Adipose tissue (AT) health issues and obesity are linked to the emergence of GDM. Recent research has nevertheless demonstrated that decreased AT health can exist even in the absence of fat. In this study, we investigated whether AT health influences the onset of GDM in non-obese women. Method: GDM-diagnosed expectant mothers were examined. Non-GDM women with matching BMI and age served as the Controls (n=6 per group). Before and during cesarian sections, samples of AT and fasting blood were taken, respectively. Using the Bioplex assay, the serum levels of adiponectin were determined. In 2002, postweaning multisystemic wasting syndrome (PMWS) was diagnosed in a European female wil... more In 2002, postweaning multisystemic wasting syndrome (PMWS) was diagnosed in a European female wild boar (Sus scrofa), based on the detection of porcine circovirus 2 (PCV2) DNA in various organs, including the uterus, and on histopathologic lesions. This is the first detection of PCV2 DNA in the uterus of a wild boar. Three years later (2005), a wild boar ,6-8 mo of age was found moribund. It presented wasting and dyspnea and finally died. PCV2 DNA was detected in tissue samples, and histopathologic lesions consistent with PMWS were observed. Both wild boars were from neighboring hunting areas in central Greece. Two PCV2 strains from the wild boars were genetically characterized and compared to other reported PCV2 sequences from wild boars and domestic pigs. The PCV-2 sequences from the wild boars in this study were closely related to each other and were grouped with two isolates from wild boars from Hungary. The phylogenetic analysis revealed that the virus might be transmitted between hunting areas. In addition, PCV2 may spread from domestic pigs to wild boars and vice versa.

We described a clinical history of a febrile adult white Yorkshire pig and the isolation of Ignatzschineria spp. from blood sample. The isolate was characterized phenotypically and further identified by 16S rRNA sequence analysis. Its occurrence may be misdiagnosed in veterinary hospitals especially in low-resource settings, often leading to the underreporting of such emerging infections, since the diagnostic facilities are still in very primitive phase in developing countries. More information on speciation is needed with much about the epidemiology and pathogenesis of this emerging pathogen in order to explore its role in the lives of animals and humans. Aim: To study the association of opportunistic infection due to Myroides odoratimimus in piglets ... more Aim: To study the association of opportunistic infection due to Myroides odoratimimus in piglets immunocompromised by porcine circovirus type 2 (PCV2) infection. Methods and Results: The clinical samples (n = 101) were analysed bacteriologically. The isolates were identified by their phenotypes and MALDI TOF-MS analysis as Myroides species. The phylogram constructed based on nucleotide sequences of the 16S rRNA gene showed identity (~99%) with the M. odoratimimus isolates. The minimum inhibitory concentration values for antibiotics revealed M. odoratimimus to be resistant against carbapenem, cephalosporins, aminoglycosides and fluoroquinolones. The presence of PCV2 in affected tissue samples was confirmed by amplification of the 565 bp region of ORF2 of the PCV2 genome. The topology of the phylogenetic tree grouped the PCV2 with cluster-2d. Conclusions: PCV2 being immunosuppressive in nature might have impaired the immunity thereby increasing the susceptibility of immunocompromised piglets to opportunistic pathogens such as M. odoratimimus leading to disease severity and high mortality. The M. odoratimimus isolates were found to be multidrug resistant and evidenced for uncertain clinical relevance and hence could act as hidden source of public health hazard. Significance and Impact of the Study: Myroides odoratimimus is a rarely reported human pathogen. We reported the incidence of infection due to seemingly rare isolates of M. odoratimimus causing an outbreak of pneumonia in piglets. This appears, to the best of authors' knowledge, to be the first outbreak due to Myroides recorded in animal clinical cases described in the literature.

In the present study, the spectrum of bacterial pathogens in the nasal shedding during disease pr... more In the present study, the spectrum of bacterial pathogens in the nasal shedding during disease process and in pneumonic lungs of dead animals was studied. A total of 288 clinical samples from cattle and buffaloes comprising of nasal swabs, blood, tracheal swabs, heart blood and lung tissue samples were collected from diseased (n = 190) and dead animals (n = 98). The recovered bacterial isolates were characterized by biochemical reactions, Matrix Assisted Laser Desorption/Ionization Time of Flight Mass Spectrometry (MALDI TOF-MS) and the 16S rRNA sequence analysis. The predominant bacterial isolates associated were Pasteurella multocida, Pseudomonas aeruginosa, Escherichia coli, Klebsiella pneumoniae and Staphylococcus aureus. The emerging pathogens causing bovine pneumonia identified were Leclercia spp., Stenotrophononas maltophila and Staphylococcus sciuri. Bacteriological examination of pneumonic lungs samples revealed 96.9% samples to be positive for polymicrobial isolation. Macroscopical lesions of lungs exhibited various stages and types of pneumonia with variable degree of haemorrhages, oedema and emphysema. Histopathologically, the fibrinous bronchopneumonia was observed to be the most frequent lesions seen in bovine pneumonia. Multi-drug resistance (MDR) was observed in 10% of P. multocida isolates. The resistance was seen for penicillin, cephalosporins and fluoroquinolones. Multi-drug resistance was seen in 90% of the E.coli tested. K. pneumoniae, E. hormaechei, E. cloacae, P. putida and Leclercia spp. identified were found to be multi-drug resistant. Understanding the etiological diversity of bacterial pathogens of bovine pneumonia may provide information for the better choice of therapeutics and health management.

The aim of the present study was to characterize and evaluate the genetic diversity of ex situ collections of E. ferox germplasm from different geographical states of India using microsatellite (simple sequence repeats (SSRs)) markers. Ten SSR markers were analyzed to assess DNA fingerprinting and genetic diversity of 16 cultivated germplasm of E. ferox. Total 37 polymorphic alleles were recorded with an average of 3.7 allele frequency per primer. The polymorphic information content value varied from 0.204 to 0.735 with mean of 0.448. A high range of heterozygosity (Ho 0.228; He 0.512) was detected in the present study. Swine pasteurellosis is an acute infectious disease and is considered to be one of the most econo... more Swine pasteurellosis is an acute infectious disease and is considered to be one of the most economically important diseases in Asia particularly in South and South East Asia leading to huge economic loss in livestock industry. A study was conducted to analyze the histopathological alterations caused by cytotoxins released by Pasteurella multocida serotype B, which caused Swine pasteurellosis in descript as well as nondescript pigs of Chhattisgarh, India. Affected animals exhibited pyrexia, dullness, staggering gait, anorexia, serous nasal discharge and dyspnoea. Typical lesions of oedematous swellings noted in the pharyngeal region. Case fatality rate was 95% in adult animals and 100% in piglets. The causative organism, Pasteurella multocida was isolated and characterized by biochemical tests and the detection of the KMT1, CapB, toxA, and ompH genes was performed using Polymerase Chain Reaction (PCR). Gross lesions included severe pneumonia and haemorrhages in lungs, petechial haemorrhages on serous membranes and other visceral organs. Lymph nodes were enlarged, oedematous and haemorrhagic. The histopathological lesions developed in lungs were typical fibrinous bronchopneumonia, multifocal suppuration and severe thickening of pleura. Heart showed presence of thrombi, haemorrhages and necrosis of myocardium. Hepatic architecture showed server haemorrhages and necrosis while kidneys and spleen showed degenerative and necrotic changes.

E... more Euryale ferox Salisbury is an important aquatic food plant cultivated largely in eastern India. E. ferox is a monotypic genus, and breeding programmes have mostly relied on the variability present in the primary gene pool. Knowledge of the genetic structure of the population is limited, and there are very few reports available on the genetic diversity of E. ferox. In this study, comprehensive research on the genetic diversity of 16 germplasms of E. ferox was carried out using random amplified polymorphic DNA (RAPD) and inter-simple sequence repeat (ISSR) markers. Out of 320 RAPD and 95 ISSR primers screened initially, 61 primers (40 RAPD and 21 ISSR) gave reproducible bands and were selected for further work. Amplification of the 40 RAPD primers gave 533 polymorphic bands with an average of 13.32 polymorphic bands per primer. Host plant resistance is an important component for management of the melon fruit fly, Bactrocera... more Host plant resistance is an important component for management of the melon fruit fly, Bactrocera cucurbitae (Coquillett), owing to difficulties associated with its chemical and biological control. Various biochemical traits including total sugar, reducing sugar, non-reducing sugar, tannins, phenols, alkaloids, flavinoid and pH contents of fruit were studied on 11varieties/ genotypes of muskmelon, Cucumis melo L., in relation to resistance against B. cucurbitae under field conditions. Significant differences were found in tested varieties/ genotypes for fruit infestation and larval density per fruit. AHMM/BR-1, RM-50 and AHMM/BR-8 were the most resistant; MHY-5, Durgapura Madhu and Pusa Sarabati were moderately resistant; AHMM/BR-13, Pusa Madhuras and Arka Jeet were susceptible; whereas Arka Rajhans and GMM-3 were the highly susceptible varieties/ genotypes to fruit fly in both seasons, 2011 and 2012. The larval density per fruit increased with an increase in percent fruit infestation and there was a significant positive correlation (r = 0.97) between percent fruit infestation and larval density per fruit. Total sugar, reducing sugar, non-reducing sugar and pH were lowest in resistant and highest in susceptible varieties/ genotypes, whereas tannins, phenols, alkaloids and flavinoid contents were highest in resistant and lowest in susceptible varieties/ genotypes. Total alkaloid and pH contents explained 97.96% of the total variation in fruit fly infestation and 92.83% of the total variation in larval density per fruit due to alkaloids and total sugar contents.

A study was undertaken to isolate and characterize tetracycline and nalidixic acid-resistant Kleb... more A study was undertaken to isolate and characterize tetracycline and nalidixic acid-resistant Klebsiella spp. in farm-raised, imported shrimp sold in the United States. Sixty-seven multiple antibiotic-resistant Klebsiella spp. strains were isolated from imported shrimp samples. Using morphological and biochemical methods, fifty-three strains were tentatively identified as Klebsiella pneumoniae and fourteen as K. oxytoca. Although all isolates were resistant to tetracycline, only 8 were resistant to nalidixic acid. These 8 isolates were further screened by PCR for quinolone resistance genes (qnrA, B, S, gyrA, B and parC). PCR protocols failed to amplify any qnr genes. The purified PCR amplicons of gyrA, gyrB and parC were sequenced and analyzed for point mutations that confer resistance to fluoroquinolone antibiotics. Analysis of the sequences of the gyrA amplicons from nalidixic acid-resistant Klebsiella spp. indicated two point mutations in gyrA at positions 83 (Ser→Phe) and 87 (Asp→Ala). Sequence analysis of the parC amplicons indicated an amino acid change at position 80 (Ser→Ile). No mutations were detected in gyrB. Template DNA from all isolates was screened for tetracycline resistance genes (tetA-E). Oligonucleotide primers specifically targeting a 305-bp region of tetB and a 477-bp region of tetD successfully amplified sequences from 91.0 and 44.0% of the isolates, respectively. None of the isolates contained tetA, tetC or tetE genes. Plasmids (2.0-16.0kb) were found in 23 of the 67 isolates. XbaI-PFGE identified 32 distinct macro restriction patterns (mrps) among the 61 multiple drug-resistant Klebsiella spp. that were typable. Our results indicate that imported shrimp is a reservoir for multidrug resistant Klebsiella spp. and potential health risks posed by such strains should not be underestimated.

The impact of gamma irradiation on growth and physiology of Euryale ferox was described in the pr... more The impact of gamma irradiation on growth and physiology of Euryale ferox was described in the present investigation. E. ferox is an underutilized aquatic food crop that grows in shallow-water bodies in lower Assam regions and north Bihar of India. The seeds of E. ferox were irradiated with different doses of gamma irradiation ranging from 0 to 500 Gy. It was observed that the germination and survival percentage was inhibited by increasing the irradiation dose. However, plants developed from seed exposed to an irradiation dose beyond 100 Gy did not survive more than 1 month. Further growth parameters (leaf size and number, number of thorns, root number and length, and number of flower and seeds) were also compared with respect to non-irradiated plants. Physiological parameters, viz. chlorophyll a, chlorophyll b, total chlorophyll, photosynthetic rate, transpiration rate, stomatal conductance, and intracellular CO 2 content was higher in the irradiation population of E. ferox. Superoxide dismutase (SOD) and ascorbate peroxidase (APX) activities were observed low in irradiated population of E. ferox. The proline and glycine betaine content was enhanced with increasing the irradiation dose. The present investigation explores the potential use of gamma rays in genetic improvement of E. ferox and improves understanding of the physiological responses inflicted by gamma irradiation.
