By weaving the old, well-worn cloth of fairy tale narrative scripts together with new schematics, Gaiman and his collaborators present sometimes radical ideologies clothed in the comfortable garb of rehearsed, longstanding traditions. As much as this action masks the changing cultural attitudes represented by the "new cloth," Gaiman's weavings also reveal to careful readers how narratives become ubiquitous and achieve metanarrativity. To understand how our minds interpret scripts, schemas, and metanarratives, this chapter will bring to bear conceptual blending and schema theories on my examination of the verbal and visual interplay of source texts and Gaiman retellings

Focusing on the dialogue between global and local influences on children’s culture, this chapter looks at how complex cross-cultural negotiations allow for innovative storytelling and production in a range of family films, including examples from Disney and Studio Ghibli and three films from the Philippines. Gutierrez anchors her study of cross-cultural exchange on the way global narrative and cinematic conventions are given local color, becoming a hybrid that is called “glocal.” As well as enriching local representations of identity and nationality, this type of blending reveals how local variations contribute to global themes of childhood. “Global stories, local imagination, glocal innovations” shows the power that can be gained from glocal exchange and hybridity.

We used different methods and perspectives to evaluate the responsiveness of PROMIS® short forms (SFs) and identify minimal and meaningful score changes. Methods: Adults with RA enrolled in a multi-site prospective observational cohort completed PROMIS Physical Function, Pain Interference, Fatigue, Participation in Social Roles/Activities SFs, PROMIS-29, and pain, patient global, and rated change in specific symptoms and RA (a little vs. lot better or worse) at the second visit. Physicians recorded joint counts, MD Global Assessment, and change in RA at visit 2. We compared mean score differences for minimal and meaningful improvement/worsening using patient and MD change ratings and distribution-based methods, and visually inspected empirical cumulative distribution function curves by change categories. Results: The 348 adults were mostly (81%) female with longstanding RA. Using patient ratings, generally 1-3 point differences were observed for minimal change and 3-7 points for meaningful change. Larger differences were observed with patient vs. physician ratings and for symptomspecific vs. RA change. Mean differences were similar among SF versions. Prespecified hypotheses about change in PROMIS Physical Function, Pain Interference, Fatigue and Participation and legacy scales were supported. Conclusions: PROMIS SFs and the PROMIS-29 Profile are responsive to change and generally distinguish between minimal and meaningful improvement and worsening in key RA domains. These data add to a growing body of evidence demonstrating robust psychometric properties of PROMIS and supporting use in RA care, research, and decision-making.

Thus designing chimeric malaria vaccine candidates involving MSP-3 and UB05 displayed upon the surface of a phage in its native form could potentiate their immunogenicity and antigenicity. In this study, we have engineered both MSP3 and UB05 upon the Qβ and assessed their antigenicity with plasma from children living in a high malaria transmission region of Cameroon. Methods: The surface of the RNA coliphage Qβ was genetically modified to display three Plasmodium falciparum derived immunogens including MSP3, UB05 and a chimera of the two UB05-MSP3. The resultant recombinant phages including QβMSP3, QβUB05 and QβUB05-MSP3 with surface displayed malaria immunogens were produced after transformation of the E. coli strain HB101. Plasma levels of antigen specific IgG antibody were then determined in samples from malaria positive and negative children living in a high malaria transmission region of Cameroon. Results: To improve yield each recombinant phage was scaled up to 10 14 pfu/ml using production strategies previously optimized in our group. This was significantly higher (P<0.001) relative to the 10 8 pfu/ml of the wild type C li nical M ic r o b io logy : O p e n Acces s

Twenty-two female collegiate soccer athletes performed ankle corrective exercises for 3 months to attempt to establish and maintain optimal ankle joint function. The corrective exercises were devised to improve ankle range of motion (ROM) and Star-Excursion Balance Test (SEBT) scores. Statistically significant improvements were seen in ankle ROM and in the SEBT for the posteromedial, posterolateral, and composite reach portions of the test; the anterior reach direction failed to show significant changes. These results have clinical implications which suggest that corrective exercises may promote optimal ankle joint function and potentially reduce the risk of injury to the ankle.

Because individuals in this region are exposed to filarial parasites from birth, most HIV-1 infected individuals invariably also have a history of filarial parasite infection. Since HIV-1 infection both depletes immune system and maintains it in perpetual inflammation, this can hamper Loa loa filarial parasite mediated immune modulation, leading to enhanced loaisis. In this study we have assessed in plasma from asymptomatic anti-retroviral (ARV) naïve Loa loa microfilaraemic HIV-1 infected people the filarial antibody responses specific to a filariasis composite antigen consisting of Wbgp29-BmR1-BmM14-WbSXP. The antibody responses specific to the filariasis composite antigen was determined by enzyme linked immunosorbent assay (ELISA) in plasma from ARV naïve Loa loa microfilaraemic HIV-1 infected participants. This implies that antiretroviral naïve HIV-infected people living within these regions are repeatedly exposed to Plasmodium species infection and consequently malaria. Natural killer (NK) cells are known to contribute to malaria immunity through the production of IFN-γ after exposure to Plasmodium falciparum-infected erythrocytes (infected red blood cells [iRBC]). However, in antiretroviral naïve HIV-1 infection, these functions could be impaired. In this study we assess the ability of NK cells from antiretroviral naïve HIV-1-infected people to respond to iRBC. Method: Magnetically sorted NK cells from antiretroviral naïve HIV-1-infected people were tested for their ability to respond to iRBC following in vitro coculture. However, some are capable of offering critical commentary on society, humanity and more broadly, life itself. Following the lead taken by the 'God of Manga' Tezuka Osamu in "Seimei-hen" in "Hi no Tori" ("Life" in "Phoenix", 1980), a number of manga and anime have produced unsettling images of clones and hybrid beings, particularly those resulting from organ transplantation. These works question, typically ahead of the technology of the time, the value of life, the integrity of its form, and its immunity from commodification. In spite of their fictionality, these narratives are associated with a great sense of reality and immediacy, due in part to rapid developments in biotechnology, computing and engineering. At the same time, humanity itself appears to have changed along with these developments and the fictional narratives can be said to embody fears, hopes, and dreams concerning life and its significance. They deal with a range of pressing social and ethical issues, especially those related to the self and its multiple boundaries, whilst entertaining their readers and viewers. Using several narratives as exemplars, this paper will explore the use of biotechnology in manga and anime as devices in envisioning 'life'-what it may be, how it is formed and how it could be dealt with, at the individual as well as collective level. In doing so, the paper will demonstrate how these manga and anime narratives and others like them are relevant in a wide range of contexts despite their apparent linguistic and cultural specificity.
