Abstract One of the big expectations of nanomedicines is progression in optimized and targeted de... more Abstract One of the big expectations of nanomedicines is progression in optimized and targeted delivery of drugs to diseased tissue without affecting the original characteristics of the tissue. Accordingly, a smart approach to increase the therapeutic index of a drug is to specifically deliver the therapeutic molecule in its active form, not only into target tissue, nor even to target organs, but more importantly, into the targeted cells. Drug therapy is based on the prototype that drug will selectively define its pharmacological activity to make the diseased person free from negative side effects and decrease the symptoms and/or cause of the disease. However, the real obstruction associated with systemic drug administration is the lack of target-specific affinity toward a pathological site, resulting in systemic toxicity and countless other side effects, as well as higher dosage requirement for efficacy. As a consequence, it is of major importance that a drug is delivered directly to its site of action within the target cell, while keeping the molecule intact. Drug delivery systems can specifically target tissue and organs of interest as well as different type of cells.

Some medicines and extracts affecting this process are already used routinely in supporting the conventional treatment of many diseases that are considered angiogenic such as cancer. However, we must be aware that the area of currently used drugs of this type is much narrower than the theoretical possibilities existing in therapeutic angiogenesis. Plant substances are a large and diverse group of compounds that are found naturally in fruits, vegetables, spices, and medicinal plants. They also have different anticancer properties. The aim of this literature review article is to present the current state of knowledge concerning the molecular targets of tumor angiogenesis and the active substances (polyphenols, alkaloids, phytohormones, carbohydrates, and terpenes) derived from natural sources, whose activity against cancer angiogenesis has been confirmed.

Bioactive compound is "a compound which has the capability and the ability to interact with one o... more Bioactive compound is "a compound which has the capability and the ability to interact with one or more component(s) of the living tissue by presenting a wide range of probable effects" .In a previous study; we discussed the bioactivity concepts and we used, for the first time, the term of "biocompounactive" to more simplify the use of bioactive compound/component in relation to their physicochemical and biological properties (e.g. chemical structure, bioavailability…etc.). The aim of this work is to prove different concepts discussed about "bioactivity" by giving some chosen examples (betalains, polyacetylenes and tocols) that demonstrate different bioactivities of these biocompounactives. Betalains, polyacetylenes and tocols are generally not categorized like the known classes of phytochemicals (phenolics, alkaloids, and terpenes), but exhibit important bioactivities such as antioxidant, antimicrobial, antifungal, anticancer, anti-inflammatory, neuroprotective and radioprotective properties… etc.

The acceptable daily intake (ADI) oh human is 0-7.5 mg/kg body weight. The objective of our study was to examine the toxicity of Tartrazine and its main metabolite Sulphanilic acid to the nematode Caenorhabditis elegans, Brine Shrimp larvae (Artemia Salina) and KGN granulosa cell line; in the aim to develop our knowledge about their toxicity effects. In this research, toxicity of Tartrazine and Sulphanilic acid were examined to the nematode Caenorhabditis elegans with Escherichia coli as a food source. Our results showed that from a 3 mM concentration of Tartrazine, and 1mM of Sulphanilic supplementation can disrupt the cell cycle nematode C. elegans even if it does not cause death. These reactions were carried out in refluxed CH3CN for 4 hours. The products were recuperated with excellent and good yields (75-89.5%). Compounds (3a-f) were screened for their antifungal activity against the budding yeast Saccharomyces cerevisiae and their antibacterial activity against Escherichia coli.

Due to the highly conserved nature of the cell-cycle machinery between yeast and humans, yeast studies are directly relevant to anticancer-drug discovery. The budding yeast is also an excellent model system for identifying and studying antifungal compounds because of the functional conservation of fungal genes. Moreover, yeast studies have also contributed greatly to our understanding of the biological targets and modes of action of bioactive compounds. Understanding the mechanism of action of clinically relevant compounds is essential for the design of improved second-generation molecules. Here we describe our methodology for screening a library of plant-derived natural products in yeast in order to identify and characterize new compounds with anti-proliferative properties.

And since there is no unifying definition, attempts to find a consensus and some common elements of the concept associated with the term "bioactive" did not stop, even if vague definitions and repeated use of the operative word "bioactive" almost integrated the term in a category of magic words! This paper tried to find a combined definition which can be a platform to develop the "bioactive" concept, and reach a solid consensus on the term "bioactive compound". We use, for the first time, the term of "biocompounactive" tomore simplify the use of bioactive compound in relation with their physicochemical and biological properties.

The philosophy that food can be health promoting beyond its nutritional value is gaining acceptance within the public arena and among the scientific community as mounting research links diet/food components to disease prevention and treatment. Biocompounactives contain chemicals that are found in small quantities in plants and certain foods (such as fruits, vegetables, nuts, oils and whole grains), they have actions in the body that can promote good health. In this work, we focus on studies that have been conducted on biocompounactives of common vegetables and legumes, and opportunities that present bioactivity of these phytochemicals to prevent many chronic diseases. As well, emphasis is placed on some challenges that face the good investissment of biocompounactives.
Capparis spinosa L., known as caper, is an aromatic plant growing in most of the Mediterranean ba... more Capparis spinosa L., known as caper, is an aromatic plant growing in most of the Mediterranean basin and some parts in the west of Asia. C. spinosa L. has been utilized as a medicinal plant for quite a long time in conventional phytomedicine. Polyphenols and numerous bioactive chemicals extracted from C. spinosa L. display various therapeutic properties that have made this plant a target for further research as a health promoter. This review is meant to systematically summarize the traditional uses, the phytochemical composition of C. spinosa L., and the diverse pharmacological activities, as well as the synthetic routes to derivatives of some identified chemical components for the improvement of biological activities and enhancement of pharmacokinetic profiles. This review also addresses the benefits of C. spinosa L. in adapting to climate change and the socio-economic value that C. spinosa L. brings to the rural economies of many countries.

All scientific studies confirm that a varied and balanced diet is a factor in protection against cancer, cardiovascular disease (CVD), diabetes, osteoporosis, obesity and high cholesterol. The philosophy that food can be a health promoting beyond its nutritional value is gaining acceptance in the public arena and among the scientific community as mounting research links diet/food components to disease prevention and treatment. In this work, we complete the discussion of the preventive approach of biocompounactives, and opportunities that present bioactivity of cereals and some animal-based foods phytochemicals to prevent many chronic diseases. As well, we finish discussing some challenges related to the evaluation of biological activity by some studies presenting important limitations that must be considered before using data types in the interpretation and extrapolation of phytochemicals bioactivities to Humans.

In this second part of the review, we focus on studies that have been conducted on biocompounactives of common fruits, and opportunities that present bioactivity of these phytochemicals to prevent many chronic diseases. As well, we discuss some challenges that face the good investissment of biocompounactives, especially that related to bioavailability and bioefficacy, on the horizon to discuss the limits of experiments interpretation, mentioned here, in the third part of this review.
