with any type of toxicities investigated. Our result indicates that GSTP1rs1695 polymorphism was strongly associated with the response of chemotherapy, whereas ABCC4rs9561778 polymorphism was significantly related with chemotherapy-induced toxicities.
