We investigated the effects of CAV1‐T29107A and endothelial nitric oxide synthase (eNOS) G894T polymorphisms on the serum levels of testosterone, NO and prostate‐specific antigen (PSA) in patients with PC. We genotyped cav‐1 and eNOS genes in 112 PC patients and 150 healthy controls by PCR‐RFLP. Serum levels of NO2‐ and NO3‐ were measured using spectrophotometry, and serum levels of testosterone and PSA were measured by ELISA. The frequencies of CAV1 genotypes A/T vs. A/A according to the dominant model AT + TT vs. AA genotype and T allele were significantly higher in PC patients in comparison with the control group and considerably increased the risk of disease by 2.19‐, 1.44‐ and 1.6‐fold, respectively. AT + TT genotypes were associated significantly with the increased risk of PC in those with smoking or diabetes by 3.08‐fold (P = .004). Individuals carrying concurrently the T allele of CAV1 A29107T and the T allele of eNOS G894T genes had a significantly increased risk of PC by 2.52‐fold (P = .009). We did not find any significant relationship between eNOS G894T genotypes and alleles with susceptibility to PC. Our results highlighted the significance of CAV1‐T29107A SNP but not (eNOS) G894T in the susceptibility to PC in our the population that we have studied.

Many of the current treatments and anticancer therapeutics have problems with severe side effects and on the other hand, the drug resistance is also another obstacle in the cancer chemotherapy. Hence, there is a strong demand for the discovery and development of effective new antineoplastic therapies. According to the in vitro effectiveness of 1,3,4-thiadiazole based compounds as anticancer agents, new 1,3,4-thiadiazole based derivatives with various electron withdrawing and electron donating moieties were synthesized and tested by MTT assay against three cancerous cell lines. PC3 (Prostate cancer), U87-C-531 (Glioblastoma) and MDA-MB-231 (Breast cancer) cell lines were applied for MTT assay and obtained results were compared to imatinib. Study of the structure activity relationship of prepared compounds showed electron withdrawing substituents such as Cl, F and NO2 enhanced the anticancer properties compared to compound without any substituent (compound 3l) or compounds with electron donating (methoxy) substituent (compounds 3j and 3k). Totally, compound 3a (IC50 = 10.6 µM) showed superior activity against PC3 cell line and compounds 3d (IC50 = 10.3 µM), 3h (IC50 = 12.5 µM) and 3j (IC50 = 11.3 µM) exhibited higher activity against MDA-MB-231 cell line compared to imatinib as reference drug.

Due to recent advances in angiogenesis and its related key factors, this process as a hallmark in glioblastoma has attracted more consideration from the research community. Temozolomide (TMZ) as the first-line treatment used to treat GBM but, resistance to TMZ limits its effectiveness and the need for better treatments is still felt. Therefore, we aimed to examine the Synergistic effects of Gefitinib (GFI) in combination with Temozolomide on VEGF and MMPs in glioma cell line (U87MG). Our results displayed that GFI could induce cytotoxic effects in U87MG with IC50 values of 11 μM. U87MG cells produced large amounts of VEGF without any stimuli, and the results showed that GFI in combination with TMZ caused a significant decrease in VEGF production in these cells. In this study, we demonstrated that after treating with TMZ and GFI, there was more decrease in the levels of MMP 2 and 9 secretions in cells than treatment with GFI and TMZ doses alone. This study indicates synergistic effects of GFI plus TMZ against glioma are mediated by the potentiated anti-angiogenesis. Therefore, it can be considered as a promising plan for future studies.

Despite advances in the diagnosis and treatment, overall survival of patients still remains poor. Hence, there is an urgent need for development of new anticancer agents. Considering promising biological activity of 1,3,4-thiadiazole derivatives, in the present study, synthesis and cytotoxicity assessment of new derivatives of this ring was done. All synthesized compounds were characterized by NMR, IR and MS spectroscopic methods. Obtained data from MTT assay showed that all compounds 3a- 3l had better anticancer activity against MDA(breast cancer) compared to PC3(prostate cancer) and U87(Glioblastoma). Compound 3 g with m-OCH3 moiety on the phenyl ring was the most potent one in this series with IC50 = 9 μM against MDA breast cell line in comparison with imatinib (IC50 = 20 μM) as reference drug.

The present study aimed to investigate the rate of accumulation, human health risk assessment, an... more The present study aimed to investigate the rate of accumulation, human health risk assessment, and nitrate-related transfer factor in vegetables irrigated with different sources, including treated wastewater effluent (TWE) of Kermanshah wastewater treatment plant, Gharasoo river water (RW) of Kermanshah, and well water with chemical fertilizer (WWF). For this purpose, three different types of vegetables, including basil, coriander, and radish, was cultivated, and each of them was irrigated by the three irrigation sources mentioned above. Finally, the amount of nitrate in different sources of irrigation, soil (before growing vegetables and after harvesting vegetables), and the mentioned vegetables was measured. Based on the study results, it can be concluded that the water of the Gharasoo River (RW), compared to the other two irrigation sources, causes more nitrate accumulation in the soil and vegetables grown in it. The highest transmission factor was related to basil vegetables irrigated with WWF. The results showed that the average daily intake of nitrate through the consumption of vegetables grown in Kermanshah with any irrigation water is less than the allowable amount, so the consumption of such vegetables is not dangerous to consumers' health. Therefore, it is suggested that the best irrigation source for vegetable cultivation in Kermanshah is TWE, provided that all of its physical, chemical and microbial parameters meet the standards for reuse in agriculture irrigation. Thus, the use of treated wastewater reduces the need for farmers to use chemical and organic fertilizers and cost-effectiveness, high frequency, and high availability.

There are reports of garlic protective effects in the cardiac hypertrophy due to overload and catecholamine. The aim of this study was to investigate the effects of garlic powder on testosterone-induced cardiac hypertrophy in rats. Materials and Methods: This experimental study was performed on Wistar male rats in 5 groups (n = 7-8) including the testosterone group (10 mg testosterone enanthate for 20 days), the garlic group (1.6 g for 10 days), the control garlic group and finally the garlic + testosterone. The separated hearts were perfused according to Langendorff method and passed three basal, ischemia and reperfusion stages. The structural and functional parameters of left ventricular fluctuations (LVDP), ventricular contractility (dp / dt) and rate pressure product (RPP) were measured. Results: In the testosterone-induced hypertrophy group, the cardiac weight, the thickness of the left ventricular wall and the functional parameters of the heart, such as LVDP, RPP, + dp / dt, and-dp / dt indicated significant increase. The use of garlic powder in the base period led to a significant decrease in these parameters (P <0.05). However, in the period of ischemic reperfusion, the effect was not significant. Conclusion: The results showed that the garlic powder modifies the cardiac hypertrophy induced by testosterone administration in rats. In other words, the use of garlic powder could potentially reduce the severity of cardiac hypertrophy in rats, due to variations in functional and structural parameters.

Materials and methods: Samples from 5 different frying oils and 12 samples of oils for cooking (natural lipids) were collected. All the frying oils as well as 6 samples of oils for cooking were subjected to fatty acid analysis using high performance liquid chromatography (HPLC). Despite advances in the diagnosis and treatment, overall survival of patients still remains poor. Hence, there is an urgent need for development of new anticancer agents. Considering promising biological activity of 1,3,4-thiadiazole derivatives, in the present study, synthesis and cytotoxicity assessment of new derivatives of this ring was done. All synthesized compounds were characterized by NMR, IR and MS spectroscopic methods. Obtained data from MTT assay showed that all compounds 3a- 3l had better anticancer activity against MDA(breast cancer) compared to PC3(prostate cancer) and U87(Glioblastoma). Compound 3 g with m-OCH3 moiety on the phenyl ring was the most potent one in this series with IC50 = 9 μM against MDA breast cell line in comparison with imatinib (IC50 = 20 μM) as reference drug.

The present study aimed to investigate the rate of accumulation, human health risk assessment, an... more The present study aimed to investigate the rate of accumulation, human health risk assessment, and nitrate-related transfer factor in vegetables irrigated with different sources, including treated wastewater effluent (TWE) of Kermanshah wastewater treatment plant, Gharasoo river water (RW) of Kermanshah, and well water with chemical fertilizer (WWF). For this purpose, three different types of vegetables, including basil, coriander, and radish, was cultivated, and each of them was irrigated by the three irrigation sources mentioned above. Finally, the amount of nitrate in different sources of irrigation, soil (before growing vegetables and after harvesting vegetables), and the mentioned vegetables was measured. Based on the study results, it can be concluded that the water of the Gharasoo River (RW), compared to the other two irrigation sources, causes more nitrate accumulation in the soil and vegetables grown in it. The highest transmission factor was related to basil vegetables irrigated with WWF. The results showed that the average daily intake of nitrate through the consumption of vegetables grown in Kermanshah with any irrigation water is less than the allowable amount, so the consumption of such vegetables is not dangerous to consumers' health. Therefore, it is suggested that the best irrigation source for vegetable cultivation in Kermanshah is TWE, provided that all of its physical, chemical and microbial parameters meet the standards for reuse in agriculture irrigation. Thus, the use of treated wastewater reduces the need for farmers to use chemical and organic fertilizers and cost-effectiveness, high frequency, and high availability.

Stresses such as fermentation play an important role in bacterial behavior and consequently in food quality. Our findings presented a significant difference between roghan and sour cream fatty acid levels only at 37°C. There are reports of garlic protective effects in the cardiac hypertrophy due to overload and catecholamine. The aim of this study was to investigate the effects of garlic powder on testosterone-induced cardiac hypertrophy in rats. Materials and Methods: This experimental study was performed on Wistar male rats in 5 groups (n = 7-8) including the testosterone group (10 mg testosterone enanthate for 20 days), the garlic group (1.6 g for 10 days), the control garlic group and finally the garlic + testosterone. The separated hearts were perfused according to Langendorff method and passed three basal, ischemia and reperfusion stages. The structural and functional parameters of left ventricular fluctuations (LVDP), ventricular contractility (dp / dt) and rate pressure product (RPP) were measured. Results: In the testosterone-induced hypertrophy group, the cardiac weight, the thickness of the left ventricular wall and the functional parameters of the heart, such as LVDP, RPP, + dp / dt, and-dp / dt indicated significant increase. The use of garlic powder in the base period led to a significant decrease in these parameters (P <0.05). However, in the period of ischemic reperfusion, the effect was not significant. Conclusion: The results showed that the garlic powder modifies the cardiac hypertrophy induced by testosterone administration in rats. In other words, the use of garlic powder could potentially reduce the severity of cardiac hypertrophy in rats, due to variations in functional and structural parameters.

Malondialdehyde (MDA) is the most important biomarker for evaluation of oxidative stress and determination of lipid peroxidation. In this study, 42 drug abusers and 22 healthy persons participated as case and control groups, respectively. MDA in volunteer sera was determined by high-performance liquid chromatography (HPLC) with fluorescence detection after pre-column derivatization using thiobarbituric acid. The analysis was performed on a ODS column by spectrofluorometer detection, operated at excitation of 515 nm and emission of 535 nm. A mixture of phosphate buffer (0.05 M, pH 6.8), containing potassium monobasic phosphate and methanol (60:40, v/v) at a flow rate of 1 ml/min, was used as the mobile phase.
