The EPCIN was evaluated from the following assays: total phenolic content (TPC – mg of Gallic Acid Equivalents (GAE)/g of dry extract), Radical Scavenging Assays (DPPH•, HOCl-scavenging assay), and protective effects against glycation of bovine serum albumin (BSA) with methylglyoxal (MGO) or a mixture of reducing sugars, fructose and glucose, as well as the potential for MGO capture by derivatization with ortho-phenylenediamine (OPD). To evaluate the antidiabetic activity of EPCIN in vitro, we used the assays of enzymes α-amylase (4 U/mL), α-glucosidase (0.25 U/mL), and dipeptidyl peptidase-4 (DPP-4 – 3.125 mU). The cell viability of EPCIN-pretreated normal human bronchial epithelial cells (BEAS-2B) alone or in the presence of the carcinogen 4-[(acetoxymethyl)nitrosamine]-1-(3-pyridyl)-1-butanone (NNKOAc) was measured u...

Estimates point to a prevalence of 3 to 15% of pregnancies. Its etiology includes intrinsic and extrinsic aspects of the progenitress, which may contribute to the pathophysiogenesis of GDM. Recently, researchers have identified that inflammation, oxidative stress, and the gut microbiota participate in the development of the disease, with potentially harmful effects on the health of the maternal-fetal binomial, in the short and long terms. In this context, alternative therapies were investigated from two perspectives: the modulation of the intestinal microbiota, with probiotics and prebiotics, and the use of natural products with antioxidant and anti-inflammatory properties, which may mitigate the endogenous processes of the GDM, favoring the health of the mother and her offspring, and in a future perspective, alleviating this critical publi...

Moreover, the term glycation describes not only the reaction per se but also the sequence of non-enzymatic reactions in which reducing sugars, such as glucose and fructose, react with nucleophilic groups of amino acids in proteins, lipids or nucleic acids generating Advanced Glycation End-Products (AGEs). These products are also formed by other metabolic routes and are associated with various physiological dysfunctions such as atherosclerosis, osteoporosis, asthma, arthritis, sarcopenia, Alzheimer's disease, cancer and various diabetic complications. Due to these biological actions, the search for molecules that inhibit the formation of AGEs is extremely important. The present review intends to relate some important chemical aspects for the formation, action and inhibition of AGEs.

Nevertheless, benign conditions may, similarly, exhibit arthritis as their major clinical finding, but with short-term duration instead, and evolve to spontaneous resolution in a few days to weeks, without permanent articular damage. Such distinction—self-limited arthritis with no need of immunosuppressive treatment or chronic arthritis at early stages?—represents one of the greatest challenges in clinical practice, once many metabolic, endocrine, neoplastic, granulomatous, infectious diseases and other autoimmune conditions may mimic rheumatoid arthritis. Indeed, the diagnosis of rheumatoid arthritis at early stages is a crucial step to a more effective mitigation of the disease-related damage. As a prototype of chronic inflammatory autoimmune disease, rheumatoid arthrit...

Here, we describe a sensing system for the determination of VC. Instead of a bare glassy carbon electrode (GCE), a prepared multi-walled carbon nanotube-polycation-GCE was used to detect VC in samples with higher sensitivity, better repeatability, and with a lower detection limit. Under the optimal conditions, the obtained sensor presented a linear response to VC in the range of 1-100 μM with a detection limit of 500 nM. The proposed electrode also successfully detected VC concentrations in real samples.

The modified electrode was denoted as CT-DNA/NiTsPc/rGO. Fourier transform infrared and ultraviolet-visible spectroscopies were performed to characterize the composite material. The electrochemical performance of the composite for NADH oxidation was investigated by cyclic voltammetry (CV), chronoamperometry and differential pulse voltammetry (DPV). The CT-DNA/NiTsPc/rGO modified glassy carbon electrode (GCE) showed an excellent electrocatalytic activity for NADH oxidation with apparent electrocatalytic rate constant (k obs) of 7.35 × 10 5 L mol-1 s-1 and linear response range for NADH from 1 up to 1350 µmol L-1 for n = 12 (r = 0.999). The proposed sensor shows sensitivity, detection limit and quantification limit of 0.014 µA L mol-1 , 0.3 and 1 µmol L-1 , respectively. The prepared sensor was further tested for the determination of NADH in artificial human urine samples, showing promising biomedical applications.

Advanced glycation (AGEs) and lipoxidation (ALEs) products are formed through specific condensati... more Advanced glycation (AGEs) and lipoxidation (ALEs) products are formed through specific condensation reactions between nucleophiles (amino groups of free amino acids or their residues in peptides, aminophospholipids or proteins) and electrophiles (carbonyls of reducing sugars, oxidized lipids or others) generating well-defined sets of covalent adducts. The ε-amino group of the lysine is the most reactive precursor in proteins and the primary target of carbohydrate attacks. AGEs/ALEs accumulation has consequences in the development of vascular, renal, neural and ocular complications, as well as in the triggering of inflammatory and neurodegenerative diseases. Therefore, AGEs/ALEs detection, quantification and, in some cases, the assessment of the extent of glycation in biomolecules of different matrices represent a factor of primary interest for science. Reliable analytical methods are thus required. Together with basic concepts, this review presents the main advances, challenges and prospects of research involving AGEs and ALEs in biological and food systems, exploring practical strategies to ensure greater reliability in the analysis of these compounds in different matrices.

Since its discovery, in the early 1900s, this enzyme has gained increasing attention, as literature reports have progressively pointed to its critical participation in regulating nitric oxide bioavailability. Indeed, accumulating evidence in the following years would picture arginase as a key player in vascular health. Recent studies have highlighted the arginase regulatory role in the progression of atherosclerosis, the latter an essentially prooxidant state. Apart from the fact that arginase has been proven to impair different metabolic pathways, and also as a consequence of this, the repercussions of the actions of such enzyme go further than first thought. In fact, such metalloenzyme exhibits direct implications in multiple cardiometabolic diseases, among which are hypertension, type 2 diabetes, and hypercholesterolemia. Considering the epidemiological repercussions of these clinical conditions, arg...

In this study, phenological patterns and their variability within and among populations located in these biomes and in transitional zones between them were assessed. During a 15-month period, individuals from two populations in Cerrado, two in the Atlantic Forest, and six in transitional zones (three in a cerrado-like environment and three in forest fragments) were evaluated in Minas Gerais State, Brazil. The individuals were evaluated monthly according to the proportion of the canopy in each vegetative phenophase (leaf fall, leaf flush and mature leaves) and each reproductive phenophase (floral buds, flowers, immature fruits and mature fruit/seed dispersal). In order to assess the phenological variability within and among populations, habitats and biomes, the Shannon-Wiener diversity index, the Morisita-Horn similarity ...

The procedure is based on the coordination reaction between hydralazine and Fe 2+ ions, which results in the formation of a purple complex monitored at 538 nm. For determination of total iron, Fe 3+ ions were reduced using ascorbic acid. Under optimized conditions, a linear calibration graph (0.1-6.0 g•ml-1 ; n = 6) was obtained. The method allows LOD (3 of blank/slope = 0.06 g•ml-1) and LOQ (10 of blank/slope = 0.22 g•ml-1). The RSD ((s/ x) × 100) for a mixed standard containing 0.60 g•ml-1 Fe 2+ and Fe 3+ was 0.10% (n = 10). Recoveries of spiked samples were 94.3%-106.0%. The analytical frequency was 60 h-1. The effect of possible interferences has been studied. The procedure was successfully applied for analysis of environmental samples. The real samples results were comparable with those obtained by the official method considering a paired t-test and 95% of confidence level.

The SOD and CAT (in erythrocytes) and GPx (in plasma) enzymatic activities, plasma levels of lipid peroxidation, and total thiols were measured in the blood of 55 subjects with type 2 diabetes and 38 subjects without diabetes (9 pre-diabetics and 29 controls) aged 40-86 years. The total SOD activity and the lipid peroxidation were higher in diabetics compared to nondiabetics. In stratified groups, the total SOD activity was different for the hypertensive diabetics compared to the prediabetics and normotensive controls. Lipid peroxidation was significantly higher in both groups of diabetics (hypertensive and normotensive) compared to prediabetic groups and hypertensive and normotensive controls. There was no significant difference in the CAT and GPx activities, as well as in the concentration of total thiols in the groups studied...

Bioassays were conducted using mosquito populations that were either susceptible or resistant to temephos by exposure to insecticide alone or in combination with sublethal doses of the ABC transporter inhibitor, verapamil (30, 35 and 40 μM). The best result in the series was obtained with the addition of verapamil (40 μM), which led to a 2x increase in the toxicity of temephos, suggesting that ABC transporters may be partially involved in conferring resistance to the populations evaluated.
