One hundred and three patients with Philadelphia chromosome or BCR-ABL positive chronic myeloid l... more One hundred and three patients with Philadelphia chromosome or BCR-ABL positive chronic myeloid leukemia (CML) in chronic phase who were on oral imatinib were included in this study. The study aimed to assess the relationship between imatinib trough serum levels and clinical outcome (as determined by molecular response) in Jordanian CML patients who have been on imatinib therapy for at least 12 months. The mean trough imatinib serum level in the group with complete molecular response (CMR) was 2891 ± 856 ng/ml, the group with major molecular response (MMR) was 2337 ± 434 ng/ml, the group with complete cytogenetic response (CCyR) was 1817 ± 563 ng/ml, and the group without CCyR was 1723 ± 673 ng/ml. A receiver operating characteristic (ROC) curve was constructed after dividing patient sample into two groups, those with MMR or better and those without MMR, in order to estimate a threshold for imatinib level that correlates with a favorable response (the former group), and analysis yielded a value of 2158 ng/ml.
Natural gas is an alternative fuel of choice in the market today due to the increase in the price... more Natural gas is an alternative fuel of choice in the market today due to the increase in the price of petroleum, as well as out of environmental concerns. Pressure requirement for a natural gas vehicle (NGV) storage tank is 3000 psig (206 bars). Thus, at NGV refueling facilities, the natural gas need to be stored at a higher pressure in order to refuel the NGV at the pressure required. Compressors are needed in the compression process at the refueling facilities. A new compressor design for natural gas refueling appliance has been developed which is the symmetrical multistage wobble-plate compressor. This compressor design is the newest variation of the axial reciprocating piston compressor. The success of the compressor design in compressing gas depends on the piston ring assembly design. Through this paper, the process of designing the piston ring assembly and considerations taken for this new compressor design were explained. The results presented are those from preliminary tests using air on the working fluid. Real tests on natural gas are to be organised utilising all the experience and lesson learnt from that on air.

Haemophilia A (HA) is an X-linked recessive bleeding disorder caused by mutations in the factor VIII gene (F8), which encodes factor VIII (FVIII) protein, a plasma glycoprotein, that plays an important role in the blood coagulation cascade. In the present study, our aim was to identify F8 gene mutations in HA patients from Jordan. One hundred and seventy-five HA patients from 42 unrelated families were included in this study. Among these patients, 117 (67%) had severe HA, 13 (7%) had moderate HA and 45 (26%) had mild HA. Severe patients were first tested for intron-22 inversion using long range polymerase chain reaction (PCR), then negative patients were tested for intron-1 inversion using PCR. Sequencing for the entire F8 gene was performed for all severe HA patients who were found negative for intron-22 and -1 inversions and it was also performed for moderate and mild HA patients. HA causative mutations were identified in all patients. Intron-22 and -1 inversions were detected in 52% and 2% of families respectively. Beside these two mutations, 19 different mutations were identified, which include 15 missense and four frameshift mutations. Five novel mutations were identified including one frameshift and four missense mutations. No large deletions or nonsense mutations were detected in patients who participated in this study. Only 17 patients with severe HA were found positive for FVIII inhibitors. The data presented will play an important role for genetic counselling and health care of HA patients in Jordan.

A highly efficient, selective and specific method for simultaneous quantitation of triprolidine a... more A highly efficient, selective and specific method for simultaneous quantitation of triprolidine and pseudoephedrine in human plasma by liquid chromatography-ion trap-tandem mass spectrometry coupled with electro spray ionization (LC-ESI-ion trap-tandem MS) has been validated and successfully applied to a clinical pharmacokinetic study. Both targeted compounds together with the internal standard (gabapentin) were extracted from the plasma by direct protein precipitation. Chromatographic separation was achieved on a C 18 ACE ® column (50.0 mm × 2.1 mm, 5 m, Advance Chromatography Technologies, Aberdeen, UK), using an isocratic mobile phase, consisting of water, methanol and formic acid (55:45:0.5, v/v/v), at a flow-rate of 0.3 mL/min. The transition monitored (positive mode) was m/z 279.1 → m/z 208.1 for triprolidine, m/z 165.9 → m/z 148.0 for pseudoephedrine and m/z 172.0 → m/z 154.0 for gabapentin (IS). This method had a chromatographic run time of 5.0 min and a linear calibration curves ranged from 0.2 to 20.0 ng/mL for triprolidine and 5.0-500.0 ng/mL for pseudoephedrine. The within-and betweenbatch accuracy and precision (expressed as coefficient of variation, %C.V.) evaluated at four quality control levels were within 94.3-106.3% and 1.0-9.6% respectively. The mean recoveries of triprolidine, pseudoephedrine and gabapentin were 93.6, 76.3 and 82.0% respectively. Stability of triprolidine and pseudoephedrine was assessed under different storage conditions. The validated method was successfully employed for the bioequivalence study of triprolidine and pseudoephedrine formulation in twenty six volunteers under fasting conditions.
