Cytotoxic T-lymphocyte (CTL) clones specific for the influenza A/PR/8/34 virus hemagglutinin (HA)... more Cytotoxic T-lymphocyte (CTL) clones specific for the influenza A/PR/8/34 virus hemagglutinin (HA) were isolated by priming CBA mice with a recombinant vaccinia virus expressing the HA molecule. The epitopes recognized by two of these clones, which were CD8+, Kk restricted, and HA subtype specific, were defined by using a combination of recombinant vaccinia viruses expressing HA fragments and synthetic peptides. One epitope is in the HA1 subunit at residues 259 to 266 (numbering from the initiator methionine), amino acid sequence FEANGNLI, and the other epitope is in the HA2 subunit at residues 10 to 18 (numbering from the amino terminus of the HA2 subunit), sequence IEGGWTGMI. These two peptides are good candidates for naturally processed HA epitopes presented during influenza infection, as they are the same length (eight and nine residues) as other naturally processed viral peptides presented to CTL. A comparison of the sequences of these two new epitopes with those of the three pr...

So far, scientists have designed BODIPY dyes with high quantum yields by introducing alkyl substituents at the 1- and 7-positions thereby restricting the rotation of the meso-substituent. Herein, we propose an easier way to achieve the same objective. Synthons 1 were prepared by the Liebeskind-Srogl cross-coupling reaction and have high quantum yields by themselves. However, it is evident that having a Br atom at the ortho-position, they could be further elaborated using other transition-metal catalyzed reactions. In this work, we present our results on the functionalization of the ortho-aryl position of 1 via the Suzuki, Stille and Sonogashira reactions.

The Rhodamine organosilane derivative (Rh-UTES) was used as metal receptor through formation of luminescence chelates within the porous silicon microcavities (PSiMc). The attachment of organic derivative into PSiMc was confirmed by FTIR, specular reflectance and scanning electron microscopy (SEM). The complexing ability of Rh-UTES receptor to Hg+2 detection was investigated by fluorescent spectroscopy and microscopy. We found that the fluorescent intensity of the PSiMc hybrid device is metal concentration dependent. Similar behavior was observed in liquid and solid phase. Metal-Ligand affinity was study by electrochemical techniques.

Ultra-short peptides 1-9 were designed and synthesized with phenylalanine, ornithine and proline ... more Ultra-short peptides 1-9 were designed and synthesized with phenylalanine, ornithine and proline amino acid residues and their effect on antimalarial activity was analyzed. On the basis of the IC 50 data for these compounds, the effects of nature, polarity, and amino acid sequence on Plasmodium berghei schizont cultures were analyzed too. Tetrapeptides Phe-Orn-Phe-Orn (4) and Lys-Phe-Phe-Orn (5) showed a very important activity with IC 50 values of 3.31 and 2.57 μM, respectively. These two tetrapeptides are candidates for subsequent in vivo assays and SARS investigations.

Data are reported in the following order: chemical shift in ppm, multiplicities (br (broadened), s (singlet), d (doublet), t (triplet), q (quartet), sex (sextet), hex (hextet), m (multiplet), exch (exchangeable), app (apparent)), coupling constants, J (Hz), and integration. Infrared spectra were recorded on a Perkin-Elmer FTIR 1600 series spectrophotometer. Peaks are reported (cm-1) with the following relative intensities: s (strong, 67-100%), m (medium 40-67%), and w (weak 20-40%). Melting points were determined on a Stanford Research Systems EZ-Melt apparatus and are not corrected. High-resolution mass spectra were obtained using a VG-70-250S (double focusing) mass spectrometer at 70 eV. Analytical thin-layer chromatography was performed on Merck silica gel plates with F-254 indicator. For the starting materials that were not colored, visualization was accomplished by UV-light and 5% phosphomolybdic acid solution in ethanol. THF was dried over activated 4Å molecular sieves. All reactions were performed under a dry N 2 atmosphere in oven-and or flame-dried glassware. STARTING MATERIALS 8-Thiomethylbodipy 1, [1] CuTC, [2] were prepared according to the literature procedures. All of the other reactants, ligands and catalysts were commercially available. Typical procedure (TP) for the cross-coupling of 1 with boronic acids. All of the reactions started with 0.06 mmol (15.0 mg) of thiomethylbodipy 1 as limiting reagent and the other components of the reaction were added in the amounts stated in the TP. An oven-dry Schlenk tube, equipped with a stir bar, was charged with the boronic acid (3 equiv.), thiomethyl bodipy 1 (1 equiv.) and dry THF (2 mL) under N 2. The stirred solution was sparged with N 2 for 10 min., whereupon CuTC (3 equiv.), Pd 2 (dba) 3 (2.5 mol%), and TFP (7.5 mol%) were added under N 2. The reaction mixture was immersed into a pre-heated oil bath at 55 °C. After TLC showed that the reaction went to completion, the reaction mixture was allowed to reach rt and was adsorbed on SiO 2-gel. After flash-chromatography (SiO 2-gel, EtOAc/hexanes gradient) purification, mesosubstituted bodipy's were obtained as highly-colored crystalline solids. For purposes of characterization, the products were crystallized from CH 2 Cl 2 /petroleum ether.

A class of structurally unique heterocycles was synthesized in good yields. Over the past decade, direct functionalization of C-H bonds has become a popular and sought after approach in CC bond forming reactions. 1 Using this strategy ensures more concise and less wasteful syntheses by foregoing the steps required to make activated starting materials necessary for conventional cross-coupling reactions. Although direct arylation of heteroaromatic compounds is widespread, 1 the use of halogenated heterocycles as electrophiles in C-H functionalizing reactions is less common. 2 Herein we discuss the use of 4-iodo-1,2,3-triazoles as electrophiles in a C-H functionalization. A common approach in the synthesis of 1,2,3-triazoles is the copper-catalyzed Huisgen cycloaddition of alkynes and azides (CuAAC). The efficacy and reliability of this reaction has demonstrated its utility in many areas of chemical science, and applications of both fused and linear triazoles in pharmaceutically relevant targets are common. 3-5 A drawback of this cycloaddition is the attenuated reactivity of disubstituted alkynes, which can limit the scope to the
